健康杂种猪6头,体重(31.3±1.8)kg,用于研究猪链球菌病对磺胺二甲嘧啶(SM2)一次和多次静注药代动力学的影响。用重氮化一偶合比色法测定血药浓度,矩量法分析药一时数据,预测方程法模拟多次给药的浓度曲线。结果显示,猪链球菌病使SM2一次给药的清除率显著下降(P<0.05),多次给药的清除率极显著升高(P<O.01),传统的预测方程对病态下SM2多次给药的动力学不适用。本文对病态下清除率变化的原因进行了讨论。  相似文献

选用21头健康杂种猪,随机分为3组,对静注、肌注及内服恩诺沙星(2.5mg/kg)的生物利用度和药物动力学进行了研究。用乙腈提取血浆中的药物,反相高效液相色谱法测定血浆中恩诺沙星及其主要代谢产物环丙沙星的浓度。所得血药浓药-时间数据用MCPKP计算机程序处理。静注给药的药时数据适合二室开放模型,主要药物动力学参数:t1/2a0.48±0.24h;t1/2a3.45±0.85h;t1/2K102.0  相似文献

本试验采用三合激素(ITC)为主的处理方案,对繁殖季节中179只母羊进行了同期发情处理。按55kg以上体重的母羊第一次注ITC2.0ml,间隔14天,第二次注1.0ml;55kg以下母羊第一次注1.5ml,第二次注0.8ml,于第二次注射同时随机分为6组。①PMSG190IU;②LRH─A_250μg+PMSG150IU;③LRH-A_350μg+PMSG150IU;④FSH20单位+PMSG150IU;⑤LH50单位+PMSG150IU;⑥PMSG500IU或LRH-A_3100μg。试验结果表明,第一次注射和第二次注射ITC,第3组同期发情率较高(P<0.05),产羔率以第2组为高(P<0.05),成活则以第4组和第6组8为最高(P<0.05)。试验初步结论为:三合激素与PNISG、LRH─A_2配合处理,可利用于绵羊同期发情。  相似文献

用二次回归正交设计研究了全玉米-豆饼饲粮的粗蛋白(CP)水平(x_1)和豆饼(占饲粮蛋白%)比例(x_2)对3周龄断奶至8周龄的仔猪日增重(ADG)、饲料利用率(F/G)、腹泻指数(DI)、血清脲素氮(SUN)和木糖吸收率的影响。除木糖吸收率外(随CP和SBM增加,木糖吸收率下降),其余指标的二次回归方程均达显著或极显著水平。方程如下:分析表明,在CP16~24%下,调整豆饼比例可使仔猪保持相近的生长速度和饲料利用率;高蛋白腹泻的实质是过敏性腹泻;当腹泻指数高到77时,日增重和饲料利用率下降。推荐3周龄断奶仔猪玉米-豆饼饲粮中适宜蛋白水平为24%,豆饼比例为60%。  相似文献

本文报道了用聚合酶链反应(PCR)获取IBvM_(41)株和广东肾变株D_(41)免疫原(Sl)基因的研究情况,所用引物为IBVBeaudette株S1基因两侧的对应序列,跨幅为1.7kb;DNA模板为IBVM_(41)和D_(41)株经病毒RNA提取后反转录而得;结果表明,2株病毒所获的PCR产物与预期一致。  相似文献

实验用PMSG或PMSG+HCG处理或未经激素处理的海狸鼠8只,共获卵巢卵母细胞138枚。激素处理对获取卵巢卵母细胞的数量没有影响,而对体外成熟发育至卵丘扩展和半成熟阶段有促进作用。三种不同培养液(Whiten+FCS;TCM199+PMSG+FCS;TCM199+HCG+FCS)共培养125枚卵母细胞,培养后卵丘扩展率及半成熟率分别为56.5%,45.7%,47.6%和21.7%,12.3%,9.5%,以Whiten液较高(分别为56.5%和21.7%),但只有TCM199+PMSG+FCS组有2枚卵母细胞出现第一极体。结果表明海狸鼠卵母细胞与其它啮齿动物的卵母细胞一样,能够在体外培养成熟,完成第一次减数分裂,排出第一极体  相似文献

