[Objective] Phosphorus is one of three major required nutrients for cotton growth and development. A phosphorus deficiency will lead to retarded cotton growth, undeveloped roots, and the abscission of buds and bolls, which results in serious reductions in cotton yield and quality. While applied phosphorus is easily fixed in soils, it is mostly transferred to occluded P, which is not available to plants. Therefore, this study aimed to provide theoretical support for the study of response mechanisms to low phosphorus stress and the efficient phosphorus uptake and utilization of different cotton genotypes. [Method] The effects of phosphorus accumulation and utilization, leaf photosynthetic characteristics and root morphology of two genotypes of cotton (CCRI 79 and SCRC 28) were studied in hydroponic cultures having different phosphorus concentrations. [Result] CCRI 79 had higher phosphorus utilization rates of 90.92 mg·mg-1 at the low phosphorus level (KH2PO4 1.0×10-5 mol·L-1) and 23.09 mg·mg-1 at the high phosphorus level (KH2PO4 0.5 mol·L-1) compared with SCRC 28. When phosphorus was deficient, the total root length, total root surface area and total root volume of SCRC 28 increased by 13.05%, 18.78% and 10.50%, respectively, while the total root length, total root surface area and total root volume of CCRI 79 decreased significantly. [Conclusion] Here, we found that SCRC 28 had a root morphology and physiological characteristics that allowed it to adapt to low phosphorusstress, while CCRI 79 utilized phosphorus more efficiently.  相似文献

[Objective] The purpose of this study was to expand upland cotton genotypes suitable for regeneration via somatic embryogenesis, so as to provide excellent receptor materials for the genetic transformation of cotton. [Method] Six different hormone combinations were studied to establish a somatic embryo regeneration system for hypocotyl explants of Xinluzao 45, a widely planted variety in Xinjiang Province. [Result] Faint yellow callus was induced on a medium consisting of hormone combinations C1 (0.1 mg·L-1 acetic acid dichlorobenzene oxide [2, 4-D] + 0.1 mg·L-1 kinetin [KT]) or C5 (0.1 mg·L-1 2, 4-D + 0.5 mg·L-1 indo-3-butyric acid + 0.1 mg·L-1 KT). The generated callus was able to successfully induce embryogenic callus on hormone-free medium containing doubled KNO3. The embryogenic callus then developed into somatic embryos and regenerated plants. Maximum rates of callus induction and regeneration were 92.2% and 40.3%, respectively. Although no significant difference was observed in the rate of callus induction of Xinluzao 45 hypocotyls between hormone combinations C1 and C5, the incidence of induced embryonic callus was higher on C1 than on C5. [Conclusion] The somatic embryo regeneration system developed in this study provides technical support that will facilitate Agrobacterium-mediated transformation of upland cotton Xinluzao 45.  相似文献

盐胁迫下棉花K~+和Na~+离子转运的耐盐性生理机制   总被引:3,自引:2,他引:1  
为了探究棉花的耐盐机制,以中棉所49、中棉所35和中51504为材料,研究了盐胁迫对棉花幼苗的生长及K+/Na+平衡生理的影响。结果表明,150 mmol·L-1 Na Cl处理对幼苗的生长具有明显抑制作用,降低了叶片的光合速率(Pn)、PSⅡ实际光量子产额(ΦPSII)和电子传递速率(ETR),增加了非光化学荧光猝灭系数(q N)。与中棉所49和中棉所35相比,中51504的干物质累积受盐胁迫影响最小,且保持较高的Pn、ΦPSII、ETR和q N值及较低的ETR/Pn值。盐胁迫提高了棉花组织中Na+的浓度,降低了K+的浓度;但中51504组织中保持了相对较低的Na+浓度和较高的K+浓度,维持了较高的K+/Na+比;通过非损伤微测技术(NMT)测定的离子流结果也表明,中51504的根系对Na+有较强的外排能力,而对K+有较强的保留和向地上部转运能力。能够有效地调节Na+和K+的跨膜转运进而维持K+/Na+平衡是棉花耐盐的重要生理机制之一。  相似文献

