在江苏扬州的罗氏沼虾(Mocrobrochium rosenbergii)养殖区,选取具“白体病”典型症状的活体仔虾和幼虾,对其腹部肌肉的病变部位进行显微和超微结构观察,未见病原生物;光镜下未见明显病理变化;其超微病理变化主要在病程中后期可见,病变部位肌丝断解、形成肌溶灶;线粒体变性、并呈空泡化;特别是肌原纤维间脂滴明显增多,呈串珠状排列。分析该病可能是一种线粒体肌病,属脂质沉积性肌病。  相似文献

Total lipid content, lipid classes and fatty acid composition were analysed in tissues from two eelpout species fed on the same diet, the Antarctic Pachycara brachycephalum and the temperate Zoarces viviparus, with the aim of determining the role of lipids in fishes from different thermal habitats. The lipid content increased with decreasing temperature in the liver of both species, suggesting enhanced lipid storage under cold conditions. In P. brachycephalum, lipid composition in the liver and muscle was strongly dominated by triacylglycerols between 0 and 6°C. In contrast, in the temperate species, lipid class composition changed with changes in the temperature. When acclimatized to 4 and 6°C Z. viviparus not only displayed a shift to lipid anabolism and pronounced lipid storage, as indicated by high triacylglycerol levels, but also a shift to patterns of cold adaptation, as reflected by an increased content of polyunsaturated fatty acids in the lipid extract. Unsaturated fatty acids were also abundant in the Antarctic eelpout, but when compared to Z. viviparus at the same temperatures, the latter had significantly higher ratios of polyunsaturated to saturated fatty acid levels, whereas the Antarctic eelpout showed significantly higher ratios of monounsaturated to saturated fatty acid levels. High δ-15N values of the Antarctic eelpout reflect the high trophic level of this scavenger in the Weddell Sea food web. Stable carbon values suggest that lipid-enriched prey forms a major part of its diet. The strategy to accumulate storage lipids in the cold is interpreted to be adaptive behaviour at colder temperatures and during periods of irregular, pulsed food supply.  相似文献

研究了低温烘干、真空冻干和微波干燥3种干燥方式对蛋白核小球藻粗脂肪、脂肪酸含量和种类组成的影响及不同保存条件下脂肪酸的变化规律。试验结果表明,低温烘干藻粉的粗脂肪含量明显低于真空冻干藻粉和微波干燥藻粉。3种干燥方式对小球藻脂肪酸含量无显著影响,但藻粉颜色明显不同。藻粉脂肪酸含量与鲜藻之间无显著差异。在小球藻粉保存过程中,脂肪酸种类组成保持不变。藻粉在常温(14.5~18.5℃)下保存半年、4℃下保存1年时,多不饱和脂肪酸含量显著减少,饱和脂肪酸和单不饱和脂肪酸含量显著增加。在-24℃下保存1年时,各脂肪酸含量无明显变化。  相似文献

The main aim of the present work was to evaluate the effect of dietary butylated-hydroxytoulene (BHT as an antioxidant) and dietary lipid level on flesh lipid quality of Beluga sturgeon during frozen storage. An initial 135-day feeding trial evaluated practical-type diets containing 0 or 250 mg BHT kg?1 with two lipid levels (12 and 24% diet on dry matter basis) in a factorial arrangement. Fillet samples were analyzed fresh or after storage at ?18 ± 1°C for 12 months. Dietary antioxidant supplementation had no significant effect on fat content and fatty acid composition in the studied fish muscle, but increasing the lipid concentration in the feed increased muscle lipid concentration. Lipid oxidation in fish muscle is related to increasing lipid concentration in the feed. In addition, oxidation was reduced for fish fed BHT. Results showed that dietary BHT supplementation can slow down the level of lipid oxidation in Beluga sturgeon muscles during frozen storage, but it had a direct relationship with dietary lipid level.  相似文献