6头体重47.00±4.57kg(X±S)的健康长白和大白杂交猪,拉丁方设计试验,按4mg/kg静注、肌注和内服克伦特罗,高效液相色谱法检测血浆中药物浓度,MCPKP药代动力学程序处理药时数据。静注给药的药代动力学参数是:t1/2α0.62±0.12h,t1/2β4.87±1.56h,Vd(area)4.48±0.56l/kg,ClB0.63±0.11l/kg/h,AUC6.39±1.27μg/ml.h。肌注给药的药动学参数是:t1/2Ka0.22±0.10h,t1/2α0.56±0.21h,t1/2β4.25±1.10h,tmax0.60±0.13h,Cmax1.27±0.35μg/ml,AUC5.48±1.29μg/ml.h,F85.40±4.69%。内服给药的药动学参数是:t1/2Ka0.28±0.15h,t1/2Ke3.15±0.36h,tmax1.46±0.19h,Cmax0.65±0.13μg/ml,AUC3.93±0.99mg/l.h,F61.02±10.90%。肌注给药的生物利用度与内服比较,差异极显著(P<0.01)。  相似文献

Vitamin D is an essential hormone in vertebrates. Most animals acquire this hormone through their diet and/or exposure to ultraviolet B (UVB) radiation. The objectives for this research were to evaluate the clinical and physiologic effects of artificial UVB light supplementation on rabbits (Oryctolagus cuniculus) and to evaluate the long-term safety of artificial UVB light supplementation over a 6-month period. Twelve New Zealand white rabbits were randomly assigned to one of two treatment groups: Group A was exposed to 12 hours of artificial UVB radiation daily and Group B received ambient fluorescent light with no UVB supplementation for 12 hours daily. All animals were offered the same diet and housed under the same conditions. Blood samples were collected every 3 weeks over 6 months to measure blood chemistry values, parathyroid hormone, ionized calcium, and serum 25-hydroxyvitamin D3 (25-OHD3) levels. Serial ophthalmologic examinations were performed at the beginning of the study and every 2 months thereafter. At the end of the study the animals were euthanized and necropsied. Mean ± SD serum 25-OHD3 concentrations differed significantly (P = 0.003) between animals provided supplemental UVB radiation (83.12 ± 22.44 nmol/L) and those not provided UVB radiation (39.33 ± 26.07 nmol/L). There were no apparent negative clinical or pathologic side effects noted between the groups. This study found that exposing rabbits to UVB radiation long term significantly increased their circulating serum 25-OHD3 levels, which was sustainable over time.  相似文献

To compare the nutrient digestibility of soybean meal (SM) and pigeon pea seed meal (PM) as well as morphological intestinal alterations in piglets fed them, three pigs per group were randomly selected at the end of the feeding experiment for ten days. Growth performance was higher in the SM group than in the PM group (p<0.05). The digestibility of crude protein, crude fat and crude fiber was 80.6%, 23.6% and 52.4% in the SM group, while in the PM group, values of 49.8%, 23.6% and 43.2% were observed, respectively. Digestible energy was 3.26 kcal g(-1) in SM and 3.17 kcal g(-1) in PM. It was concluded that the digestibility of PM was lower than that of SM; almost half of the protein in PM was digested. Dietary treatments had no effect on length of each small intestinal segment and weight of visceral organs (small intestine, liver, heart, spleen, kidney, stomach and lung) except the decreased kidney weight in the PM group (p<0.05). The epithelial cells on the jejunal villi showed a dome-like shape in the SM group, but they were a flat shape in the PM group. The present digestion trial and histological intestinal data suggest that the intestinal digestive and absorptive functions are much more atrophied in the PM group than in the SM group, and demonstrate that histological intestinal alterations might be well related with the intestinal functions.  相似文献

