为了探究贵州矮马的腹泻病原,试验以腹泻粪样为材料进行病原菌的分离鉴定。经SS和XLD培养基筛选,得到1株菌株S6,经革兰氏染色、生理生化检验以及16S rRNA基因序列比对分析,S6菌株可在SS和XLD琼脂培养基表面生长,革兰氏染色呈阴性;生理生化检验结果与沙门氏菌的生化特性相符;16S rRNA基因序列与已知的鼠伤寒沙门氏菌ATCC 13311(登录号:NR_119108)的亲缘关系最近,序列同源性为99%。形态学和分子系统学分析表明,该菌株为鼠伤寒沙门氏菌。采用PCR方法从S6菌株基因组中得到侵袭蛋白A(invasion protein A,invA)基因,与已知基因序列有6~8 bp不一致,编码的氨基酸发生了一个替换。通过改良寇氏法测定S6菌株对小鼠的半数致死量(LD50)为4.71×102 CFU,属于强致病力菌株。推测S6 invA基因的变异可能与菌株的毒力强弱有关。贵州矮马群体中沙门氏菌的检出率为57%。本研究提示贵州矮马的腹泻病原之一可能是强毒力的鼠伤寒沙门氏菌。  相似文献

猪霍乱沙门氏菌的快速分离鉴定   总被引:3,自引:0,他引:3  
猪霍乱沙门氏菌是引起仔猪副伤寒的主要病原菌,给养猪业造成重大危害。本文旨在建立一套细菌快速分离鉴定方法,为该病流行病学调查提供有效手段。从临床初诊为仔猪副伤寒的病猪采集粪便,接种亚硒酸盐亮绿增菌液增菌后,划线于SS琼脂培养基,挑无色透明菌落纯化。纯化菌落用美国B IOLOG自动细菌鉴定仪GN2肠杆菌鉴定板鉴定,同时用常规生化鉴定管鉴定,结果符合猪霍乱沙门氏菌的生化特征。PCR扩增invA基因,测序鉴定PCR产物,结果与猪霍乱沙门氏菌参考序列同源性达99%以上。分离菌株接种小白鼠,证明其对小白鼠有致病性。用沙门氏菌属标准血清检测证实其抗原式为6,7∶c∶1,5,符合猪霍乱沙门氏菌的抗原特征。应用以上方法从全国各地病料中分离鉴定了40多株猪霍乱沙门氏菌,为猪霍乱沙门氏菌病的流行病学调查提供了依据,同时证明该方法切实可行。  相似文献

建立了扩增invA基因检测沙门氏菌的PCR方法。对收集的50个血清型123株沙门氏菌及7种27株非沙门氏菌进行PCR,2%琼脂糖电泳检查,结果所有沙门氏菌都扩增出了300bp的特异性产物,非沙门氏菌都未扩增出此目的条带。产物的特异性由slot blot杂交进一步证实。通过电泳判定结果,该法可检出扩增体系中10pg染色体DNA及10~2cfu的纽波特沙门氏菌50029。为下步克隆而设计的两个酶切点(Bam HI,Eco RI)对引物的特异性没有影响。本研究为沙门氏菌的检测提供了简洁、敏感、特异的新方法,同时为克隆invA基因做属特异性探针打下了基础。  相似文献

通过16S rDNA全序列的DNA同源性分析以及PCR-RFLP分析,对从山东某奶牛场送检的腹泻死亡的犊牛内脏中分离到的一株沙门氏菌进行了血清型鉴定,得到其系统发育树状图。系统发育树分析表明,该沙门氏菌菌株与GENEBANK上发表的27株沙门氏菌的16S rDNA同源性较高,为97~99%,该沙门氏菌菌株与鼠伤寒沙门氏菌LT2(Salmonellatyphimurium LT2)的同源性最高,达到了99.7%,表明该沙门氏菌菌株为鼠伤寒沙门氏菌。  相似文献

