Five lactating Holstein cows in a 5×5 Latin square experiment were fed five high-concentrate total mixed rations(TMRs) to investigate the effects of step-wise improvement of forage combination on ruminal and milk fatty acid profiles. The ratio of concentrate to forage was fixed as 61:39, and the step-wise improvement of forage combination was applied as: TMR1, a ration containing corn stover; TMR2, a ration containing corn stover and ensiled corn stover; TMR3, a ration containing ensiled corn stover and Chinese wild ryegrass hay(Leymus chinensis); TMR4, a ration containing the ryegrass hay and whole corn silage; TMR5, a ration containing the ryegrass hay, whole corn silage and alfalfa hay. The TMRs were offered to the cows twice daily at 0700 and 1900 h. The entire experiment was completed in five periods, and each period lasted for 18 days. Diurnal samples of rumen fluids were collected at 0100, 0700, 1300 and 1900 h(day 16); 0300, 0900, 1500 and 2100 h(day 17); and 0500, 1100, 1700 and 2300 h(day 18). The step-wise improvement of forage combination increased energy and crude protein contents and decreased fibre content. As a result, the step-wise improvement of forage combination increased dry matter intake and milk yield(P0.05). The step-wise improvement increased dietary content of linolenic acid(C18:3 n-3), but did not alter dietary proportions of palmitic acid(C16:0), stearic acid(C18:0), oleic acid(C18:1 cis-9), linoleic acid(C18:2 n-6) and arachidic acid(C20:0). In response to the forage combination, ruminal concentration of C16:0, C18:2 n-6 and C18:3 n-3 linearly increased against their dietary intakes(P0.10). The step-wise improvement increased milk contents of C10:0, C12:0, C14:0, C16:0, C18:2 n-6 and C18:3 n-3(P0.10) and decreased milk contents of C18:0 and C18:1 cis-9(P0.05). Milk yields of C16:0, C18:1 cis-9, C18:2 n-6 and C18:3 n-3 were linearly increased by the increase of these fatty acids in the rumen(R2≥0.79, P0.05), and milk yields of C18:2 n-6 and C18:3 n-3 were also positively correlated with dietary intake of these fatty acids(R2≥0.85, P0.05). The step-wise improvement increased the transfer efficiencies from feed to milk for C18:2 n-6 from 11.8 to 14.2% and for C18:3 n-3 from 19.1 to 22.3%. In a brief, along with the step-wise improvement of forage combination, more dietary linoleic and linolenic acids might escape microbial hydrogenation in the rumen and consequently accumulated in milk fat though these fatty acids were present in low concentrations in ruminal fluids. The step-wise improvement of forage combinations could be recommended as a dietary strategy to increase the transfer efficiency of linoleic and linolenic acids from feed to milk.  相似文献

The objective of this study was to observe the forage yield, silage fermentative quality, anthocyanin stability, and antioxidant activity during the storage period and in vitro rumen fermentation of anthocyanin-rich purple corn (Zea mays L.) stover (PS) and sticky corn stover (SS). Forage yield of corn stover was weighed and ensiled with two treatments: (1) hybrid sticky waxy corn stover (control), and (2) hybrid purple waxy corn stover (treatment). Samples were stored in mini-silos for periods of 0, 7, 14, 21, 42, 63, 84, and 105 d. The results showed that PS had significantly higher (P<0.05) yields of dry matter (DM), organic matter (OM), gross energy (GE), crude protein (CP), neutral detergent fiber (NDF), acid detergent fiber (ADF), and total anthocyanins than that of the SS. Anthocyanin-rich purple corn stover silage (PSS) showed higher (P<0.05) levels of DM and CP relative to the sticky corn stover silage (SSS). Although anthocyanin-rich PSS displayed a lower (P<0.05) level of pelargonidin-3-glucoside (P3G), it had higher (P<0.05) levels of peonidin (Peo) and pelargonidin (Pel) compared to the control. Delphinidin (Del) and malvidin (Mal) were not detected in SSS during the ensilage period; in PSS, Del was no longer detected after 7 d of ensilage. Specifically, total anthocyanins in anthocyanin-rich PSS decreased rapidly (P<0.05) prior to 7 d of ensilage, and then remained at relatively stable (P>0.05) constants. Compared to the anthocyanin-rich PSS, SSS displayed significantly higher (P<0.05) pH value and ammonia nitrogen (NH3-N) content. Propionic acid (PA) at 0 d and butyric acid (BA) during the entire study period were not detected, whereas anthocyanin-rich PSS showed a higher (P<0.05) level of lactic acid (LA) than that of the SSS. Compared with the SSS extract, anthocyanin-rich PSS extract showed a higher (P<0.05) level of 2,2-diphenyl-1-picrylhydrazyl (DPPH) scavenging activity and displayed a lower (P<0.05) half maximal inhibitory concentration (IC50) value. Moreover, anthocyanin-rich PSS reduced (P<0.05) gas production (GP), and displayed lower levels of immediately soluble fraction and ratio of acetic acid (AA) to PA at 12 h, but the other parameters were unaffected (P>0.05) relative to the control. Taken together, the results indicated that: (1) anthocyanins could be stable in silage; (2) anthocyanin-rich PSS showed better silage fermentative quality and stronger antioxidant activity; and (3) anthocyanin-rich PSS had no negative effect on rumen fermentation parameters.  相似文献

