Summary The objective of this study was to find measurable characters to aid the plant breeder in predicting both rubber percentage and rubber yield of guayule (Parthenium argentatum Gray). A field of 234 guayule breeding lines was harvested by clipping in February 1986. Forty-two of these lines were selected for further evaluation on the basis of superior regrowth, rubber content and yield. These 42 lines were reharvested in February 1987. Nineteen variables were evaluated including percent rubber and resin, rubber and resin yield (g/plant), plant height (cm), width (cm), and volume (m3), fresh and dry weight (kg/plant), percent dry weight, stem number, mean stem diameter (cm), total and mean stem circumference (cm), total and mean stem area (cm2), total stem circumference/total stem area (cm-1), and regeneration at two dates. Dry weight (kg/plant) had the best correlation to rubber yield (r=0.85), and plant volume best predicted dry weight (r=0.83). These variables did not correlate as highly with percent rubber. The best statistical regression model for percent rubber included four variables, fresh and dry weight, percent dry weight, and plant volume. This model accounted for 51% of the variation. In comparison, the best regression of rubber yield had an R2=0.85 for plant height and width, volume, and dry weight.  相似文献

Summary To investigate genetic regulation of blossom-end scar size in tomato (Lycopersicon esculentum Mill.), a half diallel cross including 10 parents was grown under warm fall conditions in Bradenton, Florida, and cool winter conditions in Hazeva, Israel. The parents were a random sample representing all available fresh market tomato breeding lines commercially grown under subtropical field conditions. A randomized, complete block design with three replications was used and the blossom-end scar index (BSI), a measure for scar size relative to truit size, was measured on 40 and 25 fruits per plot in Bradenton and Hazeva, respectively. Analysis of variance for BSI indicated highly significant (P=0.001) variation among parents and among F1's in both environments. In Bradenton and Hazeva both, the difference in average BSI between parents and F1's was not significant suggesting insignificant overall dominance effects. Further partitioning of variation within F1's indicated that general combining ability (GCA) effects were highly significant in both environments, whereas specific combining ability (SCA) effects were significant at P=0.001 in Bradenton, but only at P=0.05 in Hazeva. Estimated variance components for GCA and SCA effects indicated that BSI inherited mainly additively in both environments. Analysis combined over environments indicated that variation in sensitivity to environments was 5-fold higher among parents than among F1's. The genotype x environment variance component was not large enough to justify testing over more than one environment for population improvement purposes and early testing of hybrids. Evaluation of inbred lines, however, may have to be done in more than one environment, especially for Suncoast-derived material. Under temperatures in Hazeva, genotype differences were more pronounced and heritabilities higher than under high temperatures in Bradenton. The estimated overall single plot heritability was 0.63. Breeding lines with a pointed blossom-end morphology (e.g. NC 8276 and NC 140 in this study) generally had small blossom-end scars, and intercrossing of these lines or crossing with non-pointed, moderately smooth breeding lines generally resulted in smooth hybrids.  相似文献

