Amplified fragment length polymorphism (AFLP) was used in genetic fingerprinting of 22 elite Chinese Populus deltoides and Populus × canadensis accessions. The results indicated that each of the nine AFLP primer pairs selected generated fingerprint profiles that were unique to each of the accessions. Therefore, each accession was definitively identified by any of the nine primer pairs. Furthermore, the inter-accession genetic relationships inferred based on 461 polymorphic fragments from the nine AFLP primer pairs were largely consistent with phylogenetic relationships based on morphologic traits. Bootstrap analysis showed that three AFLP primer pairs were required to obtain genetic similarity values with a maximum CV of 10% while 10 AFLP primer pairs could give a maximum CV of 5%. Thus, AFLP can readily be applied for a rapid and accurate evaluation of the degree of similarity between poplar cultivars. In this study, the number of AFLP fragments used was sufficient to establish a reliable estimate of genetic similarity among accessions, with a maximum CV of 5.12%. Therefore, the information on the genetic relationships among the poplar accessions generated in this study in connection with knowledge on agronomic traits may have an impact on poplar breeding and planting in China.  相似文献

Amplified fragment length polymorphism (AFLP) is a powerful DNA fingerprinting technique for studying genetic relationships and genetic diversity in insects. However, the crucial prerequisite for AFLP analysis is to extract DNA of high quality. In this study, we evaluate four different protocols (SDS method, improved SDS method, CTAB method and a complex method with SDS and CTAB) for isolating DNA from the seabuckthorn carpenter moth (Holcocerrus hippophaecolus (Lepidoptera: Cossidae)). The results indicate that the CTAB method does not produce DNA suitable for AFLP analysis. The SDS method and the complex method with SDS and CTAB are comparatively time-consuming and resulted in low yields of DNA and were therefore not used for AFLP assay. The improved SDS method is recommended for preparing DNA templates from H. hippophaecolus for AFLP analysis.  相似文献

Baobab leaves form an important part of the local diet in Sahel countries and elsewhere in Africa. Existing leaf nutritional data and agroforestry performance information are based solely on Adansonia digitata L., the baobab of continental Africa. The introduction potential of Adansonia species from the center of diversity in Madagascar and from Australia remains untapped. To assess this potential, the mineral contents and B1 and B2 vitamin levels of dried baobab leaves were determined for five-year old trees of A. digitata, A. gibbosa (A. Cunn.) Guymer ex D. Baum, A. rubrostipa Jum. & H. Perrier (syn. A. fony Baill.), A. perrieri Capuron and A. za Baill. grown in an introduction trial in Mali. Nutritional data were evaluated against survival and vigor to identify promising germplasm. Leaf vitamin and crude protein contents were highest in the Madagascar species, especially A. rubrostipa (B1 88 mg 100 g−1, B2 187 mg 100 g−1, protein 20.7% dry weight). However, the local species far outperformed the introductions in survival, tree height, basal diameter and resistance to termites. We suggest grafting as a way of harnessing the vigor of well-adapted local baobab varieties to the superior nutritional profiles of A. rubrostipa and others. Cross-species grafting tests in Adansonia were successful, thus creating new agroforestry possibilities with different scion/rootstock combinations.  相似文献

The Baobab tree (Adansonia digitata L.) is a key multipurpose species for the African region. In the recent years there has been an extended commercial interest for different A. digitata products. As a spectacular African key species there has been a growing interest from NGO′s and various research groups. A research group, focussing on the following countries Burkina Faso, Mali and Niger, has participated in a concerted research action with cross counter disciplinary cooperation between plant physiology, population genetics, tree breeding, food science, and socioeconomics. This paper presents a review and the way knowledge gaps are being addressed using the above mentioned approach. The overall work was initiated in 2005–2006 when a large collection of A. digitata seeds was carried out in 15 African countries. Fourteen populations were selected in Burkina Faso, Mali, and Niger including more than 400 superior trees, and many of the trees have been selected along with farmers in a participatory process. Studies have been initiated in all countries on geographic variation of growth, adaptive and phenological traits. This includes studies in the nursery, establishment of international provenance series, seed orchards and studies of gene flow and phylogeographic variation with various markers. Specific nursery trials have been established in order to study provenance and progeny performance related to drought stress. Several stress related characteristics are being measured. Preliminary observations showed large morphological variation between African provenances of A. digitata. Food properties of plant parts have been studied within and between populations. This includes studies of total biomass production, vitamin A, B1, B2, and various minerals. The influence of stress will be related to these parameters. Fruits, which are rich in sugar and vitamin C, are used as an ingredient in juice and other foods. Seeds can be used directly as food ingredient or in fermented condition (Maari). The fermentation process was previously poorly described. Therefore, the microorganisms associated with Baobab seeds fermentation have been identified and a starter cultures for control production has been proposed. The efficiency of dissemination of superior A. digitata trees depends on vegetative propagation. Various grafting methods are currently being tested in cooperation with farmers. Micropropagation is tested as well as a method for clone propagation. The accumulated knowledge will be applied for a domestication strategy of A. digitata in Burkina Faso, Mali and Niger. The outcome from this study will be guidelines for protection and management of the germplasm of A. digitata resources.  相似文献