The present study aimed to investigate the effect of relative humidity(RH) at either acute or chronic moderate ambient temperature(Ta) on growth performance and droppings' corticosterone metabolites of broilers.Two experiments were conducted: effect of RH(35,60 or 85%) on average daily feed intake(ADFI) and droppings' corticosterone metabolites at acute(1 d: 20–26 or 31–20°C,26 or 31°C for 6 h d–1 at 10:00–16:00) moderate Ta(experiment 1) and effect of RH(35,60 or 85%) on growth performance and droppings' corticosterone metabolites at chronic(step-wisely increasing temperature by 3°C every 3 d from 20 to 32°C within 15 d: 20–23–26–29–32°C) moderate Ta(experiment 2).Droppings were collected at the 2,4,6,8,and 22 h after Ta-RH controlled in experiment 1 and at the 2,4,6,and 22 h after Ta controlled to 32°C in experiment 2.The results showed that: 1) In experiment 1,85% RH increased(P0.05) the droppings' corticosterone metabolites at the 2,6,8,and 22 h and 35% RH increased(P0.05) it at the 2 and 22 h compared to the 60% RH.Moreover,85% RH further increased(P0.05) it compared to the 35% RH,however,no difference(P0.05) was found in ADFI among the three RH groups at acute moderate 26°C; 35 and 85% RH increased(P0.05) droppings' corticosterone metabolites at the 2,6,8 and 22 h and decreased(P0.05) ADFI compared to the 60% RH,moreover,85% RH further increased(P0.05) droppings' corticosterone metabolites and further decreased(P0.05) ADFI compared to the 35% RH at acute moderate 31°C; and the average of droppings' corticosterone metabolites in the whole period had a negative correlation(P0.02) with the ADFI.2) In experiment 2,85% RH increased(P0.01) droppings' corticosterone metabolites only at the 2 h and decreased(P0.02) ADFI and average daily gain(ADG) compared to the 60% RH,no difference(P0.05) in droppings' corticosterone metabolites was found between the 35 and 60% RH,however,35% RH decreased(P0.01) ADG compared to the 60% RH,and the average of droppings' corticosterone metabolites in the whole period also had a negative correlation(P0.02) with ADFI and ADG.In conclusion,droppings' corticosterone metabolites could be used as a RH stress index and low and high RH,especially high RH,reduced growth performance possibly through inducing RH stress at moderate temperature.  相似文献

This study investigated the effects of dioscorea opposite waste (DOW) on the growth performance, blood parameters, rumen fermentation and rumen microbiota of weaned lambs. Sixty healthy weaned Small-Tailed Han lambs (male, (22.68±2.56) kg initially) were used as the experimental animals. Four levels of concentrate: 0 (control, CON), 10% (DOW1), 15% (DOW2) and 20% (DOW3), were replaced with DOW in the basal diet as experimental treatments. The results showed that lambs fed the DOW2 diet had a higher (P<0.05) dry matter intake (DMI) than the other groups. There was no significant difference (P>0.05) among DOW groups in average daily weight gain (ADG), and replacing concentrate with DOW linearly or quadratically increased (P<0.05) the ADG, while lambs fed the DOW2 diet showed greater (P<0.05) ADG than the CON group. The relative plasma concentration of growth hormone (GH), insulin like growth factor-1 (IGF-1) and insulin were affected by DOW, replacing concentrate with DOW linearly or quadratically (P<0.05) enhanced the plasma concentration of GH, IGF-1 and insulin, which was significantly higher (P<0.05) in the DOW2 group than in the CON, DOW1 and DOW3 groups. In addition, the DOW treatment showed a lower (P<0.05) concentration of blood urea nitrogen (BUN) than the CON group. Replacing concentrate with DOW quadratically decreased (P<0.05) the ruminal ammonia nitrogen (NH3-N) and increased (P<0.05) the total of volatile fatty acids (TVFAs) at 0 and 4 h after feeding as well as linearly decreased (P<0.05) the NH3-N at 8 h after feeding. Replacing concentrate with DOW linearly decreased (P<0.05) the propionate and increased the aceate before feeding, and linearly decreased (P<0.05) propionate and quadratically increased (P<0.05) the aceate at 4 and 8 h after feeding. Lambs fed the DOW2 diet increased the phylum Firmicutes and genera Succiniclasticum and Ruminococcus_1 groups, whereas decreased (P<0.05) the relative abundance of phylum Deferribacteres and genera intestinimonas and Ruminiclostridium. In summary, replacing the concentrate with 15% DOW was beneficial for improving the rumen fermentation and ADG by increasing the DMI and modulating the rumen microbial community.  相似文献