Summary Ten peanut (Arachis hypogaea L.) lines, two from each of five centers of diversity in South America, and the F2 generation of all possible crosses among them were used to characterize the variation for yield, fruit and seed traits and protein and oil content and to determine the breeding potential of the parents. Two of the parental lines belong to subspecies hypogaea and the other eight to subspecies fastigiata.Several crosses gave heterotic responses for yield. The parents of the crosses with significant heterosis generally came from different centers of diversity.Variation among both general and specific combining abilities was significant for all traits with the exception of the SCA estimate for protein percent. The component of variation for GCA was larger than the SCA component for all traits.A Spanish line and a Valencia-like intermediate line from the Guarani region had the highest GCA effects for fruit yield, seed yield and meat content and transmitted consistently their characteristics to their F2 progeny. Some lines showed significant but small maternal effects for oil, protein and oil + protein percent. Differences between reciprocal crosses were significant for fruit length, fruit weight and oil + protein percent.A significant portion of the variation among crosses was explained by differences among parental lines within centers of diversity. This supports the fact that centers are founded because of the diversity of the peanuts grown there.The performance of parental lines per se was useful in predicting the performance of the lines in hybrid combination for all traits except fruit yield.Paper number 6141 of the Journal Series of the North Carolina Agricultural Research Service (USA). Variation for resistance to Fusarium head blight in spring barley   总被引:3,自引:0,他引:3  
Fusarium head blight (FHB) is a fungal disease of barley and other cereals, causing substantial yield and quality losses, mainly due to the contamination of the harvest with mycotoxins. We aimed to evaluate genetic variation for resistance to FHB and its association with other plant characters in diverse barley germplasm in order to identify useful lines for resistance breeding. The 143 barley lines consisted of 88 current European spring barley lines and cultivars, 33 accessions from the genebank at IPK Gatersleben, and 22 lines obtained from North American institutions. We conducted artificially inoculated field experiments with Fusarium graminearum Schwabe during two seasons. FHB severity was evaluated by repeated assessment of visual symptoms. On a set of 49 lines several trichothecene mycotoxins were analyzed. Variation for FHB severity was quantitative. The lines with lowest FHB severity were 'CIho 4196' and 'PI 566203'. Also within the European spring barley collection variation for FHB severity was highly significant. There was a significant negative correlation between plant height and FHB severity (r=– 0.55). FHB severity assessed in the field and the amount of deoxynivalenol in the harvested grains were positively correlated (r= 0.87). Several lines with a useful level of FHB resistance were found or confirmed and are recommended as crossing partners.  相似文献

Summary Sixteen promising lines (selections) of Lathyrus spp. (chicklings), comprising 11 L. sativus, four L. ochrus, and one L. cicera, were evaluated under rainfed conditions in Syria. Two locations in each of four years were treated as separate environments, to give eight environments altogether. Genotype x environment (G×E) interactions were analysed using linear regression.There was considerable variation in herbage and seed yields within both lines and environments. Genotype x environment interactions were present for both herbage and seed yields; a little over half was accounted for by the linear regression. The non-linear component was also significant, although it was smaller than that of the linear component.The most stable herbage and seed yields were obtained from L. sativus. Two selections, 347 and 311, origmating from Syria and Turkey respectively, combined both high herbage and seed yields with wide adaptation and stability, and could be considered the most widely adapted lines. Other lines were identified as suitable for favourable and unfavourable environments; in particular, all four lines of L. ochrus have great potential in frost-free climates. The importance of genotype x environment interactions in future breeding strategies is discussed.  相似文献

Summary In controlled inoculation studies with Septoria nodorum and Pyrenophora tritici-repentis, estimates of the relative proportion of each pathogen demonstrated differences in the responses of cultivars to pathogen mixtures that were not apparent from measurements of diseased leaf areas. Under field conditions estimates of the relative proportion of S. nodorum, P. tritici-repentis and S. tritici varied between field screening locations in Western Australian but also between lines within locations. Lines with known resistance to P. tritici-repentis and S. tritici, but susceptible to S. nodorum, could not be distinguished from susceptible lines on the basis of leaf area diseased or grain weight depression when S. nodorum was present in the disease complex. Such conditions, while suitable for the selection of combined resistance to these pathogens, were unsuitable for identifying resistance to individual pathogens. As symptoms were similar, the proportion of diseased leaf area sporulating with each pathogen provided a means of measuring the variation in disease development induced on lines varying in resistance. Knowledge of the components of disease and their relative importance were essential in understanding varietal response information under mixed infections of these leaf spot pathogens.  相似文献

