为了筛选出对番茄青枯病具有较好防效的生防菌,采用皿内测定法从上海地区的番茄青枯病自然衰退土壤中,分离到一株对番茄青枯病有很强抑制作用的菌株SN15-2,并进行了分子鉴定和对荧光假单胞菌SN15-2产抗生素能力和定殖能力测定。结果表明,菌株SN15-2为荧光假单胞菌(Pseudomonas fluorescens)。其菌株能产生2,4-二乙酰基间苯三酚(2,4-DAPG)、硝吡咯菌素(PRN)、藤黄绿脓菌素(PLT)。同时,可以产生HCN、噬铁素,能够形成生物膜。SN15-2在施入番茄根际后的前20 d定殖数量减少,20 d后基本稳定,60 d时,在干土中的定殖数量可达3.67×105cfu/g。盆栽防效分析表明,荧光假单胞菌SN15-2对番茄青枯病防效达到46.58%。  相似文献

生防细菌L5生防相关因子的初步分析及其种类鉴定   总被引:2,自引:0,他引:2  
从扬州郊区土壤中分离到细菌菌株L5,该菌株抑菌谱广,对多种病原真菌都有明显的拮抗能力,离体叶片生物测定结果表明菌株L5对番茄灰霉病防效达72%.对该菌株的生防相关性状进行分析,结果表明菌株L5可产生硝吡咯菌素(Pyrrolnitrin)和嗜铁素等抗菌物质.经过16S rDNA序列、16S-23S rDNA间区序列同源性分析和生理生化性状的测定,将该菌株鉴定为气味沙雷氏菌(Serratia odorifera).细菌菌株L5包含2个16S-23S rDNA的间区(IGSs),其中IGS1内含tRNA Glu基因,间区类型属于IGSG;IGS2内含tRNA lle和tRNA Aia基因,间区类型属于IGS LA.  相似文献

生防菌株YB-81的鉴定及其对番茄灰霉病的防效   总被引:3,自引:0,他引:3  
为明确生防菌株YB-81对番茄灰霉病的防病作用及其分类地位,采用平板对峙及含毒介质法测定了其对番茄灰霉病菌的抑菌作用,并测定了该菌株盆栽防病效果及抑菌谱,通过形态学、生理生化特征以及分子鉴定相结合的方法鉴定了YB-81菌株。结果表明:YB-81菌株对番茄灰霉病菌抑菌作用强,抑菌带和抑制率分别为15 mm和80.5%;盆栽防效较好,发酵产物稀释10倍的预防效果达到77.5%,治疗效果不及预防效果;YB-81菌株具有广谱的抑菌活性;根据形态特征、生理生化特征以及16S rDNA序列分析结果将YB-81菌株鉴定为枯草芽胞杆菌。  相似文献

从蔬菜根际土壤中筛选到一株生防格氏沙雷菌(Serratia grimesii)CNY-04,利用抑菌圈法测定其对灰霉病菌(Botrytis cinerea Pers.ex Fr.)的抑制效果,通过抑制孢子萌发、影响菌丝正常生长试验对CNY-04的作用机制进行研究,并对CNY-04菌株相关性状及抑菌物质的理化性质进行了研究.结果表明,CNY-04菌株对灰霉病菌拮抗能力强且遗传稳定,抑菌圈直径达到34 mm; CNY-04可对灰霉病菌菌丝结构造成破坏;可显著抑制灰霉病菌分生孢子萌发,抑制率为99%;CNY-04菌株不产生硝吡咯菌素、氢氰酸及水解酶类,可产生嗜铁索和蛋白酶;抑菌物质具有较好的热稳定性和酸碱稳定性.抑菌物质由灰霉病菌诱导产生,与已报道的沙雷菌属的作用机理完全不同.  相似文献