The objective of this study was to evaluate the effects of adding formic acid and corn flour supplementation to banana pseudostem silages on the nutritional quality of these silages, growth, digestion, rumen fermentation and cellulolytic bacterial community of Nubian black goats fed these silages. Banana pseudostem silage was prepared either conventionally without any additives(CON) or mixed with 0.6% formic acid(F), 10% corn flour(C), or both(F+C). Four experimental diets containing 40% of the corresponding silages were designed with roughage to concentrate ratio of 50:50(dry matter(DM) basis). A total of 48 Nubian black castrated goats(body weight(BW),(22.64±1.82) kg; 4-mon-old) were randomized into one of the four treatment groups with 12 replicates of one castrated goat per replicate for each treatment in a completely randomized design. Each group was fed on one of the four experimental diets for 40 days. A factorial arrangement of treatments of 2(formic acid levels: 0 and 0.6%)×2(corn flour: 0 and 10%) was adopted. Formic acid supplementation increased(P0.05) average daily gain, as well as lactic acid, propionate and butyric acid and water-soluble carbohydrate concentrations, but decreased(P0.05) the feed conversion rate, p H value, acetate/propionate ratio, and butyric acid concentration relative to the CON group. Corn flour supplementation increased(P0.05) the apparent digestibility of crude protein, neutral detergent fiber, and non-fibrous carbohydrate and Fibrobacter succinogenes, Ruminococcus albus, and Butyrivibrio fibrisolvens populations, but decreased(P0.05) the Ruminococcus flavefaciens population relative to the CON group. There were no F×C treatment interactions(P0.05) for any of the other indices except for the apparent digestibility of non-fibrous carbohydrate(NFC)(P0.05). The results demonstrated that adding 0.6% formic acid and 10% corn flour supplementation to banana pseudostem silages improved the nutritional quality of these silages and enhanced the growth performance of Nubian black castrated goats by improving apparent nutrient digestibility, and regulating ruminal fermentation and bacteria populations.  相似文献

Two experiments were conducted to investigate the effect of in ovo zinc(Zn) injection on the embryonic development, tissue Zn contents, antioxidation and related gene expressions of fertilized eggs of Arbor Acres broiler breeders. Experiment 1 was conducted to determine an optimal embryonic age for early in ovo injection. A total of 720 fertilized eggs with similar weights were randomly allotted to 4 treatments with 6 replicates per treatment and 30 eggs per replicate in a completely randomized design. The eggs were injected with 0.1 m L sterilized water at 3, 6 and 9 embryonic days of incubation(E3, E6 and E9) or non-injection(the control), respectively. The results from experiment 1 showed that E3 and E6 injections increased(P0.05) the embryonic mortalities, and decreased(P0.05) hatchabilities compared to the non-injected control, but no differences(P0.05) between E9 injection and the non-injected control were observed in either embryonic mortality or hatchability. The findings suggest that the E9 is the optimal embryonic age for early in ovo injection. In experiment 2, a total of 672 fertilized eggs with similar weights were randomly allocated to 7 treatments with 6 replicates per treatment and 16 eggs per replicate in a completely randomized design. The eggs were injected with 0(the negative control), 50, 100, 150, 200, or 250 μg Zn/egg as reagent grade Zn SO4·7 H2 O in a 0.1-m L solution, or non-injection(the positive control), respectively at E9–10. The results from the experiment 2 demonstrated that no differences(P0.05) among 50 and 100 μg Zn/egg groups and the negative control were observed in the embryonic mortality and hatchability, however, the injection of 200 μg Zn/egg increased(P0.05) the embryonic mortality, and injections of 150 and 200 μg Zn/egg decreased(P0.05) hatchabilities compared with the controls. The embryonic tibia Zn contents at E20 were increased(P0.05) by injections of 150, 200 and 250 μg Zn/egg. Zinc injection did not affect(P0.05) malonaldehyde(MDA) contents, copper-and Zncontaining superoxide dismutase(Cu Zn SOD) activities and m RNA expression levels in the liver and heart of chick embryos at E15 and E20. Compared with the negative control, injections of 50, 150 and 200 μg Zn/egg up-regulated(P0.05) the metallothionein(MT) mR NA expression levels in the embryonic liver at E20. These results indicated that in ovo Zn injections increased Zn contents in the embryonic tibia and MT m RNA expression levels in the embryonic liver at E20, however, injections of 150–200 μg Zn/egg were harmful to the embryonic development.  相似文献