Virulence of three Canadian poultry strains of Salmonella enteritidis, namely phagetypes (PT) 4, 8 and 13, and one Salmonella heidelberg strain was assessed in orally and intraperitoneally inoculated one-day old chickens and compared to the virulence of a human S. enteritidis PT 4 strain from the United Kingdom (UK). The two PT 4 strains were also compared in orally inoculated adult laying hens. In addition, orally inoculated Balb/c mice were used to evaluate virulence of the above strains and two strains of Salmonella typhimurium containing different plasmids. In orally inoculated one-day old chickens, the UK S. enteritidis PT 4 strain was more virulent than the Canadian PT 4 strain. The UK PT 4 strain was also more virulent and invasive in adult laying hens than the Canadian PT 4 strain. The S. enteritidis PT 8 strain and one S. typhimurium strain isolated from a chicken hatchery were the most virulent for orally inoculated Balb/c mice. This strain of S. typhimurium contained the 60 megadalton plasmid associated with virulence for Balb/c mice which was not present in the S. typhimurium strain isolated from a pig with septicemic disease.  相似文献

旨在确定河北昌黎县某花鸟鱼店宠物草龟发病死亡的病因,笔者从发病草龟体内分离纯化出1株优势菌CLW-1,通过解剖观察、细菌分离培养、革兰染色镜检、生化鉴定和16S rDNA基因测序对其分析鉴定,并通过药物敏感性测试检测菌株的耐药性,通过斑马鱼接种试验和毒力基因检测其致病性。结果显示,该菌在血琼脂平板形成湿润、表面光滑、呈β溶血的灰白色菌落,在RS琼脂培养基上长出光滑湿润的黄色菌落;革兰染色镜检为阴性短小球杆菌;16S rDNA基因序列同源性分析表明,分离菌株CLW-1与维氏气单胞菌为同一族,相似性均达99%以上;斑马鱼接种试验结果显示,该菌具有较高的致病力,其LD50为1.26×106CFU;毒力基因扩增结果表明,分离菌株CLW-1具有aerAExuLipSer毒力基因;药敏试验结果显示,分离菌株CLW-1对哌拉西林、头孢唑林和恩诺沙星等敏感;对呋喃唑酮、苯唑西林和红霉素等中敏;对麦迪霉素、克林霉素和万古霉素等耐药。通过试验结果确定此分离菌株CLW-1为维氏气单胞菌,携带毒力基因,具有较高致病力,应用对应的抗生素治疗效果较好,为维氏气单胞菌病的防治奠定基础。  相似文献

To determine the cause of death of Chinemys reevesii in a pet shop in Changli county, Hebei province, a dominant strain CLW-1 was isolated and purified from the diseased Chinemys reevesii. It was analyzed and identified through anatomical observation, bacterial isolation and culture, Gram staining and microscopic examination, biochemical identification and 16S rDNA gene sequencing, the drug resistance of the strain was detected through drug sensitivity test, and its pathogenicity was detected through zebrafish inoculation test and virulence gene experiment. The results showed that the bacteria formed moist, smooth surface, gray colony with β hemolysis on blood agar medium, and grew smooth and moist yellow colony on RS agar medium; Gram staining and microscopic examination showed negative coccidiosis; Homology analysis of 16S rDNA gene sequence showed that the isolated strain CLW-1 belongs to the same group with Aeromonas veronii, and the homology is above 99%; Inoculation test results of zebrafish showed that the strain had high pathogenicity, and its LD50 was 1.26×106 CFU; The virulence gene amplification results showed that the isolated strain CLW-1 had virulence genes of aerA, Exu, Lip and Ser; Drug sensitivity test results showed that the isolated strain CLW-1 was sensitive to piperacillin, cefazolin and enrofloxacin, etc; Susceptibility to furazolidone, oxacillin and erythromycin, etc; Resistance to midecamycin, clindamycin and vancomycin, etc. According to the test results, the isolated strain CLW-1 is confirmed to be Aeromonas veronii, which carries virulence genes and has high pathogenicity, it has great therapeutic effect by applying corresponding antibiotics, thus laying a foundation for the prevention and treatment of Aeromonas veronii.  相似文献

