The current review aims to establish insulin-like growth factor-1 (IGF-I) as the factor that signals nutritional status to the reproductive axis, and show that assessment of IGF-I in blood early postpartum during the negative energy balance (NEB) period could be used to predict both nutritional and reproductive status in dairy cattle. The review also explores the effect of nutritional status on circulating IGF-I concentrations and the endocrine role of IGF-I on the reproductive axis. IGF-I plays an important role in gonadotropin-induced folliculogenesis, ovarian steroidogenesis and corpus luteum (CL) function. It also modulates pituitary and hypothalamus function. IGF-I clearly has an endocrine role on the reproductive axis. Severe under nutrition significantly reduces plasma IGF-I concentrations. During the critical period of NEB in high yielding dairy cattle early postpartum, IGF-I concentrations are low in blood and its levels are positively correlated to energy status and reproductive function during this period. Changes in circulating IGF-I immediately postpartum may help predict both nutritional and reproductive status in dairy cattle. IGF-I is therefore one of the long sought factors that signal nutritional status to the reproductive axis.  相似文献

To establish sustainability in the dairy industry, it is important that cows become pregnant at a biologically optimal time and at an economically profitable interval after calving. In this review, the results obtained from Holstein cattle in an experimental herd for dairy research are summarized. First, the effect of age at first calving of heifers on productive and reproductive performance was examined. A reduction in calving age from 25.1 to 21.5 months with the same growth rate during the first 12 months after birth had no negative effects on the heifers' performance. Second, the postpartum follicular dynamics of lactating cows were traced in relation to their fertility, and the emergence and fate of cystic ovarian follicles were examined. The premature initiation of ovarian activity does not always improve the fertility of cows as indicated by the number of days open. Third, the occurrences of anestrous ovulation during the early postpartum period were analyzed with reference to the frequency of reversion to anestrus. The premature onset of estrous activity also did not improve fertility, and relapse back into anestrus after the onset of the estrous cycle often occurred during the breeding period. Fourth, some indices for the occurrence of postpartum reproductive events were evaluated as an indicator of the reproductive performance of lactating cows. The milk yield and percentage of body weight loss could be indicators for reproductive events. Finally, the potency of a pedometry system for the detection of typical and atypical estrous behaviors of heifers and lactating cows was evaluated in terms of efficiency and accuracy. The location of the pedometers and housing conditions for the animals affected the estrus detection of the system. These results represent the reproductive potential of modern high-yielding dairy cattle and provide a baseline to evaluate their reproduction.  相似文献

Using reproduction parameters as indicators for cattle welfare has limitations and, at best, these parameters should only be viewed as indirect indicators of welfare. On a farm level, measures such as fertility rates emphasize biological performance of the herd but fail to consider the welfare of individual animals. Even on an individual level, the relationship between reproductive effectiveness and animal welfare is complex. Good reproductive performance does not automatically signify good welfare, as domestication and targeted breeding programmes have led to prioritization of high productive and reproductive performance in most modern farm animal species. In this review, we synthesize literature regarding cattle husbandry, reproduction, welfare and their multidimensional relationships. We argue that practices such as artificial insemination or the use of sexed semen may provide potential welfare advantages as these practices reduce the risk of disease transmission and injury or enable selection of specific beneficial traits. Furthermore, they may offer a solution to current practices jeopardizing welfare, such as the management of surplus bull calves in the dairy industry. Conversely, the animals’ ability to perform natural behaviours such as oestrous expression, an aspect arguably contributing to welfare, is often limited on commercial farms; this limitation is particularly evident in housing systems such as tie stalls where movement is restricted. Moreover, common management practices such as oestrus manipulation may lead to negative attitudes in citizens who often regard naturalness as important element of animal welfare.  相似文献