Reactive oxygen species (ROS) are generated in all aerobic organisms. Free radicals are highly reactive ROS that cause damage to biological materials. Fish is rich in polyunsaturated fatty acids, and hence, very prone to lipid peroxidation. Both lipid and protein oxidations are important for quality loss during storage of fish, with high impact on taste and texture. Also, there are interactions between protein and secondary lipid oxidation products (aldehydes) that occur in foods because the oxidation products from one reaction can further react with both lipids and proteins respectively. This review focuses on the mechanisms and pathways of the lipid and protein oxidation and their possible relationship. Additionally, the target amino acids and final impacts of this relationship were considered. We propose that the products of lipid oxidation promote protein oxidation in fish rather than the other way around specially, during frozen storage, while during postmortem changes protein oxidation dominates. Finally, it seems that, secondary products of lipid oxidation might have more impact on the functionality of proteins from both Michael addition and Schiff base reaction rather than lipid hydroperoxides and lipid radical transfer.  相似文献

The effectiveness of milk protein concentrate (MPC) and caffeic acid in controlling lipid oxidation and texture changes in 6-month frozen stored sardine mince was investigated using two different concentrations of caffeic acid (200 and 400 ppm) with and without MPC (4%). The extent of lipid oxidation was monitored by measuring thiobarbituric acid reactive substances (TBARS) while changes in the texture of mince due to frozen storage were assessed using Texture Analyzer (TA.XT Plus) and sensory analysis. Cooking loss upon frozen storage was also determined. The addition of MPC and/or caffeic acid significantly reduced lipid oxidation throughout the storage period. However, neither added benefit nor synergistic effect in retarding lipid oxidation was shown by caffeic acid. MPC not only retarded lipid oxidation but also moderated texture hardening, cooking loss, and moistness during frozen storage. No improvement by caffeic acid alone and no significant differences in sensory firmness and moistness between MPC-added mince with and without caffeic acid indicate that caffeic acid did not play any role in texture and moisture control. This study suggests that lipid oxidation and texture of frozen sardine mince can be better controlled by using MPC alone.  相似文献

The present study investigated the effects of lipid oxidation on quality deterioration in the ordinary and dark muscles of skipjack tuna Katuwonus pelamis during the early stages of ice storage for 72 h. The lipid hydroperoxide content of the dark muscle was significantly higher (P < 0.01) than that of the ordinary muscle throughout 72 h of ice storage. The metmyoglobin content of the ordinary muscle gradually increased, and was accompanied with darkening in the fish meat color. On the other hand, the addition of sodium ascorbate or 6-hydroxy-2,5,7,8-tetramethylchroman-2-carboxylic acid (Trolox®) to the ordinary muscle of skipjack tuna significantly inhibited the formation of lipid hydroperoxide as well as metmyoglobin formation. Thus, a decrease in α-tocopherol content in the ordinary muscle with antioxidant addition was not observed during ice storage period. In conclusion, the rate of lipid oxidation of skipjack tuna ordinary muscle is closely related to metmyoglobin formation, and the addition of antioxidants to fish meat is effective at inhibiting lipid oxidation as well as myoglobin oxidation in post-mortem meat.  相似文献

病鱼为威海水产养殖场感染淋巴囊肿病的牙鲆(Paralichthys olivaceus),收集病鱼的囊肿组织,匀浆破碎,采用差速离心和蔗糖密度梯度离心方法,分离纯化淋巴囊肿病毒粒子.负染后,电镜观察证实获得的病毒纯度高,杂质极少,病毒粒子呈近似于圆形的多角形,结构完整.纯化的淋巴囊肿病毒粒子经SDS-PAGE,硝酸银染色后,电泳图谱清晰显示病毒结构蛋白带共有22条,且分子量主要集中在123~26 kD.应用Western blotting法分析病毒结构蛋白的抗原性,结果显示,分子量分别为123.55 kD、65.292 kD和54.438 kD的3条蛋白带发生了免疫反应,其中分子量为65.292 kD的蛋白带反应强度明显高于其他2条蛋白带.本研究旨为确定淋巴囊肿病毒主要衣壳蛋白提供基础依据.[中国水产科学,2006,13(3):415-420]  相似文献