 为探讨烟草根际生防细菌的生防机制,从重庆地区连作烟田健康烟株根际土壤中分离筛选到1株对烟草疫霉具有较强拮抗作用和对黑胫病具有良好防效的细菌菌株P-72-10。根据培养性状、形态特征、生理生化特性、基因组DNA的(G+C)mol%含量测定以及16S rDNA基因序列分析确定该菌株的分类地位。该菌株菌落乳白色,能产生水溶性荧光色素,革兰氏染色反应阴性,菌体杆状、大小(8.1~16.2)μm×(1.8~4.8)μm,单端生鞭毛,不形成芽孢。The BIOLOG GN2 结果显示该菌株属于假单胞菌属Pseudomonas。该菌株基因组DNA的(G+C)mol%含量为 60.72 mol%。16S rDNA基因序列分析显示该菌株与假单胞菌属荧光假单胞杆菌多个菌株的序列同源性达到99%,GenBank上的登录号为:HQ888871。结合其形态特征和生理生化特性,将菌株P-72-10鉴定为荧光假单胞杆菌P. fluorescens。平板检测拮抗代谢产物结果表明:该菌株具有分解纤维素、蛋白质和结合Fe 3+的能力,但不能分解几丁质。  相似文献

2015年,在广东省广州市番茄种植区发生番茄髓部坏死病,从番茄病样中分离到一种细菌。随机选取5株细菌进行致病性测定,结果显示,这5株细菌均可侵染番茄植株产生典型的髓部坏死症状,与田间病株的症状相同。5株细菌的16S rDNA序列间同源性为99.9%~100%,且与菊苣假单胞菌(Pseudomonas cichori)MAFF211996、TIs01和IP1-05菌株的16S rDNA序列同源性最高,为99%。利用菊苣假单胞菌hrcR基因的特异引物Hrp1a/Hrp2a进行PCR,从这5株细菌的DNA中均能扩增出大小约897 bp的特异片段;这些片段序列间同源性为99%~99.9%,且与P.cichori 83-1菌株hrcR基因的序列同源性为99.9%。这些结果表明,引起广东番茄髓部坏死病的病原为菊苣假单胞菌(P.cichorii)。生理生化试验显示,该病原菌与文献报道的菊苣假单胞菌的生理生化特征均相吻合,进一步证实引起广东番茄髓部坏死病的病原为菊苣假单胞菌。  相似文献

为获得对小麦全蚀病菌有良好拮抗效果的生防菌株,分别从河南省商丘市及驻马店市小麦全蚀病发生田块中采集小麦根际土样,采用稀释平板涂布法共分离到1051株细菌,通过与全蚀病菌G1037菌株进行平板对峙筛选,最终获得9株具有明显拮抗效果且生长状况良好的菌株。16S rDNA序列比对及生理生化性状分析结果表明:菌株P155、P154、P16及P147为荧光假单胞菌Pseudomonas fluorescens,菌株P188和P97为恶臭假单胞菌Pseudomonas putida,菌株LY3为产酶溶杆菌Lysobacter enzymogenes,菌株S38为嗜根寡养单胞菌Stenotrophomonas rhizophila,菌株B20为洋葱伯克霍尔德Burkholderia cepacia。产抗生素相关基因的检测结果发现,菌株P147含吩嗪和硝吡咯菌素合成基因,菌株B20含硝吡咯菌素合成基因。除B20、LY3、S38和P147外,其余菌株均可产生嗜铁素。9株细菌都产蛋白酶。除P97、B20和S38外,其余菌株均可产脂肽类物质。盆栽试验结果表明,9株生防菌对小麦全蚀病都具有较好的防治效果,菌株P155和P154的防治效果最好,相对防效分别为67.11%和63.82%,略高于3%的苯醚甲环唑种衣剂的防治效果。研究结果表明这些细菌具有作为小麦全蚀病生防菌的潜力。  相似文献

为筛选防治水稻稻瘟病的假单胞菌Pseudomonas,通过平板对峙试验从291株细菌中筛选获得对稻瘟病菌具有明显拮抗作用的菌株S149,对稻瘟病菌P131菌丝抑制率达到83.33%,发酵液和无菌上清液的抑制作用分别为67.39%和56.52%。16S rDNA序列分析显示,菌株S149与荧光假单胞菌P. fluorescens基因序列的同源性最高,结合形态观察,鉴定为荧光假单胞菌。对镰刀菌等10种植物病原菌有较明显拮抗作用,发酵液对稻瘟病菌分生孢子萌发和附着胞形成的抑制效果达到89%以上,无菌上清液对孢子萌发和附着胞形成的抑制作用达到81%以上。温室条件下对叶瘟的预防效果达到79.98%;田间试验结果证实对水稻幼苗有明显的促生效果,提高种子发芽7.15%、促进根长10.36%和增加株高10.33%;对叶瘟和穗颈瘟的田间防效分别达到73.32%和70.21%,与75%三环唑可湿性粉剂没有显著差异,与绿地康有显著差异。研究表明,荧光假单胞菌S149不仅可以用于防治水稻稻瘟病,而且可以促进水稻生长,具有较好的防治稻瘟病等多种真菌病害的应用潜力。  相似文献