It is generally agreed that stress can impair reproduction. Furthermore, it is often thought that cortisol, which is secreted during stress as a result of activation of the hypothalamo-pituitary adrenal axis, is associated with this stress-induced impairment of reproduction. It has been hypothesized that reproduction in females is particularly susceptible to disruption by acute stress during the series of endocrine events that induce estrus and ovulation. Nevertheless, we found no support for this conjecture when we subjected female pigs to repeated acute stress or repeated acute elevation of cortisol during the period leading up to estrus and ovulation. Conversely, studies have demonstrated that prolonged stress and sustained elevation of cortisol can disrupt reproductive processes in female pigs. Nevertheless, in each study that demonstrated this effect, there were some animals subjected to the prolonged stressor or the sustained elevation of cortisol in which the reproductive parameters that were measured were not affected by the treatment. We propose that reproduction in female pigs is resistant to the effects of acute or repeated acute stress or acute or repeated acute elevation of cortisol even if these occur during the series of endocrine events that induce estrus and ovulation. Furthermore, while reproductive processes in some individuals are compromised, reproduction in a proportion of female pigs appears to be resistant to the effects of prolonged stress or sustained elevation of cortisol.  相似文献

The significant decrease in fertility observed in dairy cattle during the last few decades and increasing requirements by the farmers have made a regular control of reproduction indispensable to urgently identify and solve potential problems affecting reproductive efficiency. Traditionally, the main diagnostic methods used for reproductive control in cattle included rectal palpation, inspection of vaginal discharge and vaginoscopy. Since the 1990 s, the use of ultrasound (US) has become a common diagnostic method as a result of the new advances made in the development of US scans: smaller size, high level of autonomy, high image quality and accessible prices. Ultrasound improves accuracy in the diagnoses of stages of the oestrous cycle, ovarian and uterine pathologies, and pregnancy diagnosis. In addition, it facilitates the diagnosis of alterations during pregnancy (embryo mortality, foetal malformations, etc.) and helps determining foetal sex from day 55 of pregnancy.  相似文献

Kisspeptins, a family of neuropeptide encoded by the Kiss1 gene, have emerged as crucial regulator of fertility and reproduction by regulating the hypothalamic–pituitary–gonadal axis. The present study was aimed to identify and associate SNPs in the KISS1 gene with reproductive traits in cattle of Indian origin. DNA samples collected from 300 individual cows of three Indian dairy breeds (Gir, Kankrej and Frieswal) of cattle were used in the study. The SNPs of KISS1 gene were identified with PCR-RFLP and sequence analysis using two sets of primer pairs. A total of 5 SNPs were identified in the targeted region of which, two were selected for screening the population and association studies. The analysis revealed that genotypes of rs442633552G>A and rs42022871C>T had a significant association with dry period. The SNP rs42022871C>T also established significant role in milk production traits, and selection of TT-genotyped animals will improve the reproduction and production potential of the animals.  相似文献

Mutations within the growth hormone receptor (GHR) gene that lead to an inactivated or truncated GHR protein cause abnormal growth and small adult size in a variety of species (Laron dwarfism). We studied a line of miniature Bos indicus cattle that have phenotypic (small mature size) and endocrine (increased blood growth hormone and decreased blood insulin-like growth factor-I concentrations) similarities to Laron dwarfs. Liver mRNA from miniature and control cattle was used to amplify a cDNA within the coding region of the GHR. The miniature cattle had GHR mRNA size (determined by Northern blot) and cDNA sequence that were similar to control cattle and, therefore, were unlike most Laron dwarf genotypes in which the GHR gene is mutated. Amounts of mRNA from liver as well as muscle (superficial neck and longissimus) were analyzed by ribonuclease protection assay for IGF-I, total GHR, GHR 1A (inducible, liver-specific GHR mRNA), and GHR 1B (constitutive GHR mRNA). Four control and five miniature bulls were tested. As expected, liver IGF-I mRNA was decreased in the miniature cattle (approximately 12% of control; P < 0.01). The amount of the total GHR as well as GHR 1A mRNA were also decreased in liver (17% and 19% of control, respectively; P < 0.01). Other GHR mRNA, including GHR 1B mRNA, were similar for miniature and control cattle. In muscle, there was a tendency (P < 0.10) for decreased IGF-I mRNA and increased GHR mRNA in miniature compared with control cattle. In summary, a novel phenotype for Laron dwarfism in Bos indicus cattle was associated with underexpression of GHR 1A mRNA, but not other GHR mRNA variants in liver. In addition to decreased GHR 1A mRNA, the miniature cattle had decreased liver IGF-I mRNA. Full expression of GHR 1A in liver, therefore, may be required for full liver IGF-I expression and normal growth.  相似文献