The objective of this experiment was to determine the effects of two levels of vitamin E (100 and 300 mg/kg diet) along with two levels of lipid (9 and 14%) and their interaction on growth performance of Indian white shrimp and consequently to evaluate the fatty acid composition and lipid stability of its muscle tissue during frozen storage. Growth of juvenile Indian white shrimp was not significantly affected by dietary vitamin E and lipid levels. Muscle lipid content of shrimp fed diets with 14% lipid was significantly higher than that of with 9% lipid. Obvious effects of the increase in dietary lipid level on muscle fatty acid composition were significant decrease in proportion of 16:0 and increase in proportion of 20:5n-3. The content of vitamin E concentration in shrimp muscle reflected dietary vitamin E concentration and ranged from 6.68 to 14.8 mg/kg muscle corresponding to two (100 and 300 mg/kg) levels of vitamin E in fish diet, respectively. Subsequently, results showed that by increasing the concentration of vitamin E from 100 to 300 mg/kg in diet, the rate of lipid oxidation in the muscle tissue during frozen storage was reduced and, as a result, caused higher HUFA retention in muscle of shrimp fed diet with high lipid level.  相似文献

Mobilization patterns of energy reserves during starvation are highly variable among crustaceans, and understanding this process is useful to satisfy the nutritional requirements of cultured organisms. The aim of this study was to elucidate the mobilization patterns on early and advanced juveniles—first free stage and one‐gram juveniles—of redclaw crayfish (Cherax quadricarinatus). A biochemical approach was used to analyse the organic carbon, total proteins, lipids and glycogen levels of the entire animal, and a histochemical approach was adopted to identify location of metabolites in the hepatopancreas and abdominal muscle. While starvation did not affect early juvenile protein levels, it showed a significant decrease in advanced juveniles. Histochemical analysis showed that lipid storage of hepatopancreas R cells was depleted near point of no return (PNR) and recovered after feeding. Glycogen storage was localized in the F cells of the hepatopancreas, and among muscle fibres of the abdomen, where after feeding, a pronounced accumulation was observed. Early and advanced juveniles of redclaw crayfish showed different patterns of consumption mainly related to the protein level, which was the most abundant reserve in advanced juveniles and the most consumed during starvation. Hepatopancreas R cells were confirmed as the principal lipid storage, whereas the abdominal muscle was the main glycogen storage. In crustacean aquaculture, the understanding of resistance to temporary feed deprivation is relevant to achieve an adequate feeding management, thereby avoiding the overfeeding and diminishing operating costs.  相似文献

As a major part of fish larval diet in nature, copepods constitute an appropriate live prey for aquaculture purposes. Considering the difficulty of mastering copepod mass production, studies on their growth performance at different environmental conditions are needed to improve their productivity. In this study a new selective approach based on temperature control is proposed to improve the physiological (body size, fecundity and lipid storage) performance of copepods. The estuarine copepod Eurytemora affinis known to have a high genetic variance in temperature tolerance was used as a biological model. First two different copepod lines were obtained after long‐term culture at constant cold (7°C) and warm (20°C) temperatures. Then both populations were transferred to a higher temperature of 24°C appropriate for aquaculture use and followed during five generations. During the first two generations (F1–F2) of a cold‐acclimated population, female body size and fecundity decreased significantly whereas the survival rate remained high. The high lipid content of this population was used by females to compensate the heat shock of more than 10°C. However, the survival rate decreased dramatically in F3 but allowed the selection of robust individuals which progressively improved their fitness during the following generations. So, compared to the warm acclimated population, the cold acclimated one showed larger body size, higher fecundity and better lipid storage. After only five generations at 24°C the cold‐acclimated population showed a significant genetic gain in prosome length compared to the warm acclimated one.  相似文献