荧光假单胞菌拮抗菌株对烟草疫霉的抑菌机制及控病效果   总被引:2,自引:1,他引:1  
为探讨烟草根际生防细菌对烟草疫霉Phytophthora nicotianae的抑菌机制,从重庆地区连作烟田健康烟株根际土壤分离获得5株荧光假单胞菌Pseudomonas fluorescens拮抗菌株。通过平板对峙及代谢产物抑菌试验筛选对烟草疫霉具有高效拮抗作用的菌株,其中,P-72-10菌株抑菌效果最强,抑菌带半径达13.0 mm,相对抑制率为68.57%,且该菌株代谢产物对烟草疫霉菌丝生长有明显的抑制作用,相对抑制率达25.39%~46.03%;显微观察发现该菌株可引起烟草疫霉菌丝的分支增多,菌丝顶端膨大呈畸形,多数菌丝中间或顶端细胞的细胞壁加厚、原生质浓缩和产生类似厚壁孢子的细胞。在温室盆栽条件下P-72-10菌株对烟草黑胫病也表现出良好的控病效果,对抗病和感病品种的相对防效分别为53.57%和66.37%。  相似文献

解淀粉芽胞杆菌YU-1对水稻纹枯病的防治作用   总被引:1,自引:0,他引:1  
本文旨在筛选对水稻纹枯病有生防作用的菌株,并初步探索其生防作用机理。收集水稻、甘蓝、黄瓜等不同植物根际土壤,采用稀释分离和对峙培养法筛选对水稻纹枯病菌Rhizoctonia solani Kühn有抑菌作用的菌株;通过离体接种防效、盆栽防效、抑菌谱、对水稻纹枯病菌菌核萌发及形成的抑制作用等方面评价其生防潜力,并对生防菌株进行鉴定。结果表明,从采集的37份根际土壤中共分离获得细菌297株,其中4株对纹枯病菌具有较好的拮抗作用,菌株YU-1对水稻纹枯病菌菌丝生长的抑制率达89.8%;对西瓜枯萎病菌、草莓灰霉病菌、番茄早疫病菌、油菜菌核病菌、小麦赤霉病菌菌丝生长的抑制率均在85%以上;对水稻纹枯病的离体和盆栽防效分别为73.1%和66.3%;对水稻纹枯病菌菌核萌发的抑菌率在92%以上;经生理生化和分子鉴定为解淀粉芽胞杆菌Bacillus amyloliquefaciens。由此可看出,菌株YU-1对水稻纹枯病的防治具有较强的应用价值,具有进一步开发成生物农药的潜力。  相似文献

Pretreatment of detached tomato leaves with o -hydroxyethylorutin reduced the percentage leaf area affected, and delayed the appearance of necrosis, following inoculation with conidial suspensions in droplets of the grey mould fungus Botrytis cinerea . o -Hydroxyethylorutin delayed, but did not inhibit, in vitro germination of conidia, although overall percentage germination was reduced compared with water controls. Both the reactive oxygen species (ROS) – superoxide anions and hydrogen peroxide – increased twice as much in o -hydroxyethylorutin-treated leaf tissue 2 and 6 h postinoculation with B. cinerea conidia in tissues under inoculation drops, as well as in surrounding tissues, whereas in plants not pretreated with the compound ROS generation was noticed later, and only in tissues under inoculation drops. Compared with these compounds, changes in the levels of hydroxyl radicals, lipid peroxidation and the activity of the enzymes superoxide dismutase, guaiacol peroxidase, ascorbate peroxidase and catalase were largely unchanged. In stimulating ROS in inoculated tomato tissue, B. cinerea appeared to be affected directly pre- and postinfection, but indirect effects increasing host resistance cannot be excluded.  相似文献