Eight calves were immunized with live auxotrophic Salmonella typhimurium mutants (aro -SL 1479, gal E 3821) and twelve calves with phenol-killed whole-cell S. typhimurium vaccine, respectively. The clinical status of the animals was followed and serial reisolation of vaccine and challenge strains from faeces was attempted. The immunization of calves with the live aro- auxotrophic S. typhimurium SL 1479 mutant proved to be unsuitable due to the death of calves after revaccination. The calves immunized with live auxotrophic gal E S. typhimurium CCM 3821 mutant proved to be protected against challenge with virulent S. typhimurium 4/5 strain administered orally at a dose of 10(6) colony forming units (CFU). The postvaccination complications showed serious shortcomings. The immunization of calves with three doses of whole-cell inactivated vaccine containing 5 strains of S. typhimurium was effective against oral challenge with virulent S. typhimurium 4/5 at a dose of 10(6) CFU.  相似文献

The purpose of the study was to evaluate the safety and efficacy of a galactose epimeraseless mutant of Salmonella typhimurium administered as an oral vaccine to one week old calves and to investigate properties of galactose epimeraseless mutants which affect their virulence and immunogenicity. The galactose epimeraseless mutant S. typhimurium strain G30D caused diarrhea and fever in three calves to which it was administered orally at a dose of 10(10) organisms; all three calves died following challenge with virulent S. typhimurium ten days postvaccination. Mild illness developed in four calves vaccinated with a dose of 9 X 10(6) organisms and one of these calves survived challenge. Three unvaccinated calves died following challenge. The vaccine organism persisted in tissues and was shed for a prolonged period by calves which received 10(10) organisms. Studies of characteristics of galactose epimeraseless mutants of S. typhimurium showed that, in the presence of galactose, there is selection for secondary mutants which are galactose resistant. The studies indicate that galactose epimeraseless mutants of S. typhimurium are not good candidate live vaccine organisms for use in calves.  相似文献

A virulent Salmonella dublin isolate was made histidine-requiring (his-) to allow recognition. The his- derivative, SL1367 (still calf-virulent), was then given by transduction and mutation, a transposon-generated non-reverting aromatic biosynthesis (aro) defect; this defect caused loss of virulence for the mouse. The his- aro- derivative strain, SL1438, was effective as a live vaccine in mice. Twenty male Holstein calves were divided into 4 groups. Groups I, II, and III were vaccinated IM at 2 weeks and at 3 weeks of age with aromatic-dependent (aro-) S dublin strain SL1438. Groups I and III received freshly prepared vaccine and group II received lyophilized vaccine. Serious adverse reactions to the vaccination were not seen. After vaccination, the mean maximum increase in rectal temperature was 1.8 C in group I and III calves and 0.6 C in group II calves. Fewer group II calves developed diarrhea (1 of 5) or positive blood cultures (0 of 5) after vaccination compared with group I and III calves (6 of 10 and 5 of 10, respectively). Postvaccination diarrhea was mild and of short duration. Group IV was comprised of 5 nonvaccinated calves. At 5 weeks of age, all calves were challenge exposed orally. Group I, II, and IV calves were challenge exposed with 10(11) virulent S dublin SL1367. Group III was challenge exposed with 10(11) virulent S typhimurium UCD 108-11. Subsequently, fever and diarrhea (lasting 1 to 3 days), but no deaths, were observed in the vaccinated calves. Four of the 5 nonvaccinated (group IV) calves died (P less than 0.001) within 8 days after challenge exposure. Aro- S dublin SL 1438 did not cause serious adverse effects and provided protection against oral challenge exposure with either virulent S dublin or S typhimurium.  相似文献