设置模拟氮沉降的控制试验,以NH4NO3作为外加氮源,设计CK(0kg N hm-2·a-1)、LN(50 kg N hm-2·a-1)、MN(100 kg N hm-2·a-1)、HN(150 kg N hm-2· a-1)4个处理,历时9个月,测定木荷(Schima superba)幼苗的光合特性、生物量和C、N、P含量及其分配格局对氮沉降的响应.结果表明:(1)木荷幼苗的最大净光合速率和光饱和点随着氮处理水平增加呈先增加后减小的特点,在中氮处理下极显著增加(P<0.01).氮处理降低了幼苗的光补偿点和暗呼吸速率,光补偿点在低氮处理下显著降低(P<0.05),暗呼吸速率在低中氮处理下极显著降低(P<0.01),高氮处理下显著降低(P<0.05).未见氮处理对表观量子效率产生显著影响.(2)氮处理促进了木荷的全株生物量以及各部分生物量的增长.随着氮处理水平的增加,叶重比呈升高的趋势,而根重比和根冠比呈降低的趋势,在高氮处理下叶重比的增加和根重比、根冠比的降低都达到了显著水平(P<0.05).(3)氮沉降促进各器官N含量的增加,在高氮处理下根和茎中N含量极显著增加(P<0.01),叶中N含量显著增加(P<0.05).而各器官C含量随着氮沉降程度的增加呈先增加后降低的趋势,在中氮处理下根和茎中C含量极显著增加(P<0.01),叶中C含量显著增加(P<0.05).但各器官P含量变化趋势各不相同,随着氮的增加,根中P含量是呈先增加后降低的趋势,而茎和叶中P含量是呈降低的趋势.氮沉降一定程度上降低了木荷各器官的C/N比值而增加了N/P比值.  相似文献

The present study was conducted to evaluate the effect of dietary mulberry(Morus alba L.) leaves powder(MLP) supplementation on meat quality of finishing pigs.A total of 40 Duroc×Landrace×Yorkshire pigs(initial body weight of(40.5±0.63) kg) were randomly allotted into two treatments,fed either with control diet or 15% MLP diet for 85 d.The results showed that MLP diet decreased(P≤0.05) average daily gain(ADG) and increased(P0.05) feed/gain ratio(F/G) in the finishing and whole period.MLP diet also decreased(P0.05) carcass weight,dressing percentage,last rib and average backfat depth.However,MLP diet increased(P0.05) intramuscular fat(IMF) content,decreased(P0.05) shear force,cooking loss and drip loss.In addition,MLP diet increased(P0.05) total antioxidative capacity,glutathione peroxidase and tended(P0.10) to increase total superoxide dismutase in serum.In longissimus thoracis,myosin heavy chain(MyHC) I and IIa mRNA levels were increased(P≤0.05) for MLP diet.In conclusion,15% MLP supplementation reduced the growth performance and carcass traits,but improved meat quality of finishing pigs possibly through the change of myofiber characteristics,enhancement of antioxidative capacity and increase of IMF.  相似文献