中国北方主要棉区土壤供钾能力及其与产量和品质的关系   总被引:1,自引:0,他引:1  
2012―2013年在山东平原县、河北威县、新疆昌吉市代表性棉田上采集土壤(0~20 cm)和棉花样品,分析北方棉区土壤钾素状况、供钾能力及其与棉花产量和纤维品质的关系。结果表明,平原、威县、昌吉棉田速效钾含量平均分别为217、128、232 mg·kg-1,供钾能力平均分别为128.2、105.1、213.6 kg·hm-2,每生产100 kg皮棉棉花地上部分别吸收K 9.77、7.82、12.25 kg。不同棉区土壤速效钾含量与皮棉产量和地上部吸钾量相关性不同,在平原和昌吉,当土壤速效钾含量分别小于123 mg·kg-1和220 mg·kg-1时,与皮棉产量和地上部吸钾量具有显著正相关。3个植棉区棉花的吸钾量与子棉产量、皮棉产量都呈显著的正相关。棉花地上部吸钾量、纤维吸钾量和纤维含钾量与纤维品质指标的相关性不同地点差异大。  相似文献

[Objective] Early initiation and early maturity are the foundation of high yield and good quality of cotton. The purpose of this study is to determine the effects of plant growth regulators applied at the seedling and squaring stage on the early initiation of flower bud and the rate of the opened cotton boll (ROCB) during later development period, and to provide practical measures for hastening the maturity of cotton. [Method] Several plant growth regulators were applied from cotyledonary to squaring stage under greenhouse and field conditions, water was used as the control. The first fruiting branch node (indicating the initiation of flower bud), the number of bud prior to blooming and the ROCB at mid-term of boll maturation period (23 September, 2017) were compared among treatments. [Result] Under greenhouse conditions, gibberellic acid (GA3) applied at the cotyledonary stage with 140 μmol·L-1 as well as the three consecutive applications of sodium nitrophenolate (CSN, 2.23 μmol·L-1) at the cotyledonary, two-leaf and four-leaf stage made the first fruiting branch node move down by about 0.9 nodes. In field experiments, the application of gibberellin4+7(GA4+7, 288 and 576 μmol·L-1) at the cotyledonary stage significantly decreased the first fruiting branch node by about 0.4 nodes. Also, the application of 6-benzylaminopurine (6-BA, 44.4 μmol·L-1) at the three-leaf stage significantly decreased the first fruiting branch node by 0.2 nodes. However, there was no significant correlation between the first fruiting branch node and the ROCB in late September. Moreover, the application of Brassinolide (BR, 0.10 μmol·L-1) at the bud stage increased the ROCB in late September, which was mainly associated with the increased boll set in the lower and middle fruiting branches. [Conclusion] The reasonable distribution of bolls (concentrated in the lower and middle fruiting branches as well as inner fruiting sites) is more important for the earliness of cotton than lowering the first fruiting branch node.  相似文献

【目的】研究脱落酸(Abscisic acid, ABA)对棉花体细胞胚胎发生过程中下胚轴脱分化和再分化的影响,优化体细胞胚胎发生体系和初步解析脱落酸调控棉花体细胞胚胎发生分子机制。【方法】以棉花品种中棉所24(CCRI 24)下胚轴为外植体,设置5个ABA浓度0、0.02、0.04、0.06、0.08μmol·L^-1,分别以A0、A1、A2、A3、A4表示,添加至MSB(MS培养基+B5维生素)培养基诱导愈伤和胚性愈伤,研究ABA对棉花下胚轴初始细胞脱分化、愈伤组织诱导和胚性愈伤组织诱导的影响。【结果】ABA促进下胚轴初始细胞脱分化;显著提高愈伤组织的脱分化率和增殖率;0.02μmol·L^-1ABA显著提高胚性愈伤分化率,0.04~0.08μmol·L^-1ABA显著降低胚性愈伤分化率。ABA处理后胚性愈伤和非胚性愈伤的增殖率均显著提高且质地受到影响。0.02~0.08μmol ABA处理下,LBD和LBD在愈伤起始期上调表达。0.02μmol·L^-1ABA处理下,在愈伤增殖早期和中期BBM、LEC1和AGL15上调表达,愈伤增殖后期FUS3、LEA、ABI3基因上调表达。【结论】脱落酸调控的棉花体细胞胚胎发生与相关标记基因的时空性表达密切相关,这些基因表达水平的增加是ABA调控愈伤和胚性愈伤分化的分子基础。  相似文献