The management of bovine reproduction is the cornerstone of health provision in elite herds. Aims and objectives for reproductive performance should be herd specific and data to monitor progress should not only be frequently collected, but also analysed and reported. Strategic monitoring of animals should include a vaginal examination for evidence of uterine disease, as well as transrectal ultrasonography of the genital tract. There has been considerable advancement in our ability to intervene in the reproduction of cattle during the last 50 years. However, it is salutary to note that during this time fertility has consistently declined, despite increasing veterinary intervention. Most elite herds use artificial insemination and success depends on accurate detection of oestrus expression, but this appears to be less overt than 25 years ago. In addition, half the cattle have abnormal oestrous cycles after parturition and conception rates are decreasing by 1% per year. Risk factors for abnormal oestrous cycles include puerperal problems, negative energy balance, which can be evaluated by body condition scoring, and uterine disease. Bacterial contamination of the uterus is ubiquitous after parturition in cattle and disease disrupts ovarian follicle growth and function. Reproduction is also disrupted by stress associated with clinical disease, pain or a sub-optimal environment. The challenge for veterinarians providing reproduction control programmes to elite herds is to transfer our knowledge of the problems underlying subfertility to the farm, in order to provide effective solutions.  相似文献

Non-additive genetic effects are usually ignored in animal breeding programs due to data structure (e.g., incomplete pedigree), computational limitations and over-parameterization of the models. However, non-additive genetic effects may play an important role in the expression of complex traits in livestock species, such as fertility and reproduction traits. In this study, components of genetic variance for additive and non-additive genetic effects were estimated for a variety of fertility and reproduction traits in Holstein cattle using pedigree and genomic relationship matrices. Four linear models were used: (a) an additive genetic model; (b) a model including both additive and epistatic (additive by additive) genetic effects; (c) a model including both additive and dominance effects; and (d) a full model including additive, epistatic and dominance genetic effects. Nine fertility and reproduction traits were analysed, and models were run separately for heifers (N = 5,825) and cows (N = 6,090). For some traits, a larger proportion of phenotypic variance was explained by non-additive genetic effects compared with additive effects, indicating that epistasis, dominance or a combination thereof is of great importance. Epistatic genetic effects contributed more to the total phenotypic variance than dominance genetic effects. Although these models varied considerably in the partitioning of the components of genetic variance, the models including a non-additive genetic effect did not show a clear advantage over the additive model based on the Akaike information criterion. The partitioning of variance components resulted in a re-ranking of cows based solely on the cows’ additive genetic effects between models, indicating that adjusting for non-additive genetic effects could affect selection decisions made in dairy cattle breeding programs. These results suggest that non-additive genetic effects play an important role in some fertility and reproduction traits in Holstein cattle.  相似文献

The welfare of cow along with profitability in production are important issues in sustainable animal breeding programmes. Along with an intense/intensive selection for increased milk yield, reproductive performance has declined in many countries, in part due to an unfavourable genetic relationship. The largely unchanged genetic trend in female fertility and calving traits for Scandinavian Red breeds shows that it is possible to avoid deterioration in these traits if they are properly considered in the breeding programme. Today's breeding is international with a global selection and extensive use of the best bulls. The Nordic countries have traditionally recorded and performed genetic evaluation for a broad range of functional traits including reproduction. In recent years many other countries have also implemented genetic evaluation for these traits. Thus, the relative emphasis of dairy cattle breeding objectives has gradually shifted from production to functional traits such as reproduction. Improved ways of recording traits, e.g. physiological measures, early indicator traits, assisted reproductive techniques and increased knowledge of genes and their regulation may improve the genetic selection strategies and have large impact on present and future genetic evaluation programmes. Extensive data bases with phenotypic recordings of traits for individuals and their pedigree are a prerequisite. Quantitative trait loci have been associated to the reproductive complex. Most important traits, including reproduction traits are regulated by a multitude of genes and environmental factors in a complex relationship, however. Genomic selection might therefore be important in future breeding programmes. Information on single nucleotide polymorphism has already been introduced in the selection programmes of some countries.  相似文献