The effects of breed, sex and birth parity on the traits of growth, carcass and meat quality were investigated in three different breeds including Swedish Landrace, British Large White and an indigenous Chinese breed — Tongcheng pigs. The results indicated that the Swedish Landrace and the British Large White had advantages over the Tongcheng pigs in growth rate, feed conversion rate (FCR) and carcass traits, but had disadvantages in meat quality. In contrast to the British Large White, the Swedish Landrace had higher average daily gain during the trial (ADG2) (877.04 g·day−1 vs. 813.95 g·day−1, P < 0.05), and marbling score (MS) (2.48 vs. 2.02, P < 0.01), larger eye muscle area (EMA) (41.80 cm2 vs. 35.14 cm, P < 0.01) and more efficient feed conversion rate (FCR) (3.06 vs. 3.29, P < 0.05), lower muscle shear force (MSF) (4.20 kgf (1 kgf = 9.80665 N) vs. 4.93 kgf, P < 0.05) and backfat thickness (BF) (all P < 0.01) including live backfat thickness (LBF), average backfat thickness at 3 points in the carcass (ABF), backfat thickness between 6th and 7th ribs (6–7 th BF) and backfat thickness at 10th rib (10 th BF). There was a difference (P < 0.05) only inMS between castrated females (CF) and castrated males (CM). Birth parity affected ADG2 (P < 0.05), some carcass characteristics and meat quality, including CL1 (P < 0.001), CL2 (P < 0.05), BF (ABF, 6–7th BF and 10th BF, all P < 0.001), EMD (P, 0.001), EMA (P < 0.001), percentage of leaf and caul fat (PLC) (P < 0.05), proportion of lean and bone of the ham (PLBH) (P < 0.05), muscle drip loss percentage (DL) (P < 0.05) and intramuscular fat content (IMF) (P < 0.05). The breed-sex interaction only impacted the average daily gain from birth to marketing (ADG1) and DL (both P < 0.05). However, there was no significant effect of sex-parity interaction on all the traits tested.  相似文献

根据2011年6-8月我国拖网船在摩洛哥南部沿岸采集的46尾金色小沙丁鱼(Sardinella aurita)和53尾沙丁鱼(Sardina pilchardus)的耳石样本,利用传统形态测量法和傅立叶分析法分别进行分析,对不同的耳石形态进行判别。传统形态测量法测量了6个耳石形态参数,并利用t检验比较两种沙丁鱼耳石形态差异;耳石轮廓经图像化处理后利用软件转化为椭圆傅立叶标码(EFDs),最后分别采用6个耳石形态参数和标准化后的77个傅立叶系数进行主成分分析和判别分析。结果表明,两种沙丁鱼耳石形态在全长(TL)、宽度(TW)、背宽(DW)、腹噱长(VL)中存在显著差异(P0.01),背长(DL)和翼叶长(WL)不存在差异(P0.05);主成分分析表明,传统测量法的前两个主成分累积贡献率达77.0%,第一、二主成分的最高变量分别为全长(TL/FL)和背宽(DW/FL),在散点图中显示出了很好的区分性;而傅立叶分析法中,前16个主成分累积贡献率为82.8%,前两个主成分因子仍有较大的重叠,区分性相对较差。判别分析结果认为,传统测量法中的3个参数值进入了逐步判别分析中,总判别正确率为83.76%;傅立叶分析法的77个傅立叶系数中,有9个系数进入了逐步判别分析中,总判别正确率为92.02%。总体来看,相比传统形态测量法,傅立叶方法在鱼类耳石的判别分析中更为有效。  相似文献

In a completely randomized block design experiment, 16 ruminally cannulated male sheep with body weights of (40 ± 2.1) kg were fed twice daily (8:00 and 16:00) with concentrate and forage (50:50 on dry matter (DM) basis). Dietary treatments were supplemented with intraruminal doses of powdered Yucca schidigera extract (YSE) at the levels of 0 (control), 100, 200 and 300 mg · kg−1. On days of 15, 16 and 17 after feeding, ruminal content was sampled at 0, 2, 4, 6 and 8 h after dosing (8:00), and blood samples were collected at the end of experiment (the days 18 and 19 after feeding). Results showed that the treatment groups’ acidity was not affected (P = 0.13) by YSE. Comparing to the control, the ruminal propionate concentration was increased by YSE addition in a dose-dependent manner by up to 29.8% (P < 0.05), and the acetic concentration was decreased by up to 17.5% (P < 0.05). The ruminal ammonia concentration 2 hours after feeding was higher (P < 0.05) in sheep fed without YSE (increased by 17.57 mg· 100 mL−1) than those fed with YSE at 200 mg · kg−1 (6.77 mg · 100 mL−1 increase in NH3) and at 300 mg · kg−1 (6.50 mg· 100 mL−1 increase in NH3). Protozoal populations in the rumen were lower (P < 0.05) with the YSE feeding dose at 300 mg · kg−1 than the control. The serum chemistries were not different among treatments (P > 0.05) and were within the normal physiological ranges for sheep 19 days after feeding. The study indicated that 200 mg· kg−1 and 300 mg· kg−1 YSE groups had particular suppressing effects on ruminal ammonia concentration, ammonia-N concentrations and protozoal populations. The effect of YSE on ruminal fermentation could be attributed to the selective inhibitory effect on rumen microbial species. High level (300 mg · kg−1) YSE as feed additives resulted no negative impact on sheep in our tests.  相似文献