为了建立高效的棉花茎尖遗传转化体系,以棕色棉品种新彩棉5号为材料,采用茎尖培养和基因枪遗传转化方法,通过对凝固剂种类、活性炭含量、卡那霉素浓度、渗透处理时间、轰击压力、轰击距离等因素的优化,建立了棕色棉新彩棉5号茎尖基因枪遗传转化技术体系。结果表明,不同凝固剂种类、活性炭含量、卡那霉素浓度、渗透处理时间、轰击压力和轰击距离对棕色棉茎尖转化率有明显影响。最佳的遗传转化体系为:前渗处理4 h,轰击压力7.586 MPa(1100 psi),轰击距离9 cm,轰击后恢复培养4 d,然后在MSB+8 g·L~(-1)琼脂+125mg·L~(-1)卡那霉素筛选培养基中筛选25 d。获得抗性芽后,在添加1 g·L~(-1)活性炭的生根培养基中诱导抗性芽生根,并获得抗性再生植株。通过PCR和q PCR检测,5200个茎尖共获得16株阳性转基因植株,转化率为0.31%。该技术体系的建立为棕色棉的遗传转化奠定了基础,为彩色棉育种提供了新的材料。  相似文献

[Objective] Cotton (Gossypium hirsutum L.) seed germination, in which melatonin plays an important regulatory role, is seriously affected by soil salinization. Cotton seed germination, antioxidant enzyme activity levels and other physiological indicators were analyzed to clarify the regulatory effects of exogenous melatonin on cotton seed germination under salt-stress conditions. [Method] The experiment was conducted in a greenhouse of Hebei Agricultural University in Baoding City, Hebei Province from 2018 to 2019 using Guoxin Cotton 9 as the material. The germination rate, germination potential, seed biomass after germination, superoxide dismutase (SOD), peroxidase (POD) and catalase (CAT) activities, as well as the malondialdehyde (MDA) content, of cotton seeds were measured after exposure to different melatonin concentrations (0, 10, 20, 50 and 100 μmol·L-1) and 150 mmol·L-1 NaCl treatments. [Result] After germination, the germination rate, germination potential, radical length and seed biomass significantly decreased, as did the SOD, POD and CAT activities. However, the MDA content significantly increased under salt-stress conditions. Low exogenous melatonin concentrations (10 and 20 μmol·L-1) increased the germination potential, germination rate and biomass of cotton seeds, and promoted the elongation of radicals; however high melatonin concentrations (50 and 100 μmol·L-1) inhibited cotton seed germination and decreased seed biomass after germination. Low melatonin concentrations (10 and 20 μmol·L-1) increased the SOD, POD and CAT activities and decreased the MDA content in cotton seeds. [Conclusion] Low melatonin concentrations could promote germination and improve the salt tolerance of cotton seeds, while high melatonin concentrations could inhibit their germination. A 20 μmol·L-1 melatonin concentration is appropriate for regulating cotton seed germination under salt-stress conditions.  相似文献

分子标记辅助选择需要大批量的DNA样品,快速提取DNA的方法是提高选择效率的关键。研究比较多种植物DNA的提取方法,通过优化改进,建立了适于棉花叶片基因组微量快速提取的新方法。该方法增加缓冲液预处理及在TPS缓冲液中添加PVP40,采用PCR板批量取样、微型手电钻磨样,省去了蛋白抽提、DNA沉淀、洗涤、风干、溶解等步骤,免去氯仿和异戊醇的使用。本方法具有低成本、快速、高通量、操作简单等特点,而且能确保提取的棉花DNA的浓度和纯度达到要求,满足大批量样本SSR分子标记辅助育种工作的需要。  相似文献