The amount of genetic variation among inbred lines and testcrosses, and covariation between both genetic materials, are of crucial importance for selection efficiency in hybrid breeding. To estimate these quantitative genetic parameters for resistance of winter rye (Secale cereale) to head blight caused by Fusarium culmorum, 88 three-way cross hybrids, produced by crossing each of 44 S2 Carsten inbred lines with two unrelated Petkus single-cross testers, were evaluated along with the parental lines over 2 years. Resistance traits were head-blight rating and grain weight per spike relative to the non-inoculated control. Significant genotypic variation occurred among lines and in both testcross series. S2 lines displayed considerably more variation than testcross series. Genotype × environment interaction was more marked among the inbred lines, while estimates of heritability were similar for both genetic materials. Testcrosses showed heterosis for head-blight resistance. No relationship existed between S2 lines and the two testcross series for any resistance trait. This might be caused by an association between inbreeding and Fusarium-head-blight susceptibility and different inbreeding depression among the S2 population. The phenotypic correlations between the testcross series were moderate for both traits (r = 0.58, P < 0.01). In conclusion, Fusarium-head-blight resistance has to be selected at the respective heterozygosity levels.  相似文献

Summary The threshing percentage (TH%) has been suggested as a selection criterion to identify the pearl millet (Pennisetum glaucum (L.) R.Br.) lines with improved ability to fill and set grains under water limiting conditions. In this study, eight genetically diverse pearl millet inbreds and their 28 crosses produced by half diallel crossing design were used to examine range in TH%, to evaluate general combining ability (GCA) of parents and specific combining ability (SCA) effects of crosses. The results showed significant variation among parental lines for TH%. The inbreds differed for their GCA effects and crosses for their SCA effects. Parents with high TH% and positive GCA effects were identified. The results of the study revealed that both additive and dominance components, with the preponderance of later, were important in the inheritance of TH%. Heritability in narrow sense was moderate (55%) indicating that selection for high TH% might be effective.  相似文献

Random amplified polymorphic DNA (RAPD) was used to estimate genetic variability among 111 different clones of Ensete ventricosum, cultivated at 9 different collection sites in southwestern Ethiopia. PCR conditions were optimized for the amplification of DNA fragments in this crop. Out of 126 oligonucleotide primers initially screened,12 were chosen that together generated 97 reproducible polymorphic bands. Each clone proved to have a unique DNA profile, in spite of 8 duplications among the vernacular names represented in this material. The high level of genetic variability encountered is in good accordance with an outcrossing breeding system, and is also indicative of gene flow from wild enset and possibly from otherEnsete species. Genetic variation within collection sites was relatively high, with values for the Shannon-Weaver diversity index ranging from 0.44 to 0.55. A PCA (principal component analysis) demonstrated some differentiation among sites but also considerable overlapping. Partitioning the total variability at different levels with the Shannon-Weaver index shows that only14% was attributable to differences between sites and 86% to variation within sites. Pairwise comparisons of the 9 sites yielded values for Nei's unbiased genetic distances that ranged from 0.054 to 0.225.Cluster analysis as well as PCO (principal co-ordinate analysis) showed that Bonga and Chencha form a nucleus within a loose cluster, which also comprises Wolkitae, Worka, Shonae and Wondo. The remaining three sites, Answae, Setunae and Seltae, take quite isolated positions. Estimates of genetic distances did not correspond with geographical distances between collection sites. Genetic distances, as well as amount of within-site diversity, may instead be connected with the pattern of distribution for various ethnic groups in the enset-based agricultural system, and their dependency on enset as a staple food. Summary A sample of 457 wild oats (Avena sterilis L.) from the world collection maintained at the Germplasm Laboratory, Beltsville, Maryland, USA, was studied for variation of six seed characters via means, ranges, variances, and frequencies from various geographical areas. It was concuded that region-specific adaptations are responsible for occurrence of certain trait(s) in specific geographical regions; e.g., Libyan and Iraqi collections had high protein percentage, Israeli strains had high oil percentage, and the Middle East and Central Asian lines had a high number of spikelets per panicle. The main conclusion of interest here is that, within the same species, estimates of the amount of variation may vary widely, depending upon the area sampled, geographical scale of sampling, etc., presumably due to the complex interrelationship between genetic and ecological factors.Journal Paper No. J-12670 of the Iowa Agric. and Home Econ. Summary Six lines of Pisum were tested in vitro for their ability to produce somatic embryos from apices. Significant quantitative variation was observed. Inheritance of the ability to form somatic embryos was studied using a diallel cross among six different lines. About 80% of the observed genotypic variation was due to additive effects. There is a tendency for the favourable genes to be recessive. It appears that there are two genetic systems involved. Analysis of the distribution of F3 families means from a cross among two extreme lines seems to indicate the presence of a few major genes in the control of somatic embryogenesis of pea.  相似文献