This experiment was conducted to evaluate the effects of Bupleurum extract(BE) on blood metabolites, antioxidant status, and immune function in dairy cows under heat stress. Forty lactating Holstein cows were randomly assigned to 1 of 4 treatments. The treatments consisted of 0, 0.25, 0.5, and 1.0 g of BE kg–1 dry matter. Supplementation with BE decreased(P0.05) blood urea nitrogen(BUN) contents and increased blood total protein(TP) and albumin(ALB) levels compared with control cows, but it had no effects(P0.05) on blood glucose(GLU), nonesterified fatty acid(NEFA), total triglyceride(TG), low-density lipoprotein cholesterol(LDL-C), and high density lipoprotein cholesterol(HDL-C). Compared with control cows, cows fed BE had higher(P0.05) superoxide dismutase(SOD) and glutathione peroxidase(GSH-Px) activity. However, supplementation with BE had no effect(P0.05) on total antioxidant capacity(T-AOC) or malondialdehyde(MDA) levels. The immunoglobulin(Ig) A and G contents increased(P0.05) in cows fed 0.25 or 0.5 g of BE kg–1. Interleukin(IL)-2 and IL-4 levels were higher(P0.05) in cows fed 0.5 and 1.0 g of BE kg–1, and IL-6 was significantly elevated(P0.05) in cows fed 0.5 g of BE kg–1. There were no treatment effects(P0.05) on the CD4+ and CD8+ T lymphocyte ratios, CD4+/CD8+ ratio, or tumor necrosis factor-α(TNF-α) level among the groups. These findings suggest that BE supplementation may improve protein metabolism, in addition to enhancing antioxidant activity and immune function in heat-stressed dairy cows.  相似文献

Marek's disease (MD), a highly cell-associated and contagious disease of chickens caused by Marek's disease virus (MDV) can result in neural lesions, immunosuppression and neoplasia in chicken. The Meq gene is an important oncogene in the MDV genome, and it is expressed highly in MD tumor tissues and MD T-lymphoblastoid cell lines. An experiment was conducted to elucidate the role of Meq in MD tumor transformation. RNA interference technology was used to block its expression, and then analyzed the biological effects of Meq knockdown on the MD tumor cell line MSB1. A small interfering RNA with an interference efficiency of 70% (P<0.01) was transfected into MSB1 cells to knock down the expression of Meq gene. The cell proliferation, cycle and apoptosis were detected post-Meq knockdown. The results showed that MSB1 cell proliferation was downregulated remarkably at 48 h (P<0.01), 60 h (P<0.05) and 72 h (P<0.01) post-Meq knockdown. The cell cycle was unaffected (P>0.05). B-cell lymphoma 2 gene (BCL2) was anti-apoptotic and caspase-6 was the effector in the apoptosis pathway. The activity of caspase-6 was upregulated (P<0.05) significantly and BCL2 gene expression was downregulated (P<0.05) significantly post-Meq knockdown, suggesting cell apoptosis might be induced. MSB1 cell migration did not exhibit any obvious change (P>0.05) post-Meq knockdown, but the expression of two genes (matrix metalloproteinase 2 (MMP2) and MMP9) that are correlated closely to cell invasion was downregulated (P<0.05) remarkably post-Meq knockdown. The Meq knockdown might affect the main features of tumorous cells, including proliferation, apoptosis, and invasion, suggesting that the Meq gene might play a crucial role in interfering with lymphomatous cell transformation.  相似文献