【目的】筛选防治烟蓟马(Thrips tabaci Lindeman)的高效生物源农药,探究其对烟蓟马3种主要解毒酶细胞色素P450单加氧酶(Cytochrome P450 monooxygenase,P450)、谷胱甘肽S-转移酶(Glutathione S-transferase,GST)和羧酸酯酶(Carboxylesterase,CarE)活性的影响。【方法】以棉田采集并室内稳定饲养的烟蓟马为供试虫源,用浸叶法测定7种生物源农药对烟蓟马若虫的毒力,选择活性好的3种药剂开展田间药效试验,并在这3种生物源农药亚致死浓度(LC25)剂量处理后测定烟蓟马体内3种主要解毒酶活性。【结果】(1)7种生物源农药对烟蓟马的毒力从高到低依次为乙基多杀菌素、多杀霉素、苦参碱、印楝素、除虫菊素、鱼藤酮、藜芦碱。乙基多杀菌素、多杀霉素和苦参碱对烟蓟马的致死中浓度(LC50)分别为0.011 mg·L-1、0.274 mg·L-1、1.479 mg·L-1。(2)田间药效试验发现,每666.7 m2用2 g、4 g、6 g的60 g·L-1乙基多杀菌素悬浮剂和10 g的5%(质量分数,下同)多杀霉素悬浮剂处理,施药后7 d防治效果均在90%以上,优于1.3%苦参碱水剂和对照药剂30%噻虫嗪悬浮剂各剂量的防效。(3)LC25剂量处理烟蓟马48 h后,乙基多杀菌素处理的烟蓟马体内P450、GST和CarE活性升高,苦参碱和多杀霉素处理后CarE活性升高,噻虫嗪处理后P450和CarE活性升高。【结论】乙基多杀菌素、多杀霉素和苦参碱对烟蓟马的毒力较高,可用于防控棉田烟蓟马;CarE可能参与了烟蓟马对多杀霉素和苦参碱的解毒代谢,3种主要解毒酶可能均参与了烟蓟马对乙基多杀菌素的解毒代谢。  相似文献

[Objective] This study investigates the effects of exogenous proline on the growth, physiological responses, and proline metabolism of cotton (Ekang No.8) seedlings treated with varying boron (B) concentrations. [Method] We conducted a randomized experiment with six treatments using the hydroponic method in a greenhouse at Huazhong Agricultural University. We applied three different concentrations of boron, including 0.02 μmol·L-1 (low-concentration B), 2.5 μmol·L-1 (medium-concentration B), and 100 μmol·L-1 (sufficient-concentration B). The first two are boron-deficient treatments. We also applied an exogenous proline treatment at 20 μmol·L-1(0 μmol·L-1 as control). When any significant difference among the treatments were observed, the related indicator was measured. [Result] The results showed that exogenous proline inhibited the growth of cotton seedlings under sufficient-concentration B treatment while promoting the absorption of B by roots under low-concentration B treatment. The application of exogenous proline to the seedlings under low-concentration B treatment reduced the contents of proline and H2O2 in leaves but increased the accumulation of MDA and H2O2 in roots. The activities of SOD and antioxidant enzymes (APX) in the roots and leaves were dramatically enhanced. Conversely, POD activity reduced significantly and there was no significant change in CAT activity relative to low-concentration B treatment. More importantly, we found that the application of exogenous proline under B deficiency increased the activities of P5CS, P5CR, OAT (synthetase), and PRODH (degrading enzyme) in proline metabolic pathways. [Conclusion] Applying exogenous proline under sufficient-B concentration inhibits growth. The application of proline to seedlings under low-B concentration promotes the absorption of B by roots, increases the activity of APX, and decreases the membrane lipid peroxidation in leaves. A B deficiency leads to proline accumulation in plants. The addition of proline under low-B concentration can reduce the proline content in leaves, which is caused by affecting the critical synthetase and catabolic enzyme activities in the proline metabolism pathway (Glu and Orn pathway). The main reason for this occurrence is the significant increase of proline dehydrogenase (PRODH) activity.  相似文献