A series of field experiments was undertaken in order to determine whether resistance to bean common mosaic virus (BCMV) could be incorporated into genotypes of the common bean (Phaseolus vulgaris L.) suitable for cultivation in Zimbabwe without recourse to backcrossing. Six inbred genotypes carrying the resistance-conferring alleles at the loci I and Bc-3 were crossed with five locally-adapted inbred genotypes. The first experiment comprised F3 progeny rows, each derived from a single unselected F2 plant, the second, F3 bulks selected for resistance, and the third, a comparison of selected and unselected F2-derived F4 lines. The number of days to flowering and to maturity, the incidence of mosaic and necrosis symptoms, seed yield and seed size were recorded. There was evidence that late flowering and maturity were associated with BCMV resistance in some crosses, though not strongly enough to present an obstacle to plant breeding. The incidence of virus symptoms and seed yield were influenced by genetic factors additional to the major resistance genes, and variation in seed yield was present not only between bulk populations of crosses, but also between single-row plots of lines within crosses. This indicates that early-generation selection for yield in the presence of BCMV, even among progeny selected for BCMV-resistace, is likely to be effective. However, the variation in yield among F4 lines was least in the highest-yielding crosses, which may represent a limit to successful selection for yield. Seed size was partly under additive genetic control, but there was also evidence of non-allelic interactions. There was no association between large seed size, preferred by consumers, and susceptibility to BCMV in the progeny, indicating that the association between these characters in the parent lines is fortuitous and will not present an obstacle to plant breeding. It is noted that a considerable amount of useful genetic information can be obtained without recourse to elaborate crossing schemes, provided that unselected progeny are included in experiments as controls. The evidence presented indicates that resistance to BCMV can be combined with appropriate values of maturity date, yield and seed size without the need for backcrossing.  相似文献