The baobab tree (Adansonia digitata L.) is of large socioeconomic importance. However, domesticated plant material of baobab is rarely available to African farmers, and very limited pre-breeding knowledge is available. To evaluate genetic differentiation between geographical origins we compared vigor and leaf morphology of trees from 17 West African and 13 East African provenances of baobab. Seedlings were grown and evaluated in a common garden test at Bamako, Mali. Germination was assessed and growth measured 3 and 12?months after germination. Leaf morphology (leaf length, leaflet number, petiole length and diameter and leaflet border) were assessed after 12?months. Significant differences between the provenances were observed for both growth rate and leaf morphology. West African provenances in general grew faster than East African provenances, but leaf characters did not reveal a particular geographic structure, and the correlations between geographic distances and multivariate Mahalanobis distances were not significant. The correlations between leaf morphological traits and climatic data were in general low at the provenance level. However, the number of leaflets was significantly higher for provenances from drier areas. Two years increment at a field site also varied significantly between the provenances, where West African provenances confirmed their tendency to higher growth rate. Still, trees are young and results regarding both growth data and leaf characteristics should be confirmed when the trees are closer to maturity.  相似文献

We analyzed the phenotypic variation of baobab (Adansonia digitata L.) fruits from Mali to define the domestication potential of this species. 269 trees, selected from 10 provenances distributed along a rainfall gradient, were characterized. Five fruits were sampled from each tree. Total individual fruit weight was partitioned into shell, pulp, and seed weight. Ratios were calculated between pulp + seed and total weight, and between pulp and seed weight. For all the measured fruit traits, we detected significant differences between provenances, as well as between trees from the same provenance. Assuming that the measured traits are under genetic control, the latter facts indicate that there are considerable opportunities for tree selection on a local scale. However, candidate plus trees with specific properties, e.g., extremely high pulp or seed weight, were found only in some locations. Mean pulp yield per fruit might be considerably increased by selecting the 5% trees with the highest pulp weight (mean: 45 ± 1 g, best 5%: >100 g). The same might be true for seed production (mean: 71 ± 2 g, best 5%: >150 g). Also the ratio between pulp and seed weight can be an important fruit characteristic for selection: in fruits with a low ratio, relatively more pulp sticks to the seeds when separating the two fruit parts by grinding, and thus more pulp will be lost for further processing. We identified several trees with a high pulp weight combined with a high ratio between pulp and seed weight. It is concluded that there is considerable phenotypic variability in traits of baobab fruit in Mali, offering opportunities for cultivar selection.  相似文献

Among the priority species identified for domestication in the Sahel region of Sub-Sahara Africa is the baobab (Adansonia digitata L.) tree. The quantification of the variation in nutrient content and fruit morphological traits of trees is one of the most important steps in identifying superior planting material for domestication. Ten provenances in Mali covering all the different agro-ecological zones were selected and the fruit morphological traits, vitamin C, calcium, iron and colour were studied. Mean pulp content was 2,149 ± 1,117 , 2,406 ± 776  and 25 ± 17 mg kg?1 for vitamin C, calcium and iron, respectively. Fruit pulp colour varied from white, creamy to pink and a significant correlation between pulp vitamin C content and reflectance in the green and blue bands was observed. Significant negative correlations were found between rainfall and pulp vitamin C content and between mean annual temperature and fruit and pulp weight and pulp fraction, suggesting that these traits are influenced by the environment. Pulp iron content correlated positively with topsoil sodicity and base saturation. Similarly, pulp vitamin C content correlated positively with topsoil sand fraction. Pulp reflectance in the blue and green bands correlated negatively with topsoil pH water and base saturation, respectively. The variation in nutritive and morphological traits offers the opportunity for selecting plus trees with a combination of desired traits for domestication.  相似文献

