本研究以长牡蛎(Crassostrea gigas)F1全同胞家系为作图群体,在已构建的基于120个微卫星和66个SNP标记的长牡蛎性别平均连锁图谱上,利用PROC QTL 2.0软件对出肉率和壳形(壳宽和壳深)性状进行QTL定位分析。结果表明,共检测到13个相关的QTL,分布在3个连锁群上;其中,与出肉率相关的4个QTL定位在1号和3号连锁群上(LG1和LG3),表型解释率为0.25%~47.53%;与壳宽相关的3个QTL定位在10号连锁群上(LG10),表型解释率为0.71%~45.39%;与壳深相关的6个QTL也定位在LG10,表型解释率为3.37%~24.78%。根据QTL连锁群分析和性状相关性分析结果可以推测,出肉率与糖原含量性状以及壳宽与壳深性状分别具有相近的遗传特征,利用与相关性状共同关联的分子标记可以同时对出肉率与糖原含量性状、壳宽与壳深性状进行遗传改良。本研究结果为今后长牡蛎相关性状候选基因克隆和分子标记辅助育种提供了参考。  相似文献

The locations and effects of quantitative trait loci (QTL) were estimated for nine characters for growth‐related traits in the Pacific abalone (Haliotis discus hannai Ino) using a randomly amplified polymorphic DNA (RAPD), amplification fragment length polymorphism (AFLP) and SSR genetic linkage map. Twenty‐eight putatively significant QTLs (LOD>2.4) were detected for nine traits (shell length, shell width, total weight, shell weight, weight of soft part, muscle weight, gonad and digestive gland weight, mantle weight and gill weight). The percentage of phenotypic variation explained by a single QTL ranged from 8.0% to 35.9%. The significant correlations (P<0.001) were found among all the growth‐related traits, and Pearson's correlation coefficients were more than 0.81. For the female map, the QTL for growth were concentrated on groups 1 and 4 linkage maps. On the male map, the QTL that influenced growth‐related traits gathered on the groups 1 and 9 linkage maps. Genetic linkage map construction and QTL analysis for growth‐related traits are the basis for the marker‐assisted selection and will eventually improve production and quality of the Pacific abalone.  相似文献

为了克服单个家系数量性状位点(QTL)检测效率低、假阳性高等缺点,实验利用250对微卫星(SSR)标记对镜鲤8个全同胞家系的522尾子代进行基因组扫描,采用半同胞家系的分析策略对镜鲤体长(SL)和体质量(BW)性状进行QTL分析。结果显示,基于父系的QTL分析,共检测到4个QTL区间,其中,3个体长的QTL中,1个为95%基因组水平(genome-wide)显著性,位于LG24,可解释表型变异率为20.3%;其余2个均为95%染色体水平(chromosome-wide)显著性,分别位于LG6和LG30,可解释表型变异率分别为11.9%和11.6%。1个体质量的QTL达到99%基因组水平,位于LG24,可解释表型变异率达到38.3%,且与体长QTL区间重叠。基于母系的QTL分析,共检测到8个QTL区间,其中,5个体长的QTL中,1个为99%染色体水平,位于LG8,可解释表型变异率为16.6%;其余4个均为95%染色体水平,分别位于LG24、LG30、LG31和LG45,可解释表型变异率为9.6%~14.2%,且位于LG24和LG30上的QTL为父母本共有;3个体质量的QTL均与体长QTL区间重叠,1个为95%染色体水平,位于LG24,其余2个均为99%染色体水平,位于LG30和LG45,可解释表型变异率分别为14.1%和13.6%。进一步分析发现,位于LG24上的体长和体质量QTL区间重叠且均为父母本共有,体质量的3个QTL均与体长QTL存在重叠区域且呈现成簇分布的特点。本研究结果不仅可以为鲤分子育种提供更可靠的标记,而且为家系和品种间QTL变异规律的探索提供基础数据。  相似文献