Summary Relationships among the traits protein percentage, grain yield, and protein yield of oats were studied with F2-derived lines in F3 and F4 from 27 matings obtained by crossing high-protein with high-yield oat lines. High-protein parents were (a) selections from an Avena sativa bulk, (b) selections from three-way matings in which an initial parent was A. sterilss, and (c) cultivars. High-yield parents were derived from backcross populations involving A. sterilis accessions as donor parents.Significnnt genetic variation existed among F2-derived lines for grain and protein yield in all matings and for protein percentage in all but one mating.Protein percentage had a highly significant negative correlation with grain yield (r=–0.33**) when pooled over all matings, but in five, these two traits were not correlated. Overall, protein percentage showed a small negative correlation with protein yield (r=–0.09*), and protein and grain yields had a high positive association (r=0.98**). F2-derived lines with both high protein percentage and high grain yield were obtained.High transgressive segregates for protein percentage occurred in two matings, for grain yield in nine, and for protein yield in 14. Summary Maintaining maize (Zea mays L.) inbred lines without genetic change is an important concern of maize breeders and seed producers. Long-time inbred lines, however, have been shown to be genetically unstable, and with selection pressure acting on the resulting genetic variation, these lines have evolved into different strains. The objective of this study was to compare maintenance of line integrity under reproduction by sib-mating and self-pollination in ear-to-row progenies. Ten lines, varying from 5 to 35 generations of previous ear-to-row selfing, were maintained for 11 successive generations under each method of reproduction. Alternate generations were compared by using a randomized complete-block design with each family of lines as a separate experiment. Data were collected for 10 plant and yield traits: pollen shed and silk emergence dates, plant and ear heights, tassel branch number, kernel row number, ear length and diameter, grain yield, and 300-kernel weight. Summarizing for F-tests over all traits and experiments, 30% were significant among sib-mated generations and 56% among selfed generations. Also, overall generation means of the two methods were different for 51% of the comparisons, and the selfed lines were less vigorous for 79% of the significant comparisons. It was concluded that sib-mating lessens the effect of genetic instability and that a reproduction system of sib-mating with intermittent generations of selfing may be more beneficial than continuous ear-to-row selfing for certain lines.Joint contribution: USDA-ARS, and Journal Paper No. J-11880 of the Iowa Agric. and Home Econ. Summary The relation has been studied between the high molecular weight glutenin (HMWg) subunit alleles and the bread-making quality of 226 lines of winter wheat (T. aestivum L.), grown in The Netherlands. The lines represented a wide range of genetic backgrounds, and had not been selected for quality, in contrast to the established varieties used by other authors.The variation in HMWg subunit genotypes accounted for about 20% of the total variation in loaf volume among the lines. Most important was the allelic variation at the Glu-D1 locus. The Glu-D1 allele encoding the subunits 5+10 was superior to its allelic counterpart, encoding 2+12. The difference in average of loaf volume between groups of lines containing 5+10 or 2+12 was negatively related with protein content of the flours. When protein content was below 9.2%, no effect of allelic variation at the Glu-D1 locus was present. Epistatic effects between the Glu-I loci also contributed to the variation in loaf volume of the lines: i.e. the effect of allelic variation at Glu-A1 and Glu-B1 depended on the allele present at the Glu-D1. The contribution of the epistatic effects was about half the contribution of the additive effects, and should therefore be included in predictive models for bread-making quality.  相似文献

RFLP variation and genetic relationships in cultivated cucumber   总被引:8,自引:0,他引:8  
Summary Two sets of cucumber (Cucumis sativus L.) germplasm were used to determine the potential use of restriction fragment length polymorphisms (RFLPs) for estimating genetic relationships. Sixteen accessions [15 domesticated variety sativus and one feral variety hardwickii (PI 183967)] of diverse origin were used to assess RFLP variation in cucumber, and to determine if genetic relationships based on RFLPs were similar to those obtained by isozyme analysis. Additionally, 35 commercial lines or cultivars were surveyed to determine genetic relationships among and within common cucumber types (narrow genetic base). The 16 accessions were surveyed with 440 low copy clones from two libraries (Pst I partial genomic and cDNA) using two restriction enzymes. Data from a subset of 104 random (mapped and unmapped) and a set of 30 mapped RFLPs were used to estimate genetic relationships among the 16 cultigens. Variability was low among RFLPs (33% of all probes) and putative alleles ( 2.2 polymorphic fragments/probe). RFLP variation between sativus lines and hardwickii (21±4%) was greater than among sativus lines (12±2%). RFLPs among the 16 accessions revealed genetic relationships which agree with those obtained using isozymes. Genetic relationships estimated using mapped and unmapped RFLPs were similar. The 35 elite lines were surveyed using a set of 40 RFLPs from 3 libraries (Pst I and EcoR I partial genomic and cDNA) to evaluate the discriminatory value of RFLPs among and between commercial cucumber types. The RFLP-derived genetic relationships among this germplasm were in agreement with predictions based on fruit type and pedigree information. Thus, RFLPs are a useful addition to the morphological characters and isozyme loci currently used for taxonomic classification and plant variety protection of cucumber.  相似文献