从表型性状和分子水平上研究了小果油茶农家品种间的遗传差异及亲缘关系。结果表明:小果油茶农家品种在果实、叶和花性状上的变异极为丰富,每500 g鲜果数的变异系数最大(76.91%),不饱和脂肪酸含量变异系数最小(3.31%);果实性状的变异程度高于叶和花性状;果纵径和果横径与叶长、叶周长和花冠直径呈显著或极显著正相关;农家品种宜春白皮中籽表型性状的总变异程度高于龙眼茶、羊屎茶和珍珠茶。利用25对AFLP引物组合在4个小果油茶农家品种和普通油茶中共扩增出921条带,其中多态性条带706条,多态性条带比率达75.7%;羊屎茶与宜春白皮中籽的遗传相似系数最大(0.698 2),龙眼茶与普通油茶的相似系数最小(0.558 1)。AFLP标记和表型性状的聚类都可将小果油茶农家品种和普通油茶分开,但对小果油茶农家品种间的归类并未完全一致。  相似文献

Domestication of Uapaca kirkiana Müell. Arg is a high priority for improving rural livelihoods of smallholder farmers in southern Africa. Domestication efforts require knowledge of adaptive traits and intra-specific variation. Morphological traits and amplified fragment length polymorphic (AFLP) markers were used to assess genetic variation in twelve provenances of U. kirkiana collected from southern Africa. Assessment of morphological traits showed significant differences (P < 0.05) between provenances. Provenances from Zimbabwe and Zambia showed faster growth than those from Malawi (except Phalombe) and Tanzania. Morphological traits exhibited strong genetic differentiation between provenances and within provenances. The degree of provenance differentiation of traits (Q ST = 0.03–0.139, Q ST mean = 0.092) was in the same range with that of AFLP loci (F ST mean = 0.089) detected among provenances and within provenances (0.002 < F ST < 0.259). The differentiation for some morphological traits could be attributed to local adaptation and human selection of the U. kirkiana trees at the site of origin. Mean Nei’s (H) genetic diversity of AFLP showed high diversity within the provenances (H = 0.181–0.321, H mean = 0.256). An analysis of molecular variance (AMOVA) revealed higher genetic variation (90.8%) within provenances than among provenances (9.2%). There was no geographical pattern of variation in growth and morphological traits among the seed sources. Chipata provenance from Zambia was the most diverse while Mapanzure from Zimbabwe was the least diverse, though it was superior in height and earliest in fruiting. The pattern of genetic diversity indicates low selection in some areas and high gene flow which would be counteracting it. Therefore regional and country collections and conservation strategies should consider differences by focussing on the main range of the species by paying particular attention to unique populations within countries.  相似文献

Accurate identification of varieties (clones) and knowledge of their genetic relationships are essential for poplar breeding and variety management. In this study, 21 elite poplar varieties of Tacamahaca and Aigeiros in China were fingerprinted using amplified fragment length polymorphism (AFLP) markers. Four AFLP primer pairs developed generated totally 181 AFLP polymorphic fragments, and in particular, each primer pair generated fingerprint profiles specific to each of the tested varieties. The genetic relationships among the varieties were evaluated by dendrograms and multidimensional scaling (MDS). The results showed that the tested poplar can be classified into five groups, and indicated the clear separation of varieties of different sections of poplar and the primary distinction between native and exotic poplar varieties. This study indicated that tested poplar varieties could be identified by their fingerprint profiles and that genetic relationships deduced from the study were consistent with their genealogy. The green oak leaf roller (Tortrix viridana L.) is a major pest on oaks. The green oak leaf roller is one reason within a disease complex for the oak decline, and it leads to defoliation of oaks in spring. In Germany, Quercus robur L. is mainly affected. The partitioning of genetic variation within and among populations of forest pests is linked to their capacities for migration. And the capacity for migration is the basis for dispersal of the pest species to different host trees and host populations and, therefore, for further outbreak events. Thus, the within population spatial genetic structure (SGS) of T. viridana was investigated based on nuclear (AFLP) markers to analyse the dispersal capacity of this insect pest species at a very small spatial scale. A total of 401 individuals of the green oak leaf roller from four stands in North Rhine-Westphalia (Western Germany) were examined. In three of four populations, the AFLP markers revealed clearly SGS up to 40 m, which can be explained by the mating behaviour within this species.  相似文献