The objective of this study was to investigate the effect of the different concentrations of the seed film coating salicylic acid on the cold tolerance of cotton seedlings using the sand culture method. The physiological response of cell membrane permeability, protective enzyme activity and osmotic adjustment of cotton seedling were measured before and after at low temperature stress and at normal temperature. The results showed that 1-10 mmol·L-1 of seed film coating with salicylic acid seed coating treatment could effectively alleviate damage to cotton seedlings at low temperature, and the accumulation of the relative electrical conductivity and malondialdehyde were reduced on cotton seedlings. The activity of superoxide dismutase, peroxidase, catalase, and soluble protein, soluble sugar and proline contents increased sharply. These results indicate that film coating with salicylic acid can alleviate the effects of low temperature stress on seedling growth inhibition and improve the cotton seedlings by providing chilling tolerance, and the optimum treatment concentration salicylic acid is 5 mmol·L-1.  相似文献

杂交棉中棉所70后代分离群体产量品质的表型变异分析   总被引:3,自引:3,他引:0  
中熟转基因抗虫杂交棉中棉所70具有高产优质特点,由高产且综合性状优良的抗虫棉SGK 156与品质优异的渐渗系材料901-001为亲本配制。本文对该杂交种F1及其自交后代F2、F2:3、F2:4和F2:5共4个分离世代材料的皮棉产量和品质性状进行了详细调查和相关性分析。结果表明,杂种F1纤维品质表现突出,上半部平均长度平均为33.00 mm,断裂比强度为33.3 c N·tex-1。在分离群体中,各性状均存在大量的超亲变异,衣分与纤维长度及强度为极显著负相关,与马克隆值为极显著正相关,说明棉花产量与品质间的同步改良仍存在困难。此外,从F2:5株系中挑选出11个优异品系,其中4个材料的皮棉产量超对照,上半部平均长度均高于31.80 mm,断裂比强度在31.0 c N·tex-1以上,铃重均大于6.26 g,衣分最高为38.64%,这表明通过高代自交是可以筛选出具有优异产量及品质性状的品系。本研究为棉花数量性状遗传、基因功能分析、基因聚合育种改良等研究储备了大量的基础材料。  相似文献

[Object] The study was conducted to investigate the plant architecture characteristic of direct sowing cotton planted after barely harvested with high yield and centralized boll-setting in the Yangtze River basin. [Method] In 2014, the cultivars Guoxin 12-1, Yijimian and Lumianyan 36 were used and the two conventional fertilizer (CF) application rates (namely pure nitrogen 45 kg·hm-2, 150 kg·hm-2) and Guoxin 12-1 was used in 2015. Two slow release fertilizer (SR) utilization rates (namely pure nitrogen 150 kg·hm-2 and 225 kg·hm-2) and two SR topdressing at different growth stages (namely 100% topdressing at seedling stage, 70% topdressing at seedling stage + 30% at flowing stage) were set with CF (pure nitrogen 150 kg·hm-2) and no fertilizer treatment as the controls. [Result] While the pure nitrogen (CF) amount was 150 kg·hm-2, the seed cotton yield of Guoxin 12-1 were 4 014.72 kg·hm-2. In 2015, the seed cotton yield for the treatment, application SR (pure nitrogen 150 kg·hm-2)and application ratios of seedling stage and flowering stage of 100% and 0, respectively, increased by 30.96%. The ratios of bolls setting from 08-15 to 08-30 to total bolls (RBT) for the two treatments were 31.8% and 26.1%, respectively. Then a significantly positive correlation between the seed cotton yield and RBT was found(r2014=0.948**, r2015=0.976**). Based on the analysis of relationship between the plant architecture indexes and RBT, plant architecture characteristics of cotton population with high yield and centralized boll-setting was proposed. [Conclusion] These indexes would be used to supervise the cotton culture management to achieve high yield and centralized boll-setting for the direct sowing cotton planted after barely harvested.  相似文献