Growth‐related traits are the main target of genetic breeding programmes in grouper aquaculture. We constructed genetic linkage maps for tiger grouper (Epinephelus fuscoguttatus) and giant grouper (E. lanceolatus) using 399 simple sequence repeat markers and performed a quantitative trait locus (QTL) analysis to identify the genomic regions responsible for growth‐related traits in F1 hybrid grouper (E. fuscoguttatus × E. lanceolatus). The tiger grouper (female) linkage map contained 330 markers assigned to 24 linkage groups (LGs) and spanned 1,202.0 cM. The giant grouper (male) linkage map contained 231 markers distributed in 24 LGs and spanned 953.7 cM. Six QTLs affecting growth‐related traits with 5% genome‐wide significance were detected on different LGs. Four QTLs were identified for total length and body weight on Efu_LG8, 10, 13 and 19 on the tiger grouper map, which explained 6.6%–12.0% of the phenotypic variance. An epistatic QTL with a reciprocal association was observed between Efu_LG8 and 10. Two QTLs were identified for body weight on Ela_LG3 and 10 on the giant grouper map, which explained 6.9% of the phenotypic variance. Two‐way analysis of variance indicated that the QTL on Efu_LG13 interacts with the QTLs on Ela_LG3 and 10 with large effects on body weight. Furthermore, these six QTLs showed different features among the winter, summer and rainy seasons, suggesting that environmental factors and fish age affected these QTLs. These findings will be useful to understand the genetic structure of growth and conduct genetic breeding in grouper species.  相似文献

The swimming crab (Portunus trituberculatus) is an economically important species in Asian aquaculture. Implementing growth‐related traits of P. trituberculatus into genetic breeding programmes is an ongoing effort. We used a previously published genetic linkage map of P. trituberculatus, containing 55 simple sequence repeat (SSR) markers and 172 amplified fragment length polymorphism (AFLP) techniques to map quantitative trait loci (QTL) for growth‐related traits in a single full sibling F2 family. Ten growth‐related traits were measured for QTL mapping. Composite interval mapping identified 9 QTL on the female map and 16 on the male map. Individual QTL with additive effects explained 11–38% of the phenotypic variance for various traits using the female parent's map, and from 1% to 21% using the male parent's map. Two QTL explaining a large percentage of variation in body weight were detected on chromosome 17 on the female map, and on chromosome 16 on the male map, and contributed 38% and 18% of the phenotypic variance respectively. This is the first study to report the detection and positioning of major QTL affecting growth in a true crab species (Brachyura). The mapping of growth‐related QTL in this study raises the possibility of improving the growth of P. trituberculatus through marker‐assisted selection.  相似文献

为挖掘镜鲤头长及头长体长比性状的主效QTL区间,实验利用368个SSR、336个SNP标记对镜鲤良种后代杂交F1群体的68个个体进行基因型检测,运用JoinMap 4.0软件包构建遗传连锁图谱。该图谱包含535个分子标记并被分配到50个连锁群上,覆盖基因组总长度为2 244.66 cM,标记间平均距离为4.63 cM。利用MapQTL 5.0(interval mapping,IM)区间作图法进行QTL检测。结果显示,共得到2个与头长相关的QTL区间,分别分布在LG21和LG42,可解释型变异分别为28.2%、32.6%;6个与头长体长比性状相关的QTL位于LG8、LG15、LG18、LG21、LG39、LG40,可解释表型变异范围是16.4%~49.3%。全部QTL区间中贡献率大于20%的主效QTL有7个,HL-21和HL-42是头长性状的主效区间;HBR-8、HBR-15、HBR-21、HBR-39和HBR-40是头长体长比性状的主效QTL区间。利用SPSS的一般线性模型(GLM)针对另一群体进行验证,结果表明HLJ692与镜鲤头长体长比显著相关。  相似文献