Several film-forming polymers reduced the amount of grey mould on various crops in a dew chamber and in a plastic house under natural conditions. The polymers Wilt Pruf, Biofilm and Colfix reduced germination of conidia and germ tube length of Botrytis cinerea . The most effective inhibition of linear growth of the pathogen on potato dextrose agar was obtained by Safe Pack and Biofilm. All polymers significantly reduced grey mould on detached leaves of Capsicum, Phaseolus , tomato, cucumber, rose and pelargonium. Grey mould on rose flowers was not controlled, apparently due to latent infection. The substances Biofilm and Vapor Gard were applied either alone or with chlorothalonil fungicide on cucumber plants in a commercial greenhouse. The polymers had no harmful effect to the host. Disease on senescing female fruits of cucumber was reduced by 46–67% with no additive effect to the mixture with fungicides. Stem infection also was reduced.  相似文献

从安徽省寿县农田土壤中分离筛选到1株对番茄灰霉病菌Botrytis cinerea有强拮抗作用的放线菌菌株HNU-1。根据菌体形态特征、培养特征、细胞壁组分、生理生化特性和16S rRNA基因序列分析,将该菌株鉴定为吸水链霉菌Streptomyces hygroscopicus。菌株HNU-1发酵滤液可抑制灰霉病菌菌丝生长和分生孢子萌发,且浓度越高,抑制能力越强;当发酵滤液稀释6.67倍时则完全抑制灰霉病菌菌丝生长和分生孢子萌发。盆栽试验结果表明,菌株HNU-1发酵滤液6.67倍稀释液对番茄灰霉病的预防与治疗效果分别为87.8%和77.9%,均显著高于50%多菌灵可湿性粉剂500倍稀释液。  相似文献

 番茄灰霉病菌(Botrytes cinerea)的分生孢子萌发需要高湿的环境,一般要求相对湿度在80%以上或水滴中萌发最好。在适温下,随着湿度的增加,病菌致病性增强,孢子在相对湿度85%以上、菌丝在相对湿度80%以上的高湿环境能够发生侵染,并形成危害性的软腐斑。在一天中高湿的时间直接影响番茄灰霉病的发生,高湿(相对湿度超过85%)时间8h以上病菌才能够连续侵染。  相似文献

离体试验表明,醚菌酯对番茄灰霉病菌菌丝生长、孢子萌发、孢子产量和菌核产量均有一定的抑制作用,其EC50分别为935.670、0.667、0.479和6.975mg/L。其中对菌丝生长的抑制作用较低。但是,用10mg/L和20mg/L的醚菌酯叶片喷雾处理前24h,接种灰霉病菌菌碟,醚菌酯对灰霉病的保护效果分别达到92.3%和100%;处理后24h接种,治疗效果分别达83.3%和100%,说明醚菌酯对番茄灰霉病具有很好的保护和治疗作用。醚菌酯具有优异的内吸活性,能在植物体内向顶传导。  相似文献

Eighteen free radical scavengers (antioxidants) were tested for their ability to control grey mould. Most of the compounds reduced disease significantly in at least one of the test hosts–leaves of tomato, pepper, Senecio sp., bean, eggplant, or rose flowers; however, the effective concentration varied between 0.1 and 100 m m . Selected antioxidants were tested further. Butylated hydroxy toluene (BHT), tannic acid, ascorbic acid and dimethyl sulfoxide (DMSO) at a concentration of 1 .0 m m controlled grey mould of tomato fruits. All these compounds except BHT controlled the disease on cucumber fruits. Antioxidants affected Rhizopus stolonifer on grape berries but not Botrytis cinerea or Aspergillus spp. Some combinations of antioxidants were found to be more effective than either compound alone when tested on pepper or tomato. The synergists ascorbic acid and citric acid improved the control activity of BHT, propyl gallate, benzoic acid and tert -butylhydroquinone on tomato leaves. Ethylene production was inhibited in tomato leaves treated with propyl gallate, ascorbic acid and benzoic acid, but not in pepper leaves. Ethephon or H2O2 increased the severity of grey mould on leaves of Senecio sp. Their effect was controlled by BHT and benzoic acid or by BHT, respectively. Four to six compounds reduced linear growth of B. cinerea isolates in culture at a concentration of 1.0 m m , and six more compounds were effective at 10.0 m m . However just five compounds inhibited conidial germination at the high concentration alone. Gluconic acid lactone, thiourea and propyl gallate reduced Sclerotinia sclerotiomm on lettuce by 51-76%. The multiple activity of antioxidants on the host plant interaction is discussed.  相似文献