The kinetics of astaxanthin degradation and lipid oxidation in shrimp oil from hepatopancreas of Pacific white shrimp (Litopenaeus vannamei) as affected by storage temperature were studied. When shrimp oil was incubated at different temperatures (4, 30, 45 and 60 °C) for 16 h, the rate constants (k) of astaxanthin degradation and lipid oxidation in shrimp oil increased with increasing temperatures (p < 0.05). Thus, astaxanthin degradation and lipid oxidation in shrimp oil were augmented at high temperature. When shrimp oils with different storage conditions (illumination, oxygen availability and temperature) were stored for up to 40 days, astaxanthin contents in all samples decreased throughout storage (p < 0.05). All factors were able to enhance astaxanthin degradation during 40 days of storage. With increasing storage time, the progressive formation of primary and secondary oxidation products were found in all samples as evidenced by the increases in both peroxide values (PV) and thiobarbituric acid reactive substances (TBARS) (p < 0.05). Light, air and temperatures therefore had the marked effect on astaxanthin degradation and lipid oxidation in shrimp oils during the extended storage.  相似文献

The influence of dietary protein and lipid sources on the quality of organic rainbow trout (Oncorhynchus mykiss) was studied. The protein and oil sources were fishmeal, fish oil, and organic vegetable protein and oils. Sensory profiling was performed during 3 to 14 days of ice storage along with lipid analyses of the fillet. Overall, the results showed that the sensory characteristics of the trout were affected in different ways during ice storage. The source of lipid seemed to affect the sensory quality at the beginning of the storage period, while the protein source seemed to have a more pronounced impact at the end of the storage period.  相似文献

>ABSTRACT: The lipid profile was studied along the branch length, from the top, middle to base portion, of coral Montipora digitata to gain more insight into the physiological significance of lipids in the coral energy budget. The lipids of M. digitata consisted of seven major lipid classes: polar lipid, sterol, free fatty acid, unknown lipids 1 and 2, triacylglycerol (TG), and wax ester. The concentration of storage lipids, TG, and wax ester showed a top–base gradient along the length, whereas the levels of free fatty acid and unknown lipids showed a base–top gradient. The proportions of polar lipid and sterol in the top portion of the branch were slightly higher than those in the base portion. This observation appeared to be compatible with the view that the increased energy expenditure for proliferation enhanced the mobilization of the storage fuel lipids of wax ester and TG rather than the structure lipids of polar lipids and sterols at the top portion of the branch. Compositions of fatty acid also showed a length-wise diversity. The top portion had a lower proportion of palmitic acid (16   :   0) in all lipid classes of fatty acid ester, suggesting that this fatty acid was preferentially mobilized at the top portion, probably for the growth of coral cells.  相似文献

Herbivorous grass carp (Ctenopharyngodon idella) has been reported to exhibit low capacity to utilize high dietary lipid, but different lipid sources might affect this limited capacity. In order to compare the effects of different lipid sources with different lipid levels, juvenile grass carp were fed one of nine diets containing three oils [lard, plant oil mixed by maize and linseed oil, and n‐3 high unsaturated fatty acid‐enriched (HUFA‐enriched) fish oil] at three lipid levels (20, 60 and 100 g kg?1 dry diet) for 8 weeks. Decreased feed intake, poor growth performance, hepatic pathology and higher blood lipid peroxidation were found in 60 and 100 g kg?1 fish oil groups. Conversely, in lard and plant oil groups, even at 100 g kg?1 dietary lipid level, feed intake and growth performance did not decrease, despite histological observation revealed hepatic pathology in these groups. Plasma triglyceride and cholesterol contents increased significantly in all 100 g kg?1 dietary lipid groups. In the comparison of hepatic FA β‐oxidation among three oil groups at 60 g kg?1 dietary lipid level, impaired mitochondrial and peroxisomal FA oxidation capacity was observed in fish oil group. The results confirmed the relatively low capacity of grass carp to utilize high dietary lipid, and furthermore excess HUFA intake will result in more serious adverse effects than other FA.  相似文献