In strawberry, Fragaria × ananassa Duch., fruit, two different endo-β-1,4-glucanase genes, FaEG1 and FaEG3, also named Cel1 and Cel2, are expressed during the softening process that occurs during fruit ripening. It has also been suggested that FaEG3, which contains a putative cellulose-binding domain, could play a key role in fruit development, since previous attempts to down-regulate this gene through transgenesis have been unsuccessful. In this investigation, we obtained transgenic strawberry plants containing an antisense sequence of the FaEG3 gene under the control of the 35S promoter. Ripened fruit from transgenic lines (Acel lines) showed large variation in FaEG3 silencing, but fruit firmness was similar to control fruit in all the lines. Two Acel lines showing almost 95% reductions in FaEG3 mRNA levels were selected for further study. In these lines, FaEG3 down-regulation was high, from 78 to 95%, at all fruit developmental stages, whereas FaEG1 was only slightly suppressed. In spite of the high FaEG3 silencing achieved, EGase activity was not modified in ripe fruit. At the cell wall level, walls from transgenic ripe fruit contained a significantly higher amount of the 4 M KOH fraction, which is enriched in hemicellulosic polymers. The analysis of this fraction by size exclusion chromatography showed that transgenic cell walls contained a smaller amount of higher molecular mass polymers than controls. Altogether, these results indicate that FaEG3 does not play a key role either in fruit development or fruit softening. However, its silencing affects the amount and, in a minor way, the size of hemicellulosic polymers.  相似文献

In a previous study, we developed male sterile lines of Japanese bunching onion (Allium fistulosum L.) possessing the cytoplasm of a wild species, A. galanthum Kar. et Kir., by backcrossing. To evaluate seed productivity of the male sterile lines in practise, they were crossed with the male fertile line, cultivar 'Kujyo', using honeybees as pollinators under field conditions. The number of florets and seeds per inflorescence, seed set and seed germination of the material were investigated. Although variation was observed among the male sterile lines, there were several lines having seed productivity equal to cultivar 'Kujyo'. Our data demonstrate that the male sterile lines of A. fistulosum possessing the cytoplasm of A. galanthum are useful as seed parents for the commercial F seed production of A. fistulosum.  相似文献

Summary A total of 749 genotypes from a number of white clover (trifolium repens L.) cultivars and populations, many collected from dryland areas, were cloned and grown in field titles. Several morphological characters, including leaf size, number and diameter of large nodal roots (taproots), and proportion of root weight that was classified as taproot were measured. There was large variation between lines and genotypes for all characters measured, and differences between genotypes within lines are also reported. Broad sense heritability estimates were higher (>0.5) for leaflet width, petiole length and stolon diameter, than for all root character heritability estimates which were between 0.2 and 0.4.  相似文献

Summary In vitro selection of salt tolerant plants of Brassica juncea L. (Indian mustard) cv. Prakash has been accomplished by screening highly morphogenic cotyledon explant cultures on high NaCl media. Out of a total of 2,620 cotyledons cultured on high salt medium, 3 survived, showed sustained growth and regenerated shoots. They were multiplied by axillary bud culture on NaCl free medium. The salt-selected shoots retained salt tolerance following 3 month of growth and multiplication on control medium. While two of these somaclones flowered and set seeds, third one grew slowly, had abnormal leaf morphology and was sterile. The seed of the two fertile plants were sown in the field to raise R1 segregating generation. Data were recorded for field, other agronomic components and oil content. The somaclonal lines, both selected salt-tolerant and non-selected, showed tremendous amount of variation for all the characters studied. One of the two tolerant somaclones invariably showed reduced height, longer reproductive phase and higher 1000 seed weight. Based on the agronomic performance of R1 plants of these somaclones, some plants were selected and their progeny were evaluated for agronomic performance under standard field conditions and salt-tolerance in the greenhouse using sand pot culture method. Most of the lines bred true for their specific characteristics. In the greenhouse, selected salt-tolerant somaclones (SR-2 and SR-3) performed better for plant growth, yield and other agronomic traits at higher salt treatments, indicating thereby that salt-tolerance trait selected in vitro was expressed in the whole plants and is genetically stable and transmitted onto the progeny. The two tolerant lines, however, differed in their salt-tolerance during vegetative and reproductive phases as indicated by their salt-tolerance and stress susceptibility indices. The mechanism of salt-tolerance is not clear and needs to be further investigated.  相似文献