Fruit trees play an important nutritional role for livelihoods of rural people in the West African Sahel through provision of energy and nutrients such as vitamins, minerals, and proteins. Research on the domestication of local fruit trees has started recently through projects concentrating on some of the most important indigenous species of dry West Africa, i.e. Adansonia digitata, Parkia biglobosa, Tamarindus indica, Vitellaria paradoxa and Ziziphus mauritiana. We present a status of finalised and ongoing domestication research with the aim of defining research gaps that would need to be covered by future research activities to obtain higher yields and better quality fruits. Germplasm collection in central West Africa has been intense compared to elsewhere in the species?? distribution areas, but conservation status of the material is poor since it is only planted in few trials. Knowledge of genetic parameters, especially for fruit traits, is almost absent, but characterisation of genotypes is underway for some of the species. Mating systems and patterns are still unknown for many species. Efficient vegetative propagation based on simple techniques was shown to be possible for all species except P. biglobosa. In order to secure immediate as well as long term gains, we recommend combining clonal propagation of selected plus individuals with recombination and breeding of selected genotypes. We discuss whether local institutions in the Sahel have the financial capacity to carry out long term breeding programmes, and suggest that efforts should be made to find new ways of disseminating improved germplasm.  相似文献

A common objective in commercial reforestation is to plant seedlings that will produce superior adult trees. Long forest management planning horizons and life spans of most forest trees complicate the prediction of growth and performance of adult trees originating from planted seedlings of unknown genetic origin. We used multivariate statistics to identify and rank a set of seedlings characteristics that could predict the performance of mature trees in Pinus ponderosa. Results indicated that metabolic heat rate (q), a measure of total metabolism, of one-year-old foliage during the peak growth in May, was the most important seedling characteristic that predicted mature-tree performance. Increased metabolic heat rate in seedlings corresponded with greater vigor of mature trees. Additionally, seedling basal stem diameter (D), height (H), and needle length (NL), measured in November, were, in order of importance, other variables that defined the vigor class of the mature clones. However, these seedling morphological characteristics correlated negatively with vigor classification of the mature clones, contradicting the notion that greater D and/or H during the seedling stage may indicate a greater vigor at maturity.  相似文献

[目的]研究山地木麻黄种源间抗风性、生长及形质性状的遗传变异规律,为山地木麻黄的良种选育和种质资源的合理开发利用提供科学依据。[方法]以27个山地木麻黄种源为试验材料,于造林后2、5、7 a时测定山地木麻黄种源的树高、胸径、单株材积和保存率等数量性状,并于造林后7 a时调查主干分叉习性(AP)、主干通直度(SFS)、侧枝密度(DPB)、侧枝直径(TPB)、绿色小枝长度(LDB)、侧枝分枝角(APB)、侧枝长度(LPB)等形质性状以及抗风性(RES),通过方差分析、相关性分析及遗传参数估算揭示其遗传变异规律。应用坐标综合评定法对山地木麻黄种源进行综合评定。[结果]表明:造林后2、5、7 a时,27个山地木麻黄种源间保存率和抗风性差异显著(P0.05);对造林后7 a时保存率较高的18个种源进一步分析显示,上述3个年份各种源间树高、胸径和单株材积等生长性状均存在极显著差异(P0.01);7 a时,TPB、APB、LDB、AP和SFS等形质性状在种源间亦存在显著或极显著差异;生长性状的种源遗传力明显高于形质性状,二者分别受中度或中度偏下和低度遗传控制;随着林龄的增长,树高的遗传变异系数变化不大,而胸径和单株材积的遗传变异系数呈先增加后降低的趋势,树高、胸径、单株材积的遗传变异系数分别为11.89%~12.30%、11.67%~13.67%、30.20%~38.11%;7 a时,形质性状的遗传变异系数为3.84×10~(-5)%~5.56%。性状间相关分析表明:树高作为山地木麻黄早期选择性状较适宜。[结论]依据坐标综合评定法,筛选出17877、19489和19490等3个优良种源,可在生产上大面积推广。  相似文献

Adansonia digitata (baobab tree), a multipurpose tree species, occurs throughout semi-arid and arid zones of Africa. Its survival is, however, threatened by bush fire, over-exploitation, grazing and a lack of natural regeneration. The extent of variation in fruit characteristics, seed germination and seedling traits of the baobab tree in Benin, was evaluated at climatic zone level. 1,200 fruits were sampled in each of the three climatic zones of Benin for morphological assessment and to assess germination rate and seedling growth dynamics according to the climatic zones, the used substrate and the scarification of the seed coat. There were significant differences in fruit characteristics not only between climatic zones but also between individuals from the same zone and within-trees. Using mechanical scarification on freshly-collected baobab seeds negatively affected the germination rate of baobab seeds sampled in the Guinean and Sudano-Guinean zones of Benin. The best-germination rate was recorded for non-treated seeds from the Guinean zone, up to 57% on day 25. All seeds germinated best on the sand substrate, but supplying organic matter promoted further seedling growth after 11 days of germination. Based on these observations we propose some strategies for efficient ex situ conservation of baobab in Benin.  相似文献