Pacific bluefin tuna (Thunnus orientalis) has high market value, but its wild populations have decreased in recent years. The broodstock of Pacific bluefin tuna that were hatched artificially and reared under aquaculture conditions is beginning to be used for production. The creation of broodstock with commercially valuable traits, such as rapid growth, is therefore of great interest. Genetic linkage map‐based identification of markers associated with quantitative trait loci (QTLs) facilitates marker‐assisted selection (MAS) breeding and allows efficient genetic improvement of broodstock. Single nucleotide polymorphism (SNP)‐based genetic linkage map construction using the genotyping‐by‐sequencing method can expand the number of mapped markers and help identify growth‐related QTLs. In this study, we constructed sex‐specific maps for 24 linkage groups consisting of 677 SNP and 651 microsatellite markers. The total lengths of 93 progenies in the mapping population followed normal distribution, with an average length of 9.4 mm. We performed composite interval mapping in the mapping population. QTL analysis revealed one significant QTL in LG10 on the female linkage map. The genetic linkage map—the second such map generated for Pacific bluefin tuna—and the growth‐related QTLs detected in this study will be useful for tuna aquaculture MAS programs.  相似文献

Pacific oysters, Crassostrea gigas, are the most abundantly harvested shellfish in the world and are ecologically significant. The content of nutrients, including protein, glycogen, lipid, zinc (Zn), and selenium (Se), is important for oyster meat quality, but heritability estimates of such traits have rarely been reported. In this study, 64 full‐sib families were generated using a nested mating design. Finally, 18 full‐sib families, of which there were nine half‐sib families, with each containing 2 full‐sib families, were sampled for heritability estimates. The narrow‐sense heritabilities of glycogen, protein, lipid, Zn, and Se contents were 0.29 ± 0.02, 0.38 ± 0.02, 0.58 ± 0.08, 0.02 ± 0.02, and 0, respectively. Negative genetic correlations existed between both glycogen and protein content (?0.95 ± 0.004) and between lipid and protein content (?0.59 ± 0.05), whereas a positive correlation was observed between lipid and glycogen content (0.16 ± 0.06). Weak genetic and phenotypical correlations (r = 0–0.2) were observed between shell height and nutritional quality traits. These data demonstrated that glycogen, protein, and lipid content can be chosen in a selective breeding program, but glycogen and lipids cannot be selected together with protein. Furthermore, performing indirect selective breeding for quality traits by selecting traits related to growth is impossible. This study provides information for the development of breeding strategies for oyster quality traits.  相似文献

采用牙鲆日本群体和韩国群体杂交的92个F1个体作为分离群体, 利用微卫星标记和Joinmap 4.0作图软件构建了牙鲆遗传连锁图谱。共有221个SSR标记用于连锁图谱构建, 雌性图谱中, 共178个微卫星标记定位到22个连锁群上, 观测总长度为(G oa )599.0 cM, 覆盖率(C oa )达76.27%。雄性图谱中, 共194个微卫星标记定位到23个连锁群上, G oa 为693.4 cM, C oa 为78.82%。对全长、体质量、体高3组数据进行主成分分析处理, 得到可解释3个性状的89.6%特征的一组数据, 命名为牙鲆生长性状GT。用WinQTLCart 2.5软件的复合区间作图,在已构建的遗传连锁图谱上对牙鲆生长性状GT进行QTL定位, 取LOD经验值2.5为QTL存在的阈值; 对微卫星标记进行性状—标记之间的回归分析。本研究共定位3个与牙鲆生长性状GT相关的QTLs, qGT-f4 qGT-m20 qGT-f20,可解释表型变异率分别为27.60%, 13.74%, 10.27%。在性状—标记之间的回归分析中, 得到22个与生长性状GT相关(P<0.05)的微卫星标记, 单个标记可解释表型变异率介于3.70%~10.42%, 其中6个微卫星标记scaffold558_51720、scaffold558_26183、scaffold903_69232、scaffold485_47120 、scaffold1262_77386、scaffold809_65154与生长性状GT之间呈极显著相关(P><0.01), 可解释表型变异率分别为10.42%、7.31%、10.07%、10.07%、8.39%和11.26%。  相似文献