The overall objective of one of the major research programs in the Co-operative Research Centre (CRC) for Beef Genetic Technologies is to 'Improve female reproductive performance' in tropical, northern Australian beef cattle herds. To address this overall objective, a quantitative genetics project focused on investigation of male reproductive traits was designed and linked to three female reproduction-focussed projects, (i) discovery of genes associated with post-partum re-conception and age at puberty; (ii) expression of genes associated with post-partum re-conception; and (iii) early predictors of lifetime female reproductive performance. During the initial planning of this male reproductive traits project, the CRC Scientific Review Committee recommended that the research team investigate and evaluate potentially new, early-life (i.e able to be measured before 2 years of age) predictors of both male and female reproductive performance. To address this recommendation, the following was carried out: (i) criteria for selection of traditional and candidate traits were established; (ii) methodology for tabulation of potential traits/phenotypes that define male and female reproductive function was developed; and (iii) a systematic scientific review of early-life predictors of male and female fertility was prepared. This review concluded that although factors that might be useful in predicting male reproductive performance have been studied for many years, there was relatively little useful information available to meet the objectives of this review. It was also concluded that the direction of future research should be guided not only by previous research which was scarce, but also by speculative hypotheses arising from an understanding of the physiological, endocrinological and genetic processes active in reproduction. A small number of new traits were recommended in addition to traditional sperm morphology, sexual behaviour, anatomical structure and growth traits. Potential additional traits include measurement of gonadotrophin-releasing hormone-stimulated luteinizing hormone (GnRH-stimulated LH); inhibin; several seminal plasma proteins (osteopontin, spermadhesin and seminal plasma proteins BSP30 and phospholipase A(2) could be used in an index); 11β-hydroxysteriod dehydrogenase; and leptin. In addition, the potential also exists to screen animals for a number of genetic markers associated with age of puberty, follicular recruitment and ovulation rate and genes associated with bovine seminal plasma protein and testosterone production. Insulin-like growth factor-1 (IGF-1) measurements are included because of their association with growth parameters, and an additional analysis demonstrated associations with male and female reproductive traits. Some of these factors have been previously evaluated in small numbers of animals of various species under intensive management conditions. Therefore, there is a need to evaluate these factors in much larger numbers of beef cattle grazing semi-extensive tropical production systems in northern Australia to determine their value in improving beef cattle enterprise profitability through improved herd fertility.  相似文献

近年来,中国肉牛产业不断调整优化,母牛存栏量持续增长,但国内可繁殖肉用母牛数量持续减少,母牛受胎率普遍不高,犊牛成活率低,已成为制约肉牛产业发展的瓶颈环节。同期排卵-定时输精技术通过应用生殖激素处理调控母牛发情周期,促进母牛发情,从而提高母牛参配率,在提高母牛繁殖力中得到广泛应用,是当今家畜繁殖技术的一个重大突破。由于中国肉牛养殖集约化程度低,同期排卵-定时输精技术在肉牛上的应用尚未得到广泛重视。不同同期排卵-定时输精处理程序各有特点,在实际应用中影响同期排卵-定时输精程序处理母牛发情配种妊娠率的因素很多,如何对该技术实现进一步优化是当前面临的重大难题。国内外针对于同一生殖激素的不同浓度、不同激素组合及激素注射的间隔时间、激素的替换等做了大量的研究。文章综述了定时输精技术原理、主要程序及国内外肉牛同期排卵-定时输精技术研究进展,以期为建立高效的肉牛同期排卵-定时输精技术体系实现该技术在肉牛产业上的应用提供参考。  相似文献

Background

The present study hypothesized that GH-AluI and IGF-I-SnabI polymorphisms do change the metabolic/endocrine profiles in Holstein cows during the transition period, which in turn are associated with productive and reproductive parameters.