The purpose of this study was to evaluate the effectiveness of an aromatic-dependent mutant of Salmonella typhimurium as a parenteral vaccine for prevention of fecal shedding of Salmonella spp. Pigs and chickens were vaccinated IM, with 1 x 10(9) and 1 x 10(8) organisms, respectively, followed by a second identical vaccination 2 weeks later. Salmonella organisms were not detected by analysis of fecal or cloacal swab specimens from any animal after vaccination. Deleterious side effects were not noticed after vaccination. Pigs were challenge-inoculated PO with 1 x 10(12) virulent S typhimurium 1 week after the second vaccination. Chickens were challenge-inoculated PO with 3 x 10(8) organisms of either S enteritidis or the virulent parent strain of S typhimurium 3 weeks after the second vaccination. Vaccinated pigs shed Salmonella spp significantly less frequently than did nonvaccinated pigs. Vaccinated chickens challenge-inoculated with either S enteritidis or S typhimurium also shed Salmonella less frequently than the corresponding nonvaccinated control birds; however, the difference was not significant.  相似文献

Ten Holstein calves were divided into 2 groups. Five calves served as nonvaccinated controls, and 5 calves were vaccinated IM at 2 and 3 weeks of age with 10(9) aromatic-dependent (aro-) Salmonella typhimurium strain SL1479 containing O antigens 1, 4, 12. Serious adverse reactions to vaccination were not observed in the calves. Mean maximum rectal temperature increase in the vaccinated calves was 1.5 C. One calf had diarrhea and depressed appetite for 1 day after vaccination. At 5 weeks of age, all calves were challenge exposed orally with 1.5 X 10(11) virulent S dublin strain SL1367 (O antigens 9,12). After challenge-exposure inoculum was given, 1 of 5 vaccinated calves died and 4 of the 5 nonvaccinated calves died (P less than 0.05). Thus, some cross serotype protection against S dublin was induced by parenteral vaccination of calves with aro- S typhimurium strain SL1479, although protection was not complete.  相似文献

In order to understand the characteristics of S gene of porcine epidemic diarrhea virus (PEDV) in Guizhou province, and grasp the genetic variation of PEDV, according to 3 pairs of specific primers designed in the test, S gene of 6 strains of PEDV from Guizhou were amplified by RT-PCR, cloned and sequenced. Nucleotide sequence and phylogenetic tree of 6 strains of PEDV from Guizhou and reference strains were analyzed by biological information software. The result showed that the length of S gene genome of 6 strains of PEDV from Guizhou was 4 161 bp, encoding 1 387 amino acids, and the nucleotide homologies of domestic and foreign PEDV reference were from 93.3% to 98.8%, the amino acid identity were from 91.6% to 98.9%. Phylogenetic tree analysis results showed that the 6 strains were close to the American strain, Vietnam strain and the Shandong strain.They were far from the CV777, LZC, Brl and 83P-5.The experiment showed that the S gene of PEDV in Guizhou had a certain degree of amino acid change in recent year with a trend of variation. The study provided a theoretical basis for the prevention and control of PEDV in Guizhou province.  相似文献

为了解贵州省猪流行性腹泻病毒(porcine epidemic diarrhea virus,PEDV)S基因的特点,掌握其遗传变异情况,本试验设计3对特异性引物对6株PEDV贵州株S基因进行全基因RT-PCR扩增、克隆及序列测定;应用生物信息学软件将6株PEDV贵州流行株与参考毒株进行同源性与系统进化分析。结果显示,6株PEDV贵州流行株S基因的全基因长为4 161 bp,编码1 387个氨基酸,与国内外PEDV参考毒株核苷酸同源性在93.3%~98.8%之间,氨基酸同源性在91.6%~98.9%之间;进化树分析结果显示,6株流行毒株与美国毒株、越南毒株和山东毒株亲缘关系较近;与CV777株、LZC、Brl、83P-5亲缘关系较远。结果表明,PEDV贵州地方流行毒株S基因编码的氨基酸存在一定程度变化,近年来呈现变异趋势。本研究可为贵州省PEDV的防控提供一定的理论依据。  相似文献