Growth rate is one of the important economic traits in aquaculture species that is influenced by quantitative trait loci (QTLs). Recently published complete genetic map of common carp (Cyprinus carpio) with 307 markers (including 109 SSR, 31 EST-SSR and 167 SNP markers) was used to determine the number and position of QTLs effecting body weight (BW) and growth rate. Ten-, 11- and 12-month-old individuals of common carp were measured, and seven QTLs were detected in three linkage groups (LG1, LG6 and LG20); three were significant QTLs (qBW2-6, qBW3-6 and qBW3-20, P < 0.001), and four were suggestive QTLs (qBW1-1, qBW1-6, qBW2-20 and qBW3-1, P < 0.01). The three significant QTLs related to BW2 and BW3 were in LG6 and LG20 and covered 18 and 41 cM, respectively. The explained variances of qBW2-6, qBW3-6 and qBW3-20 were 34.4, 32.1 and 33.4 %, respectively. The four suggestive QTLs affecting BW1, BW2 and BW3 were in LG1, LG6 and LG20 and had variances that ranged from 17.9 to 26.7 %. The similarity of the QTL intervals for BW1, BW2 and BW3 in LG6 suggested that these genes may affect growth continuously during the early stage of growth and may be important for growth. The identification of QTLs for growth rate and BW in C. carpio will lead to a better understanding of the factors that influence these traits at a genetic level and will promote the growth to higher production through marker-assisted breeding.  相似文献

The trace element contents of Corbicula clam shells collected from Japan, Russia, China, and the Republic of Korea were analyzed to determine their geographic origin. The crushed shells were decomposed with nitric acid–hydrogen peroxide, and the concentrations of 14 elements (Li, Mg, V, Mn, Co, As, Rb, Mo, Ba, Ce, Pb, U, Sr, and Ca) were measured by inductively coupled plasma mass spectrometry and inductively coupled plasma optical emission spectrometry. Some of the elements identified in samples displayed a geographic trend. The average content of manganese in Japanese samples was twice that of Russian samples. Conversely, the arsenic content in Japanese samples was approximately half of that in Russian samples. Linear discriminant analysis was applied to the data from Japanese and Russian samples, and a discriminant model was constructed. The discriminant model was used to determine the geographic origin of Corbicula clams produced in Japan, with 89.8 % of those identified as Japanese and 92.2 % of those identified as Russian being classified correctly. Therefore, trace element analysis of the shells of Corbicula clams is a useful technique for the identification of their country of origin.  相似文献

Corticotropin-releasing hormone (CRH) is a potent mediator of endocrine, autonomic, behavioral, and immune responses to stress. For a better understanding of the structure and function of the CRH gene and to study its effect on feeding regulation in cyprinid fish, the cDNA of the CRH gene from the brain of Schizothorax prenanti was cloned and sequenced. The full-length CRH cDNA consisted of 1,046 bp with an open reading frame of 489 bp encoding a protein of 162 amino acids. Real-time quantitative PCR analyses revealed that CRH was widely expressed in central and peripheral tissues. In particular, high expression level of CRH was detected in brain. Furthermore, CRH mRNA expression was examined in different brain regions, especially high in hypothalamus. In addition, there was no significant change in CRH mRNA expression in fed group compared with the fasted group in the S. prenanti hypothalamus during short-term fasting. However, CRH gene expression presented significant decrease in the hypothalamus in fasted group compared with the fed group (P < 0.05) on day 7; thereafter, re-feeding could lead to a significant increase in CRH mRNA expression in fasted group on day 9. The results suggest that the CRH may play a critical role in feeding regulation in S. prenanti.  相似文献