Baobab (Adansonia digitata L) belonging to Bombacaceae family, is one of the most widely used indigenous priority tree species in sub-Saharan Africa, valued in the cosmetic industry for its seed oil, and powdery fruit pulp for juice making. Baobab has high potential for domestication in southern Africa, therefore understanding its genetic diversity and population structuring is warranted. The study investigated the level of genetic diversity and differentiation of five populations of A. digitata L. sampled from four diverse silvicultural zones in Malawi. Variation at nine microsatellite loci were examined in 150 individual trees. Low mean genetic diversity was expressed through genetic diversity indices: Nei’s genetic diversity (h, 0.18 ± 0.03), Shannon Information Index (I, 0.21 ± 0.07), observed number of alleles (na, 1.47 ± 0.10), effective number of alleles (ne, 1.23 ± 0.04) and percentage polymorphic loci (pp, 48 %). The low genetic variation found is attributed to the population growing in marginal areas of genetic centre of diversity of the species, anthropogenic factors and founder effects. Moderate genetic differentiation was observed among populations (Gst = 0.13) indicating the presence of a large number of common alleles resulting in a homogenisation effect. Clustering of individual trees by genetic similarity coefficients indicated that mainland trees were genetically closer than the trees on Likoma Island. Mantel’s test showed a weak positive insignificant correlation (Z = 0.12; P = 0.64) between genetic distance among populations and actual distance on the ground implying that geneflow was not directly influenced by isolation by distance. The results suggest that seed distribution and tree improvement should recognise the presence of ecotypes and conservation measures should protect all the populations due to existence of private alleles which are of adaptive importance.  相似文献

Acacia senegal (L.) Willd. is an African leguminous multipurpose tree species belonging to subfamily Mimosoideae and subgenus Aculeiferum, highly valued for gum arabic production. This study estimated the genetic diversity within and among Kenyan populations of the species in the Bulla Sambul, Kutulo, Wamba and Meisori populations based on 12 quantitative traits, which included; tree height, diameter at breast height (DBH), number of branches, bark thickness, gum weight and various pod and seed traits on 20 randomly selected trees in every population. Duncan’s multiple range test (DMRT) and the dendrogram distinctively divided the populations into two groups; Wamba and Meisori in one group while Bulla Sambul and Kutulo in the other, indicating geographical structuring of the genetic variability. The trees were larger, taller with greater pod and seed traits in the Wamba and Meisori than the Bulla Sambul and Kutulo populations. Contrarily, trees had lighter seeds, more branches with higher gum weight in the Kutulo and Bulla Sambul than the Wamba and Meisori populations. The Shannon–Weaver diversity index (H), depicted 1,000-seed weight as the most varied trait across the populations. These findings provide essential information on the genetic diversity of the species, necessary for delineation of particular zones for seed sources for germplasm conservation, selection for domestication and improved gum arabic production.  相似文献

Teak(Tectona grandis L.f.) is a popular hardwood species native to South and South-East Asia. The possible association of amplified fragment length polymorphism(AFLP) marker with morphological variables of eleven important characters viz. girth at breast height(GBH), height of tree, bole height, branch knots, presence of fluting, spiral stem, leaf hair, leaf length/breadth ratio, branching pattern, bark colour and petiole shape for nine natural populations comprising 180 genotypes of teak(9 populations × 20 trees). The phenogram constructed using Euclidean distances for the eleven morphological characters showed that the populations were not grouped according to their geographical origin. The Mantel's test for pairwise correlation between Euclidean distances of different morphological variables and genetic distances from AFLP data revealed that only petiole character(r =0.269; p =0.046) and height of tree(r =0.200; p =0.001) were significantly correlated with that of AFLP data matrix. The nine populations in this study covered a geographic area of about 1000 km stretch along the Western Ghat of South India. A test of correlation between genetic and geographic distance matrices revealed a significant positive correlation(r =0.475; p =0.009). The lack of perfect congruence between morphological and molecular data except for geographic distance, tree height and petiole character suggested that the morphological system might be useful for the morphotypes management but not appropriate to study the genetic structure of the teak populations.  相似文献