为了解贵州省三穗县鸭疫里默氏杆菌(Riemerella anatipestifer,RA)的流行血清型、毒力和耐药情况,本试验从贵州省三穗县6个规模养鸭场临床疑似RA感染的病鸭体内分离出6株分离菌,对病原菌进行分离鉴定、毒力基因检测和耐药性分析。细菌分离鉴定结果显示,分离菌在巧克力琼脂培养基上长出表面光滑、圆形半透明的滴状菌落;革兰氏染色呈阴性短小杆菌,瑞氏染色呈两极浓染;分离菌均不具运动性,尿素、触酶和氧化酶试验均为阳性,符合RA生化特性,将6株分离菌分别命名为SS-RA1~SS-RA6;6株分离菌的16S rRNA基因序列与NCBI上RA参考菌株的基因序列相似性≥98%,说明6株分离菌均是RA;SS-RA1~SS-RA4为血清2型且含有8种毒力基因(OmpA、CAMP、wza、AS87_04050、Fur、SIP、TbdR1和luxE基因),SS-RA5和SS-RA6为血清11型,缺失AS87_04050基因,仅含有上述其余7种毒力基因;动物回归试验结果显示,攻毒组雏鸭均全部死亡,对照组雏鸭未表现明显临床症状,表明6株分离菌对雏鸭均有致病力;药敏试验结果显示,6株分离菌仅对羧苄西林、哌拉西林、头孢他啶、头孢氨苄、头孢曲松、头孢拉定和头孢哌酮7种抗菌药物敏感,对其他13种抗菌药物均表现不同程度的耐药,对氨基糖苷类和大环内酯类抗菌药物的耐药率为100%。本试验成功分离得到6株RA,为贵州省三穗县鸭疫里默氏杆菌病的疫苗选择和药物防治提供理论依据。  相似文献

Porcine carcasses contaminated with Salmonella typhimurium pose significant public health problems. Prolonged faecal shedding of Salmonella in pigs contributes to the contamination level of carcasses. Although the mechanism of prolonged faecal shedding is not yet clarified, the CS54 Island, and more specifically the shdA gene encoding a fibronectin binding autotransporter protein, was identified as an important locus for intestinal colonization and persistence of Salmonella typhimurium in mice. The aim of this study was to assess the contribution of ShdA in faecal shedding of Salmonella typhimurium in pigs. Pigs were orally inoculated with a Salmonella typhimurium wild type field strain or its isogenic shdA mutant strain. For the first few days after inoculation, the shdA mutant strain was excreted more, the diarrhoea was more pronounced and higher numbers of internal organs were infected. No effect on long-term shedding was found. In a porcine ileal loop model, the wild type strain and shdA mutant strain did not show any differences in the induction of neutrophil influx into the intestinal wall and lumen. In conclusion, we have shown that a Salmonella typhimurium deletion mutant in shdA is more virulent during the first days after inoculation and is not significantly impaired in persistence or prolonged shedding in pigs.  相似文献

Migration of bovine macrophages under agarose was used to assess cellular immunity in 7 nonvaccinated calves and 9 calves vaccinated with Salmonella typhimurium. The 9 vaccinated calves were allotted to 4 groups. Group I calves were vaccinated twice orally with small doses of virulent S typhimurium; group II calves were vaccinated twice orally with genetically altered aromatic-dependent (aro-) S typhimurium SL3261; group III calves were vaccinated twice IM with small doses of virulent S typhimurium; and group IV calves were vaccinated twice IM with aro- S typhimurium SL1479. Samples of blood were obtained from these calves at 2 weeks after the 2nd vaccinal dose was given, and lymphocytes were harvested, using lymphocyte separation medium. Lymphocytes in serum-free medium were then incubated with S typhimurim antigen for 48 hours. Lymphocytes were then transferred to antigen-free medium and incubated for 48 hours, and the supernatant was assayed for the migration-inhibition factor (MIF). Lymphocyte supernatant was assayed for MIF by incubating it for 48 hours with 2.0 X 10(4) alveolar macrophages in agar wells. The macrophage migration distance was measured and compared with control values. Macrophage migration was inhibited in the presence of supernatant of lymphocytes from vaccinated calves that had been incubated with antigen, indicating the presence of the MIF in the supernatant. Migration distances, as a percentage of control, were 33% for group I calves (oral vaccination, virulent vaccinal organism), 60% for group II calves (oral vaccination, aro- vaccinal organism), 41% for group III (IM vaccination, virulent organism), and 25% for group IV (IM vaccination, aro- vaccinal organism).(ABSTRACT TRUNCATED AT 250 WORDS)  相似文献