The present study was conducted to evaluate the effects of different folic acid supplemental levels on growth performance, serum biochemical indicators, and hepatic folate metabolism-related gene expressions in weaned piglets. There were 160 piglets with initially average bodyweight of 7.33 kg randomly assigned to diets containing five levels of folic acid: basal diets (C), 0.5 mg·kg−1 folic acid (FS 0.5), 2.5 mg·kg−1 folic acid (FS 2.5), 5.0 mg·kg−1 folic acid (FS 5.0), or 10.0 mg·kg−1 folic acid (FS 10.0). Blood samples were collected from a subset (n = 20; 4 pigs per treatment) of the piglets on day 0, 14, and 28. Liver samples were collected from the blood-taken piglets on day 28 of the experiment. Pigs fed basal diet supplemented with 2.5 mg·kg−1 folic acid grew faster (P<0.05) and consumed more feed (P<0.01) than groups of C, FS 5.0, and FS 10.0 during the last two weeks. Dietary treatment had no effect on F/G throughout the experiment. Pigs in the FS 2.5 group showed greater concentrations of Growth Hormone (GH) (P<0.05) and Insulin-like Growth Factors (IGF-1) (P<0.01) in serum than C and FS 10.0 on day 28. RT-PCR analysis revealed that FS 0.5, FS 2.5, and FS 5.0 had a greater abundance of the mRNA encoding 5,10-methylenetetrahydrofolate reductase than C and FS 10.0 (P<0.01). The mRNA expressions of folate binding protein in FS 0.5 and FS 2.5 were upregulated compared with pigs fed with basal diet (P<0.05). These results demonstrated that folate supplemental level of 2.5 mg·kg−1 significantly enhanced the growth performance of piglets. Folic acid had an impact on folate metabolism and the homocysteine concentrations. No folate supplementation or folate supplemental level of 10 mg·kg−1 could not increase the growth performance to the greatest degree.  相似文献

An experiment was conducted to investigate the effect of relative humidity(RH) at chronic temperature on growth performance, glucose consumption, and mitochondrial adenosine triphosphate(ATP) production of broilers. A total of 180 28-day-old Arbor Acres broilers(half males and half females) were randomly allocated to three treatments, each containing six replicates of 10 birds per treatment, using a completely randomized design. Birds were reared at 35, 60 or 85% RH at 32°C for 15 days(temperature increased by 3°C every 3 days from 20 to 32°C within 15 days: 20–23–26–29–32°C). RH affected(P0.05) average daily feed intake(ADFI), average daily gain(ADG), average daily water consumption(ADWC), blood glucose concentrations, muscle glycogen levels, avian uncoupling protein(av UCP) mRNA expression, and cytochrome c oxidase(CCO) activity in liver of broilers at 42 days of age. The 85% RH decreased(P0.05) ADFI, ADG and ADWC; 35% RH decreased(P0.02) ADG. Both 85 and 35% RH increased(P0.01) blood glucose and decreased(P0.05) muscle glycogen. Both 85 and 35% RH increased(P0.05) av UCP mRNA expression. 35% RH decreased(P0.05) CCO activity. In conclusion, both high and low RH inceased glucose consumption and reduced mitochondrial ATP poduction, leading to a decline in growth rate.  相似文献

Mechanized grain harvest of maize becomes increasingly important with growing land plot size in Northeast China. Grain moisture is an important factor affecting the performance of mechanized grain harvest. However, it remains unclear what influences grain dehydration rate. In this study, maize grain dehydrating process was investigated in a two-year field experiment with five hybrids under two planting densities in 2017 and 2018. It was found that damaged-grain ratio was the main factor affecting mechanized harvest quality, and this ratio was positively correlated with grain moisture content at harvest (R2=0.6372, P<0.01). To fulfill the national standard of <5% damaged-grain ratio for mechanized grain harvest, the optimal maize grain moisture content was 22.3%. From silking to physiological maturity, grain dehydrating process was mostly dependent on the thermal time (growing degree days, GDDs) (r=–0.9412, P<0.01). The average grain moisture content at physiological maturity was 29.4%. Thereafter, the linear relationship between GDDs and grain moisture still existed, but the correlation coefficient became smaller (r=–0.8267, P<0.01). At this stage, grain dehydrating process was greatly affected by genotypes. Grain dehydrated faster when a hybrid has a smaller husk area (r=0.6591, P<0.05), larger ear angle (r=–0.7582, P<0.05), longer ear peduncle (r=–0.9356, P<0.01) and finer ear (r=0.9369, P<0.01). These parameters can be used for breeders and farmers to select hybrids suitable for mechanized grain harvest.  相似文献