壳白长牡蛎品系生长和壳色性状遗传参数估计   总被引:2,自引:2,他引:0  
以经过连续4代家系选育获得的壳白长牡蛎(Crassostrea gigas)选育系为亲本,通过巢氏平衡设计建立了30个全同胞家系混合养殖,采用微卫星多重PCR技术进行家系鉴定,基于REML法估算24月龄壳白长牡蛎的生长性状和壳色性状的遗传参数。结果表明,壳白长牡蛎品系壳高、壳长、总重、壳重、L~*(明度)、a~*(红绿轴色品指数)和b~*(黄蓝轴色品指数)的遗传力为中高等水平,依次为0.35±0.13、0.18±0.09、0.20±0.09、0.16±0.08、0.16±0.08、0.27±0.11和0.19±0.08,壳宽、肉重、出肉率、壳型指数A和壳型指数B的遗传力为低等水平,依次是0.07±0.02、0.11±0.06、0.02±0.03、0.08±0.06和0.11±0.06。壳高、壳长、壳宽、总重、壳重和肉重之间的遗传相关和表型相关均为正相关,其中,壳高、壳宽和总重与其他生长性状的相关性较高,分别为0.40±0.65~0.90±0.14、0.39±0.55~0.97±0.24和0.50±0.66~0.99±0.02。壳型指数A和壳型指数B与壳高均为较高的负相关,分别为–0.94±0.16和–0.77±0.19,表明仅以壳高性状为选育目标时,可能不会对长牡蛎壳型改良产生作用。壳白长牡蛎壳色参数与生长性状之间的遗传相关范围为–0.09±0.42~0.91±0.74,不同性状间的遗传相关差异很大,其中L~*与生长参数遗传相关较高,为0.49±0.29~0.91±0.74,表明以壳高、壳长、总重和L~*任一个为选育目标时,其他生长性状都可以获得提高。壳色参数间L~*与a~*负的相关性最高,为–0.96±0.04,L~*与b~*和a~*与b~*相关性较低,分别为–0.08±0.36和0.21±0.31,表明以L~*为选育目标时,可间接降低a~*值。本研究为合理制定壳白长牡蛎新品系育种方案和选择反应预测提供了参考依据。  相似文献

To evaluate the impact of mass selection on genetic structure in artificially closed populations of the Pacific oyster Crassostrea gigas, we performed mass selection over six generations on two stocks from Japan and Korea and analyzed their temporal genetic variation and structure using 18 microsatellite makers, which were compared with the base populations of the two selected lines and one wild population from China. The average numbers of alleles (Na), mean observed heterozygosities (Ho), and expected heterozygosities (He) varied over generations in the two selected lines (selected lines of Japan, Na = 10.7–14.9, Ho = 0.757–0.846, He = 0.778–0.871; selected lines of Korea, Na = 9.4–17.3, Ho = 0.736–0.865, He = 0.744–0.854). There was no significant reduction in heterozygosity in the two selected lines. However, the average number of alleles per locus was significantly lower in the fifth and sixth generations of the two selected lines compared with that in the base population and wild population (P < 0.05), suggesting that the successive mass selection in closed populations may increase the sensibility of rare alleles to genetic drift. Equalizing the sex ratio of parents and reducing the selection intensity properly with the increase of selective generations is recommended to minimize the deleterious effect of genetic drift and bottleneck caused by successive mass selection. The information obtained in this study is useful for the design of appropriate management strategies for selective breeding of C. gigas.  相似文献

The effect of taurine supplementation to a microencapsulated diet at 0 % (T-0 %), 1 % (T-1 %) and 2 % (T-2 %) level on growth performance and taurine content of Nibea albiflora larvae was evaluated. The microencapsulated diet was prepared using wet granulation and fluidized bed coating process. Scanning electron microscopy microphotography of the microencapsulated diet showed the appearance with a dense film around the core. More than 50 % of diet particles were between 250 and 590 μm. A 30-day feeding experiment was conducted with larvae at 15 days after hatching (DAH), in which larvae were weaned at 20 DAH from copepods to the experimental diets or fed copepods throughout the experimental period. The survival and total length of larvae were significantly higher in T-1 % group compared with T-0 % group (P < 0.05). The survival, total length and wet weight of larvae were significantly higher in T-2 % group compared with T-0 % group (P < 0.05). Fish larvae fed copepods had the best growth performance in survival, total length and wet weight, which were significantly higher than those in other treatments (P < 0.05). The taurine content of larvae fed with supplemental taurine (T-2 %, T-1 %) was significantly higher than that of larvae fed without supplemental taurine (T-0 %) (P < 0.05). These results confirmed that taurine is an essential nutrient for N. albiflora larvae.  相似文献