【目的】本研究旨在科学合理地利用浅层咸水资源。【方法】依托短期定位试验开展了在第3年和第4年不同咸水利用方式下(CK:造墒和蕾期灌淡水;T1:造墒和蕾期灌咸淡混配矿化度3 g·L^-1的微咸水;T2:淡水造墒蕾期灌矿化度5 g·L^-1咸水;T3:造墒和蕾期灌矿化度5 g·L^-1咸水;T4:淡水造墒蕾期不灌水)棉花长势、叶绿素荧光参数、土壤盐分累积及其运移的变化。【结果】T1和T2处理的齐苗率、株高、干物质质量、叶面积指数、叶绿素荧光参数、产量及霜前花率与CK无显著差异,土壤盐分含量较CK有所增加,但未对棉花生长产生明显抑制。T3处理的棉花长势指标、叶绿素荧光参数较CK显著降低,0~100 cm土壤盐分含量较CK明显增加。【结论】从土壤质量安全和咸水高效利用的角度考虑,连续4年用3 g·L^-1的咸淡混合水灌溉或用淡水与5 g·L^-1的咸水轮灌不仅能节约淡水,且不影响棉花产量。本研究结果为当地在棉花生产中安全利用咸水提供技术参考。  相似文献

[Objective] The aim of this study is to evaluate nitrogen efficient cotton germplasms and improve nitrogen use efficiency. [Method] Eighty cotton germplasms were selected and evaluated in the hydroponic experiment under low (0.25 mmol·L-1) and high (5 mmol·L-1) nitrogen concentration. Different traits for screening were identified and nitrogen use efficiency types were classified. Field experiments were also performed for comparison and confirmation of the identified germplasms. [Result] The results showed that there were significant differences in the total plant dry matter, shoot nitrogen accumulation and nitrogen absorption efficiency in cotton germplasms at the two nitrogen levels. Based on coefficient of variation, principal component analysis and correlation, six traits including total plant dry matter, shoot dry matter, root dry matter, total nitrogen accumulation, shoot nitrogen accumulation and nitrogen absorption efficiency were used as screening indicators. According to the Heatmap clustering analysis and the nitrogen efficiency comprehensive index, two germplasms (Lu05R59 and CCRI 69) were identified as low nitrogen tolerant and nitrogen efficient, and two germplasms (Coker 201 and Xinluzhong 30) as low nitrogen sensitive and nitrogen inefficient. The results of field experiment were consistent with the results of the hydroponic culture at the seedling stage. [Conclusion] It was finally determined that Lu05R59 and CCRI 69 were the low nitrogen tolerant and nitrogen efficient germplasms, and Coker 201 and Xinluzhong 30 were low nitrogen sensitive and nitrogen inefficient germplasms. The results of these studies provide the possibility for screening and rapid identification of nitrogen use efficiency in cotton at the seedling stage, and provide the ideal materials and theoretical basis for further study of cotton nitrogen efficient.  相似文献

【目的】研究盐胁迫下施钾调节棉花纤维断裂比强度的糖代谢机制,为盐碱地适量施钾提供理论依据。【方法】以中棉所79(耐盐型)和泗棉3号(盐敏感型)为试验材料,通过设置3个土壤电导率(低盐1.68~1.78 dS·m^-1、中盐6.21~6.42 dS·m^-1、高盐10.59~11.08 dS·m^-1),3个施钾量(0、150、300 kg·hm-2),研究了盐胁迫下施钾对棉花纤维断裂比强度、纤维加厚发育期纤维素累积和蔗糖、β-1,3-葡聚糖及相关酶活性的影响。【结果】(1)盐胁迫显著降低了棉花纤维断裂比强度;施钾显著缓解了中、高盐胁迫下盐分对纤维断裂比强度的影响,但施钾150、300 kg·hm^-2处理间无显著差异。盐碱地施钾,中棉所79的纤维断裂比强度增幅高于泗棉3号。(2)盐胁迫降低了纤维加厚期纤维素的累积量,降低了纤维蔗糖含量并提高了β-1,3-葡聚糖含量;盐碱地施钾则提高了纤维加厚发育期纤维素最大累积速率,提高了花后28 d磷酸蔗糖合成酶以及β-1,3-葡聚糖酶的活性,提高了蔗糖及β-1,3-葡聚糖含量,且施钾缓解作用随盐胁迫程度加重而逐渐减弱。施钾条件下,中棉所79的纤维素最大累积速率及β-1,3-葡聚糖酶活性的增幅高于泗棉3号。【结论】盐碱地适量施钾可缓解盐胁迫对棉花纤维断裂比强度的影响。  相似文献