A 6-wk feeding trial was conducted to evaluate the effect of various dietary lipid levels on quantitative requirements for essential fatty acids (EFA) by juvenile Pacific white shrimp Litopenaeus vannamei . A factorial experiment with three dietary lipid levels (3, 6, and 9%) and three dietary levels (0.5, 1, and 2%) of a mixture of n-3 highly unsaturated fatty acids (HUFA) was used. An increase in quantitative requirements for EFA with increasing supplementation level of dietary lipid was not demonstrated. Results suggested that this shrimp species appeared to be able to satisfy its n-3 HUFA requirements when they were supplied at 0.5% of diet; this dietary level may be even lowered. Moreover, depressed growth was observed in shrimp fed diets with the HUFA mixture supplemented at 2%. In addition, the present study showed that increasing the dietary lipid level had an effect on lipid composition of shrimp, by increasing lipid deposition in hepatopancreas and muscle tissue, but without a significant effect on growth.  相似文献

This work is focused on the effect of using whitening agents (WA) during a process followed by frozen storage (4 months at ?18°C) on the whiteness, quality parameters, and mechanical properties of sardine surimi. The whiteness of surimi exhibited a significant improvement when whitening agents (calcium carbonate and peroxide hydrogen) were used (48.75 to 60.24). Dynamic rheological measurement showed that storage moduli, G′, was considerably higher than loss moduli, G″, for both surimi with WA and control; however, the highest viscoelastic moduli were found in the treated surimi. In addition, the microstructure of surimi with WA showed more compact matrix and higher water holding capacity. On the other hand, most tested indexes showed quality losses throughout the frozen storage in both samples. Proteins underwent denaturation as indicated by the reduction in band intensities, especially myosin heavy chain (MHC) bands. Peroxide value (PV) and thiobarbituric acid reactive substances (TBARS) measurements showed that lipid oxidation took place during storage; however, the degree of lipid oxidation was not relevant. Therefore, it was proved that the addition of whitening agents had a marked effect on the production of surimi with better functional attributes including whiteness, water holding capacity, and mechanical properties.  相似文献

Effects of dietary cholesterol levels on moulting performance, lipid accumulation, ecdysteroid concentration and immune enzymes activities of juvenile Eriocheir sinensis were investigated. Crabs were fed with feeds contained various cholesterols of 1100, 2100 and 3200 mg kg?1 in both paddy fields and laboratory experiments. In paddy fields trial, the crabs fed with diets contained 3200 mg kg?1 cholesterol achieved higher growth rate than those fed with diets contained no supplemental cholesterol (1100 mg kg?1). In laboratory trial, moulting frequencies of crabs fed with diets contained 3200 mg kg?1 cholesterol were higher than those of crabs fed with diets contained no supplemental cholesterol from the 6th to 10th moult. Further laboratory experiment indicated that intermoults of crabs fed with diets contained 3200 mg kg?1 cholesterol significantly shortened compared with crabs fed with the basal feeds (1100 mg kg?1 cholesterol). In the intermoult, total lipid content, ecdysterone concentration and three immune enzymes in crabs were increased with the increment of dietary cholesterol levels between the 7th and the 8th moult in laboratory experiments. Taken together, dietary cholesterol not only enhanced moulting performances of growth, survival and moult frequency, but also enhanced total lipid storage, ecdysterone concentration and three immune enzymes activities in the intermoult period.  相似文献

Lipid distribution pattern is an important trait, which is related to flesh quality and the yield rate of aquaculture products. Here we investigated lipid distribution patterns by oil red O staining for nine economically important fish in Thailand. The most remarkable feature was that Siluriformes species had a thick layer of subcutaneous lipids, possibly to compensate the evolutionary loss of scales. In addition, we found several other characteristic staining patterns: (1) dotted stains at the boundary of red and white muscles and along the subcutaneous tissue in crystal‐eyed catfish, (2) double lines of staining along the subcutaneous layer in crystal‐eyed catfish, (3) dotted stains along the horizontal septum lines in hybrid catfish, (4) pronounced retroperitoneal fat accumulation in Nile tilapia and red tilapia, (5) pronounced fat accumulation in the inclinator muscle of the fins in Indian spiny turbot, (6) intense staining under the scales of common carp and (7) three dotted stains in soft finrays in clown knifefish. Although lipid distribution patterns are known to be affected by environmental conditions, the observed species differences suggest the strong genetic control of lipid distribution. This trait should be considered as a target of selective breeding especially for Siluriformes, in addition to commonly used parameters such as growth rate and disease resistance.  相似文献