This study was conducted to establish the relationship between histological alterations of the intestinal villus and intestinal function, and to histologically assess the enteral nutrient absorption of raw pigeon pea seed meal (PM). Piglets were allotted into five groups: feed withdrawal for 3 days or 5 days, refeeding soybean meal (SM) or PM for 2 days after the 3‐day fast, and access ad libitum to conventional feed for the last 2 days without fasting. After the end of each experiment, the intestinal segment and duodenum of each piglet were processed and examined by light and scanning electron microscopy, respectively. After fasting, villus height, cell area and cell mitosis number were decreased in most parts of each intestinal segment, and duodenal villus tips had a smooth surface. After refeeding SM or PM, the decrease in body weight improved, but there was not a significant difference between them. The atrophic morphology of the intestine was reversed by refeeding SM, suggesting that the intestinal villi might be activated by absorption of SM. On the other hand, in the piglets refed PM, the recovery of light microscopic parameters and duodenal villus tip morphology was less clear than with SM. The present histological results suggest that although the absorption of PM is less than that of SM, the PM nutrients might be effectively absorbed, even in the raw form, and establish that in pigs intestinal histological alterations correspond well with intestinal absorptive function.  相似文献

The purpose of this study was to determine the specific effects of extending the interval between dwell time and the duration of scalding on pork quality attributes. Sixty-four Duroc x Yorkshire pigs were randomly assigned to a 2 x 2 factorial treatment arrangement. Treatments included extending the dwell duration from 5 to 10 min and extending the scald duration from 5 to 8 min. All carcasses entered the cooler 50 min after exsanguination. At exsanguination, blood was collected for three 1-min intervals and then for a final 2-min period. Temperature and pH of the LM and semimembranosus muscle (SM) were measured at 45 min, and at 2, 4, 6, and 24 h postmortem (PM). Hunter L*, a*, and b* values were determined on the LM, SM, and biceps femoris (BF). Purge loss was measured on the SM, BF, and the sirloin end of the loin. Drip loss was measured in duplicate from LM chops after 1 and 5 d of storage. Warner-Bratzler shear force (WBS) measurements were determined on LM chops aged 1, 3, 5, and 7 d PM. Over 99% of the collected blood was obtained during the first 3 min after sticking. Carcasses scalded for 8 min had greater (P < 0.05) semi-membranosus 2 h temperature (28.8 degrees C) than carcasses scalded 5 min (27.3 degrees C). An 8-min scald process resulted in longissimus dorsi chops with lower hue angle and greater WBS values than the 5-min scald process. Increasing dwell time from 5 to 10 min resulted in biceps femoris chops with greater hue angle and loin chops with greater WBS values at 3 d PM. Harvest processes did not significantly affect subjective quality scores, Hunter L* values, purge or drip loss. Lengthening the duration of dwell and scalding may result in a more rapid PM pH decline. Reducing the duration of scalding may lead to increased time for manual removal of hair. Because of differences in facilities, it is recommended that individual facilities monitor dwell and scald durations to determine how to best minimize time of entry into the cooler.  相似文献