Methods

Holstein cows (Farm 1, primiparous cows, n = 110, and Farm 2, multiparous cows, n = 76) under grazing conditions were selected and GH and IGF-I genotypes were determined. Blood samples for metabolic/endocrine determinations were taken during the transition period and early lactation in both farms. Data was analyzed by farm using a repeated measures analyses including GH and IGF-I genotypes, days and interactions as fixed effects, sire and cow as random effects and calving date as covariate.

Results and Discussion

Frequencies of GH and IGF-I alleles were L:0.84, V:0.16 and A:0.60, B:0.40, respectively. The GH genotype was not associated with productive or reproductive variables, but interaction with days affected FCM yield in multiparous (farm 2) cows (LL yielded more than LV cows) in early lactation. The GH genotype affected NEFA and IGF-I concentrations in farm 1 (LV had higher NEFA and lower IGF-I than LL cows) suggesting a better energy status of LL cows.There was no effect of IGF-I genotype on productive variables, but a trend was found for FCM in farm 2 (AB cows yielded more than AA cows). IGF-I genotype affected calving first service interval in farm 1, and the interaction with days tended to affect FCM yield (AB cows had a shorter interval and yielded more FCM than BB cows). IGF-I genotype affected BHB, NEFA, and insulin concentrations in farm 1: primiparous BB cows had lower NEFA and BHB and higher insulin concentrations. In farm 2, there was no effect of IGF-I genotype, but there was an interaction with days on IGF-I concentration, suggesting a greater uncoupling somatropic axis in AB and BB than AA cows, being in accordance with greater FCM yield in AB cows.

Conclusion

The GH and IGF-I genotypes had no substantial effect on productive parameters, although IGF-I genotype affected calving-first service interval in primiparous cows. Besides, these genotypes may modify the endocrine/metabolic profiles of the transition dairy cow under grazing conditions.  相似文献

The effect of both environmental and management related stressors on fertility is discussed. While environmental heat as a stressor is significant in disrupting normal reproductive cyclicity, management induced stress is becoming more important when related to the requirements of modern production methods. Deviations in hormonal patterns are noted, and the clinical manifestations brought about by these changes are described. Since reproduction is the ultimate measure of an animal's ability to adapt to an ever changing external milieu, as well as forming the basis of life-time productivity, research should be aimed at obtaining greater clarity of the hormonal interactions involved. The role of neurotransmitters in these physiological mechanisms should not be overlooked. Psychobiological studies must be extended so as to provide a positive input into management procedures that should be used in intensive production units for optimal fertility and productivity.  相似文献

Background: Beef cattle breeding programs in Brazil have placed greater emphasis on the genomic study of reproductive traits of males and females due to their economic importance. In this study, genome-wide associations were assessed for scrotal circumference at 210 d of age, scrotal circumference at 420 d of age, age at first calving, and age at second calving, in Canchim beef cattle. Data quality control was conducted resulting in 672,778 SNPs and 392 animals.Results: Associated SNPs were observed for scrotal circumference at 420 d of age(435 SNPs), followed by scrotal circumference at 210 d of age(12 SNPs), age at first calving(six SNPs), and age at second calving(four SNPs). We investigated whether significant SNPs were within genic or surrounding regions. Biological processes of genes were associated with immune system, multicellular organismal process, response to stimulus, apoptotic process, cellular component organization or biogenesis, biological adhesion, and reproduction.Conclusions: Few associations were observed for scrotal circumference at 210 d of age, age at first calving, and age at second calving, reinforcing their polygenic inheritance and the complexity of understanding the genetic architecture of reproductive traits. Finding many associations for scrotal circumference at 420 d of age in various regions of the Canchim genome also reveals the difficulty of targeting specific candidate genes that could act on fertility; nonetheless,the high linkage disequilibrium between loci herein estimated could aid to overcome this issue. Therefore, al relevant information about genomic regions influencing reproductive traits may contribute to target candidate genes for further investigation of causal mutations and aid in future genomic studies in Canchim cattle to improve the breeding program.  相似文献