采用菌丝生长速率法和孢子萌发法,分别测定了烟草灰霉病菌对多菌灵、嘧霉胺、异菌脲和丙环唑的敏感性,同时通过离体叶片法评估了这4种杀菌剂对烟草灰霉病的保护和治疗作用。结果表明:4种杀菌剂对烟草灰霉病菌的菌丝生长和孢子萌发均表现出了不同程度的抑制活性,并对灰霉病同时具有保护和治疗作用。其中多菌灵对菌丝生长的抑制活性最强,EC50平均值为0.06 mg/L,其次为丙环唑、嘧霉胺和异菌脲,EC50平均值分别为0.36、0.53和0.60 mg/L;异菌脲和丙环唑对烟草灰霉病菌孢子萌发的抑制活性较强,EC50平均值分别为2.05和2.21 mg/L,其次为嘧霉胺和多菌灵,EC50平均值分别为10.56和131.23 mg/L。异菌脲和多菌灵对灰霉病的保护作用和治疗作用均最强,当药剂质量浓度为200 mg/L时,其对离体叶片的保护和治疗作用防效分别为100%、100%和98.3%、91.8%。研究结果可为烟草灰霉病的科学防治提供依据。  相似文献

Biocontrol of Botrytis cinerea with Trichoderma spp. is generally believed to result from direct interaction of the biocontrol agent with the pathogen or from a Trichoderma-induced change in environmental conditions that affects B. cinerea development. In this work we provide arguments for the participation of induced plant defence in T. harzianum T39 control of B. cinerea. In tomato, lettuce, pepper, bean and tobacco, T. harzianum T39 application at sites spatially separated from the B. cinerea inoculation resulted in a 25–100%percnt; reduction of grey mould symptoms, caused by a delay or suppression of spreading lesion formation. Given the spatial separation of both micro-organisms, this effect was attributed to the induction of systemic resistance by T. harzianum T39. The observation that in bean the effect of T. harzianum T39 was similar to that of the rhizobacterium Pseudomonas aeruginosa KMPCH, a reference strain for the induction of systemic resistance, confirmed this hypothesis. Since B. cinerea control on tobacco leaves sprayed with T. harzianum T39 was similar to the control on leaves from T. harzianum T39 soil-treated plants, induction of plant defence might also participate in biocontrol on treated leaves.  相似文献

冬青卫矛内生放线菌YDG17菌株发酵液抑菌活性研究   总被引:1,自引:1,他引:0  
从冬青卫矛植株中筛选得到一株内生链霉菌YDG17,其发酵液对多种植物病原真菌有较强的抑制作用。室内生物测定结果表明,YDG17菌株发酵液对供试的11种植物病原真菌菌丝生长均有一定的抑制作用,其中对小麦赤霉病菌Fusarium graminearum、番茄灰霉病菌Fulvia fulvum和番茄早疫病菌Alternaria solani的抑制作用最强,EC50值分别为259.98、336.13和100.72 mg/L;对供试的3种植物病原真菌孢子萌发也均有一定的抑制作用,其中对番茄灰霉病菌孢子萌发的抑制作用最强,EC50值为87.84 mg/L。离体子叶法测定结果表明,发酵液原液对番茄灰霉病的保护效果为97.62%,治疗效果为79.63%。盆栽试验结果表明,发酵液原液对番茄灰霉病的保护效果为71.34%,治疗效果为64.23%。捷克八溶剂系统纸色谱测定结果表明,该发酵液的主要活性物质为碱性水溶性抗生素。选用弱酸性离子交换树脂吸附法对YDG17发酵液活性成分进行了初步分离,并对其活性馏分进行ESI-MS/MS分析,表明其活性物质主要为链丝菌素类化合物,该类化合物为首次从植物内生菌中分离得到。  相似文献