The objective of this study was to determine whether differences in pork tenderness and water-holding capacity could be explained by factors influencing calpain activity and proteolysis. Halothane-negative (HAL-1843 normal) Duroc pigs (n = 16) were slaughtered, and temperature and pH of the longissimus dorsi (LD), semimembranosus (SM), and psoas major (PM) were measured at 30 and 45 min and 1, 6, 12, and 24 h postmortem. Calpastatin activity; mu-calpain activity; and autolysis and proteolysis of titin, nebulin, desmin, and troponin-T were determined on muscle samples from the LD, SM, and PM at early times postmortem. Myofibrils from each muscle were purified to assess myofibril-bound (mu-calpain. Percentage drip loss was determined, and Warner-Bratzler shear (WBS) force was analyzed. Myosin heavy-chain (MHC) isoforms were examined using SDS-PAGE. The pH of PM was lower (P < 0.01) than the pH of LD and SM at 30 and 45 min and 1 h postmortem. The PM had a higher (P < 0.01) percentage of the MHC type IIa/IIx isoforms than the LD. The-LD had the greatest proportion of (P < 0.01) MHC IIb isoforms of any of the muscles. The PM had the lowest (P < 0.01) percentage of MHC IIb isoforms and a greater (P < 0.05) percentage of type I MHC isoforms than the LD and SM. The PM had less (P < 0.01) drip loss after 96 h of storage than the SM and LD. The PM had more desmin degradation (P < 0.01) than the LD and SM at 45 min and 6 h postmortem. Degradation of titin occurred earlier in the PM than the LD and SM. At 45 min postmortem, the PM consistently had some autolysis of mu-calpain, whereas the LD and SM did not. At 6 h postmortem, some autolysis of mu-calpain (80-kDa subunit) was observed in all three muscles. The rapid pH decline and increased rate of autolysis in the PM paralleled an earlier appearance of myofibril-bound mu-calpain. The SM had higher calpastatin activity (P < 0.05) at 45 min, 6 h, and 24 h and had higher WBS values at 48 h (P < 0.01) and 120 h (P < 0.05) postmortem than the LD. At 48 and 120 h postmortem, more degradation of desmin, titin, and nebulin were observed in the LD than in the SM. These results show that mu-calpain activity, mu-calpain autolysis, and protein degradation are associated with differences in pork tenderness and water-holding capacity observed in different muscles.  相似文献

Rabbit hemorrhagic disease (RHD) is an important cause of disease and mortality in wild and domestic European rabbits (Oryctolagus cuniculus) throughout the world. Testing for 2 distinct RHD virus types (RHDV/RHDVa and RHDV2) was carried out on samples collected from 684 rabbits submitted from veterinary practices and private owners throughout Europe between January 2015 and June 2017. Four (0.6%) were positive for RHDV/RHDVa and 257 (37.4%) were positive for RHDV2. RHDV/RHDVa was detected in individual samples from Germany and the Netherlands, while RHDV2 was found in animals from Germany, Great Britain, Luxembourg, The Netherlands, Spain, Switzerland, Poland, Belgium, Austria, Sweden, and Finland.  相似文献

The objectives of this study were to determine the effects of storage time (ST) and packaging method (PM) on tenderness and changes in intramuscular connective tissue (IMCT) strength of chevon. Spanish does (8 mo of age, average BW 25 kg) were harvested (n = 12), chilled at 4 degrees C for 24 h, and then fabricated into 2.5-cm-thick leg, shoulder/arm, and loin/rib cuts. The cuts from six carcasses were vacuum-packed and aged at 2 degrees C for 0, 4, 8, or 12 d. To assess the influence of a packaging method that favors oxidation on postmortem tenderization, the cuts from the remaining six carcasses were placed on styrofoam trays, overwrapped with polyvinyl-chloride film, and stored at 2 degrees C for similar periods. At each ST, longissimus (LM), semimembranosus (SM), and triceps brachii (TB) muscles were assessed for Warner-Bratzler shear (WBS) values. The WBS of uncooked meat, myofibrillar fragmentation index (MFI), and collagen solubility were assessed on LM. The IMCT samples were prepared to assess changes in mechanical strengths and for scanning electron microscopy (SEM). Intact honeycomb structures of endomysium, with no muscle fiber elements, were observable under SEM. The PM or ST did not influence the mechanical strength of IMCT preparations, as measured by a texture analyzer. Collagen solubility of LM muscles also did not change during aging. For both PM, cooked meat WBS values were higher (P < 0.01) in SM and TB than in LM. In the SM samples, the average WBS values were higher (P < 0.01) at d 0 than at other ST. Although MFI of LM increased with increasing aging time (P < 0.05), changes in WBS over ST were minimal in TB and LM samples. The WBS of uncooked LM decreased sharply up to 8 d postmortem in both PM (P < 0.05). However, there was no PM x ST interaction to indicate any adverse influence of packaging on tenderization of chevon. The results suggest that aging chevon cuts for more than 4 d may not result in significant additional improvement in tenderness.  相似文献