[Objective] Novel germplasm of cotton was generated for development of cultivars with high resistance to glyphosate. [Method] The glyphosate resistance gene G10eve was preliminarily analyzed using bioinformatics methods and introduced into 'Coker 312' via Agrobacterium-mediated transformation. The expression of G10eve was estimated by PCR (Polymerase chain reaction) and qRT-PCR (Quantitative real-time PCR). The content of shikimic acid and tolerance to glyphosate were determined in response to application of glyphosate to 'Coker 312' and transgenic plants. [Result] A total of 28 independent positive transgenic lines were obtained. The transformation efficiency and the rate of seedling differentiation were up to 49.3% and 40.6%, respectively. PCR and qRT-PCR analysis showed that G10eve was integrated into the cotton genome, and the G10eve expression level differed significantly among the transgenic lines. At the cotyledon stage, the transgenic cotton showed resistance to 8 mL·L-1 glyphosate, whereas 'Coker 312' showed phytotoxicity to the 2 mL·L-1 glyphosate treatment. No significant accumulation of shikimic acid was detected in transgenic plants before or after glyphosate treatment, which indicated the typical physical characteristic of glyphosate resistance. [Conclusion] Overexpression of G10eve in cotton improved resistance to glyphosate. Thus, novel germplasm of cotton resistant to glyphosate was generated.  相似文献

【目的】本研究旨在揭示棉花钾离子(K~+)吸收机制。【方法】以国欣棉3号和中棉所41为材料,应用病毒诱导基因沉默技术研究棉花K~+通道基因GhAKT1和K~+转运体基因GhKT2的功能。【结果】在低水平供K~+(0.1 mmol·L~(-1))和充分供K~+(2.5 mmol·L~(-1))条件下,沉默GhAKT1和GhKT2基因使棉花幼苗的K~+积累量分别降低15%左右和25%以上。此外,沉默GhAKT1和GhKT2基因导致棉花幼苗对K~+的最大吸收速率和亲和能力出现不同程度的下降。药理学试验结果表明,在外界K~+浓度低于250μmol·L~(-1)的条件下,高亲和系统对棉花K~+吸收的贡献大于K~+通道;而且GhAKT1蛋白可能是1个对NH4+比较敏感的组分。【结论】棉花K~+通道蛋白GhAKT1和K~+转运体蛋白GhKT2具有吸收K~+的功能,而且表现出双亲和吸收K~+的特性,可作为改善棉花钾营养的候选基因。  相似文献

棉花短季直播栽培模式对产量构成及纤维品质的影响   总被引:4,自引:1,他引:3  
为探索棉花短季直播高产栽培技术模式,2014年在湖南农业大学浏阳试验基地,采用"311"最优回归设计方案研究了播期、密度和施氮量对早熟品种JX0010的产量构成及纤维品质的影响。对不同处理的棉铃时空分布、棉花产量及纤维品质的分析结果表明:不同处理棉铃时间分布上以秋桃比例最大,空间分布上纵向分布以中下部成铃数所占比例大,横向分布以内围铃为主。根据结果,建立了3个栽培因子施氮量(x1)、密度(x2)、播种期(x3)与子棉产量的多项式回归方程:y=213.522+3.9331x1+6.414x2-21.4619x3-5.7541x12-4.321x22-7.5348x32~(-1)1.982x1x2-0.1645x1x3+3.5747x2x3。分析回归方程得出,单因子效应表现为播种期密度施氮量,交互作用表现为施氮量与密度的互作效应密度与播种期的互作效应施氮量与播种期的互作效应,根据回归方程得到子棉高产的最佳组合为施氮量202.35 kg·hm-2、密度6.20万株·hm-2、播种期5月19日,产量为5510.25 kg·hm-2。棉花纤维品质性状受影响的顺序为马克隆值断裂伸长率断裂比强度上半部平均长度长度整齐度指数。  相似文献