将猪链球菌溶血素(suilysin,SLY)基因克隆入原核表达栽体pBV220,将重组质粒再导入减毒鼠伤寒沙门氏菌SV4089株,经PCR和酶切鉴定,构建成携带猪链球菌溶血素基因的重组减毒鼠伤寒沙门氏菌。结果表明:该减毒株具有相对安全性;用酶切和PCR鉴定法证实在无抗生素存在的条件下携带重组质粒的减毒株比较稳定;SDS-PAGE显示SLY能在宿主菌中进行表达。该结果为进一步研究制备猪链球菌口服活疫苗奠定了基础。  相似文献

The purpose of this study was to investigate the molecular types and antimicrobial resistance of Salmonella isolated from pigeon in Shanghai from 2011 to 2014. A total of 92 fecal samples were collected from markets. XLD plate and Salmonella chromogenic medium were used to isolate suspected Salmonella colonies, and then determined the number of Salmonella through gram staining and biochemical tests. Among them, 24(26.1%) were positive for Salmonella. Kauffmann-White method and the K-B method were used respectively for serotype identification and susceptibility testing. In result, serological identification showed that 24 isolates from pigeon could be divided into 3 serotypes including S.typhimurium (66.7%), S.agona (25.0%) and S.corvallis (8.3%). Drug susceptibility test showed that 75.0% of the isolates were resistant to one antibiotic at least. The highest level resistance were found for tetracycline as well as sulfisoxazole (62.5%), followed by streptomycin (58.3%), nalidixic acid (50.0%) and ampicillin (20.8%). Multi-drug resistant strains accounted for 62.5% (15), in which the largest number of strains (7, 16.7%) were resistant to four drugs. In addition, isolates were 100.0% susceptible to cefepime, cefotaxime, ceftazidime, imipenem and ofloxacin, but 12.5% were moderately sensitive to the amoxicillin/clavulanic acid and ciprofloxacin. The study showed that Salmonella had a high separation rate of pigeon in Shanghai farmers market and performed serious multidrug resistance, which would bring great challenges and risks to the prevention and control of Salmonella in food, so that the prevalence and drug resistance of Salmonella in pigeon deserved close attention.  相似文献

本试验旨在构建表达猪繁殖与呼吸综合征病毒(PRRSV) GP5蛋白的口服重组减毒鼠伤寒沙门氏菌活载体疫苗株.PCR克隆除去信号肽序列的PRRSV ORF5基因,将其插入到表达载体pYA3341中,构建重组质粒pYA3341-ORF5.将重组质粒电转入鼠伤寒沙门氏菌疫苗株X4550(缺失Asd、Cya、Crp基因),获得重组疫苗菌株X4550(pYA3341-ORF5).鉴定重组菌GP5蛋白的表达;测定重组菌定居特性及安全性;检测免疫小鼠血清抗体;流式细胞仪检测重组菌对小鼠T淋巴细胞CD4+和CD8+亚群的影响;最后进行免疫猪血清抗体检测.结果表明,酶切鉴定证实重组质粒构建成功;Western blot证实重组菌表达的GP5蛋白能与PRRSV阳性血清特异性结合;重组菌在体内可较稳定地定居于小鼠的肠系膜淋巴结和脾脏中,并在其中表达出GP5蛋白;小鼠口服试验证实重组菌无毒性作用;重组菌口服免疫小鼠可以产生抗GP5蛋白抗体且抗体具有中和活性;重组菌株能不同程度地使CD4+、CD4+/CD8+升高,而使CD8+下降,表明重组菌对细胞免疫功能具有调节作用;淋巴细胞增殖试验表明,重组菌能诱发小鼠产生较强的细胞免疫应答;重组菌口服免疫猪可以产生抗GP5蛋白抗体.本试验成功构建了能稳定表达PRRSV GP5蛋白的口服减毒鼠伤寒沙门氏菌疫苗株,为研究PRRSV口服基因工程疫苗奠定基础.  相似文献