Early life intervention is important to shape the gut microbiome profiles of adult animals due to the tremendous alteration of diet components. Nevertheless, there is still no unified understanding about its long-term effects in lambs. In this study, sixty 20-day-old lambs were assigned into ewe-rearing (ER) and artificial-rearing (AR) treatments to evaluate the effects of AR strategy on ruminal microbiota, fermentation, and morphology of pre-weaning lambs (from 20 to 60 days of age) and its long-term effects in the fattening stage (from 61 to 180 days of age). During the pre-weaning stage, ER lambs were breastfed and supplemented starter, while AR lambs were artificially fed with milk replacer and starter. During the fattening stage, all lambs in both treatments were fed with the same fattening diets. At 60, 120 and 180 days of age, 6 lambs from each group were slaughtered to collect rumen content and tissue samples. Compared with ER lambs, the dry matter feed intakes of AR lambs increased (P<0.05) from 20 to 180 days of age, companying an increased average daily gain (ADG) from 61 to 120 days of age (P<0.05) and from 121 to 180 days of age (0.05<P<0.1). Although there was no difference in short-chain fatty acid (SCFA, including acetate, propionate, and butyrate) between treatments before weaning (P>0.05), it was higher (P<0.05) in AR lambs compared with ER lambs at the fattening stage. The rumen keratin layer of AR lambs was thinner (P<0.05) than that of ER lambs. Along with lamb growth from 60 to 180 days of age, the differences in rumen bacterial diversity between AR and ER treatments grew more distinct (P<0.05). Compared with ER lambs, AR lambs increased (P<0.05) rumen bacteria abundance, such as phylum Spirochaetes and genus Treponema at 60 days of age, phylum Actinobacteria and genus Succiniclasticum at 120 days of age, and phylum Proteobacteria at 180 days of age, but decreased genus Selenomonas from 60 to 180 days of age, and Anaerovibrio at 180 days of age. In summary, the early interventions before weaning could improve dry matter feed intake of lambs, which triggered robust rumen development and produced positive long-term effects on rumen fermentation and noticeable weight gain of fattening lambs. It suggests that the artificial rearing strategy is effective in improving rumen fermentation and microbial maturity of intensive fattening lambs.  相似文献

用4头装有瘤胃瘘管和十二指肠双筒T形瘘管的黑白花公犊,研究颗粒精料对复胃消化代谢的影响。试验分粉状精料期(Ⅰ期)和颗粒精料期(Ⅱ期)。两期精料成分和量完全相同。粗料为青草,计量自由采食。试验结果表明:Ⅱ期与Ⅰ期相比,平均每kg体重青草采食量和干物质摄入量分别提高了42.97%(P<0.01)和57.35%(P<0.01);复胃纤维素、干物质和有机质消化率分别提高了7.94%(P<0.01),13.03%(P<0.01)和10.61%(P<0.05);瘤胃总挥发性脂肪酸浓度变化不明显(P>0.05)。但乙酸(C_2)百分比升高(P<0.01),丙酸(C_3)和戊酸(C_5)百分比降低(P<0.01),C_2/C_3升高(P<0.01),丁酸(C_4)变化不明显(P>0.05)。两期氨氮浓度无明显差异(P<0.05);瘤胃液平均pH由6.46升到6.67(P<0.01)。Ⅱ期十二指肠食糜流量增加了18.20%(P<0.01);食糜总蛋白、总徽生物蛋白、过瘤胃蛋白和非氨氮量分别提高了26.02%(P<0.01),38.28%(P<0.05),30.63%(P<0.01)和24.66(P<0.01).结果表明颗粒精料能促进犊牛粗饲料采食量,提高复胃纤维素消化率,改变瘤胃发酵类型,减少饲料蛋白在瘤胃的分解,增大进入十二指肠食糜量和非氨氮,从而提高了饲料蛋白和纤维素的利用效率。  相似文献

This study was carried out to determine the effect of replacement of forage fiber sources from alfalfa and Leymus chinensis with nonforage fiber sources(NFFS) from dried distillers grains with solubles and corn germ meal on calves growth, rumen development and blood parameters. 48 female and 12 male calves((110.55±15.36) kg of body weight and 12 wk of age) were assigned randomly to four dietary treatments(15 calves/treatment) in a completely randomized design. Experimental diets were: 0% NFFS(control), 9% NFFS(group 1), 18% NFFS(group 2), 27% NFFS(group 3), and contained equivalent neutral detergent fiber and total digestible nutrients, respectively. The dry matter intake was similar among diets, averaging 3.33 kg d-1, and no differences were detected for body weight, withers height, body length and heart girth. In addition, the development of rumen, reticulum, omasum and abomasum also were similar among diets. Dry matter, crude protein, and neutral detergent fiber digestibilities increased with the increasing levels of NFFS in the experimental diets, but had no significant effect. Blood urea nitrogen, total protein and glubulin were not affected by the dietary treatment, but group 3 resulted in the highest(P〈0.05) concentrations of glucose and the lowest(P〈0.05) concentrations of triglycerides and albumin. In conclusion, dried distillers grains with solubles(DDGS) and corn germ meal(CGM) were available and alternative fiber sources for Holstein calf diets.  相似文献