The objective of this study was to determine the extent of variation in, and relationships among, biochemical and palatability traits within and among 11 major beef muscles. Longissimus thoracis et lumborum (LD), psoas major (PM), gluteus medius (GM), semimembranosus (SM), adductor (AD), biceps femoris (BF), semitendinosus (ST), rectus femoris (RF), triceps brachii (TB), infraspinatus (IS), and supraspinatus (SS) from one side of 31 Charolais x MARC III steer carcasses were vacuum-packaged, stored at 2 degrees C until 14 d postmortem, and then frozen at -30 degrees C. The 2.54-cm-thick steaks were obtained from two or three locations within muscles in order to assess biochemical traits and Warner-Bratzler shear force, and from near the center for sensory trait evaluation. The PM was most tender and was followed by IS in both shear force and tenderness rating (P < 0.05). The other muscles were not ranked the same by shear force and tenderness rating. The BF had the lowest (P < 0.05) tenderness rating. The PM, GM, and LD had lower (P < 0.05) collagen concentration (2.7 to 4.5 mg/g muscle) than muscles from the chuck and round (5.9 to 9.0 mg/g), except for the AD (4.9 mg/g). Desmin proteolysis was highest (P < 0.05) for BF and LD (60.7 and 60.1% degraded), and was lowest (P < 0.05) for PM (20.2%). The PM, TB, IS, RF, and ST had relatively long sarcomere lengths (> 2.1 microm), whereas the GM had the shortest (P < 0.05) sarcomere length (1.7 microm). Cooking loss was lowest (P < 0.05) for BF (18.7%) and was followed by LD and IS (20.7%); it was highest (P < 0.05) for ST (27.4%). Across all muscles, tenderness rating was highly correlated (r > 0.60) with shear force, connective tissue rating, sarcomere length, and collagen content. Within a muscle, correlations among all traits were generally highest in LD and lowest in AD. Within muscle, location effects were detected (P < 0.05) for shear force (PM, ST, BF, SM, and RF), sarcomere length (PM, ST, BF, LD, SS, IS, SM, and RF), collagen concentration (PM, BF, SS, IS, SM. AD, TB, and RF), desmin degradation (PM, GM, BF, SM, AD, and, RF), and cooking loss (all muscles except SS and AD). There is a large amount of variation within and among muscles for tenderness traits and tenderness-related biochemical traits. These results increase our understanding of the sources of variation in tenderness in different muscles and provide a basis for the development of muscle-specific strategies for improving the quality and value of muscles.  相似文献

The pharmacokinetics of sulfadimidine (SDM) and its N4-acetyl metabolite (N4SDM) were investigated after intravenous bolus injection of a single dose (200 mg/kg) of SDM in normal and diseased New Zealand white rabbits. The apparent distribution volume at steady state, total body clearance and elimination half-life of SDM in normal animals were 0.7 +/- 0.3 l/kg, 0.57 +/- 0.24 l/kg/h and 1.6 +/- 1.3 h, respectively. Of the administered dose, 62.1% was metabolized by N4-acetylation, and 12.7 +/- 1.1 and 2.8 +/- 1.8% of the dose was excreted as free drug by the kidney and gastrointestinal tract, respectively. The 'apparent' formation and elimination half-lives of N4SDM were 0.6 +/- 0.4 and 2.2 +/- 1.1 h, respectively. The metabolite was eliminated mainly by excretion through the kidney. There was no significant effect of acute pasteurellosis on the pharmacokinetics of either SDM or N4SDM in rabbits.  相似文献

用从甘肃省马气喘病自然病例肺中分得的5株放线菌和5株霉菌,对34只家兔和23只豚鼠进行了实检性农民肺研究。通过吸入与气管内注入上述菌粉,在这些动物中首次成功地复制出农民肺模型。染菌动物的症状与病变同在气喘病马所看到的很相似。尸检时见肺膨大、气肿,还有密散的结节。镜检动物的肺组织,观察到单核巨噬细胞性肺泡炎、间质性肺炎、支气管周炎与上皮样-巨细胞肉芽肿形成,但豚鼠比家兔的淋巴网状组织增生更明显。结果表明,所分得的霉菌和放线菌包括热吸水链霉菌参与了甘肃马农民肺的病理发生。  相似文献