黑色素是一种广泛存在于动物界的生物色素,而酪氨酸酶被认为是一种重要的调控黑色素合成的关键酶。实验以3种壳色的长牡蛎选育品系为材料,使用PCR-SSCP的方法对长牡蛎酪氨酸酶基因CgTyr1进行SNP分型筛选,将突变位点与不同壳色性状进行关联分析。结果显示,酪氨酸酶基因的外显子上存在23个SNP位点,其中11个SNP位点与壳色性状极显著相关;在这11个S N P位点中,检测到有3个S N P位点为有义突变(c.591C/T、c.632G/A和c.1155T/C),分别导致不同的氨基酸突变(Ala122Val、Gly136Ser和Phe310Ser);利用极显著关联的11个SNP位点,为每种壳色群体建立了1种单倍型,并在验证组中得到了确认。研究表明,长牡蛎酪氨酸酶基因的单个碱基突变和以此构建的单倍型与壳色性状存在显著的关联。本研究筛选出的SNP位点和构建的单倍型为长牡蛎壳色品系选育提供了重要的参考资料。  相似文献

Corticotropin-releasing hormone (CRH), urotensin I (UI) and urotensin II (UII) are found throughout vertebrate species from fish to human. To further understand the role of crh, uI and uII in teleosts during development, we investigated the expression pattern of crh, uI, uIIα and uIIβ genes, and their response to acute salinity and temperature challenge during early development of zebrafish, Danio rerio. The results reveal that crh, uI, uIIα and uIIβ mRNA are detected from 0hpf, and the expression levels increase to a maximum at 6 days post fertilization (dpf), with the exception of uIIα that peak at 5dpf. Exposure of zebrafish embryos and larvae to acute osmotic (30ppt) stress for 15 min failed to modify expression levels of crh, uI, uIIα and uIIβ mRNA from levels in control fish except at 6dpf when uIIα and uIIβ were significantly (P < 0.05) modified. Exposure of embryos and larvae to a cold (18 °C) or hot stress (38 °C) generally down-regulated mRNA levels of crh, uI, uIIα and uIIβ apart from at 3dpf. The results indicate that the contribution of crh, uI, uIIα and uIIβ genes to the stress response in zebrafish may be stressor-specific during early development. Overall, the results from this study provide a basis for further research into the developmental and stressor-specific function of crh, uI, uIIα and uIIβ in zebrafish.  相似文献

The F1 progeny from parents selected randomly for differences in performance traits from different families of mirror carp were used as a mapping panel. Linkage maps constructed using Join Map 4.0 software with 992 microsatellite polymorphic markers ultimately yielded 51 groups. Composite interval mapping (CIM) of superoxide dismutase (SOD) trait of three tissues (liver, anterior and posterior intestine) was conducted for quantitative trait loci (QTL) location using Map QTL 6.0 software. Results shows that seven QTLs significantly (P < 0.05) explained 8.2–18.5% of the phenotypic variations in liver, five QTLs explained 7.9–11.1% of the variation in anterior intestine (P < 0.05) and six QTLs were located and explained 6.1–15.8% of the phenotypic variations in posterior intestine (P < 0.05). NCBI BLASTn was used for homology analysis between the sequences from markers used in the located QTL regions and sequences from the zebrafish database, and many protein relating with the SOD expression were blasted (similarity >85%). These results showed that it is highly probable that these QTLs are the main regions of SOD expression. The data also support the value of forward genetic studies on organism development and serve as a basis for selection.  相似文献

以镜鲤(Cyprinus carpio L.)全同胞家系为材料,用940对微卫星(SSR)标记进行基因组扫描,采用半同胞家系的分析策略对体质量(BW)和体长(SL)进行QTL定位分析。QTL检测显示:基于父系的QTL分析,共检测到8个QTL区间。5个与体质量相关的QTL区间中,1个QTL为95%基因组水平(genome-wide)显著性,位于LG26连锁群,其他4个QTL均为95%染色体水平(chromosome-wide)显著性;3个与体长相关的QTL区间与体质量的QTL区间重叠,其中,1个QTL为99%基因组水平显著性,其余2个QTL均为95%染色体水平显著性。基于母系的QTL分析,共检测到11个QTL区间。6个与体质量相关的QTL区间中,1个QTL为99%基因组水平显著性,位于LG26,2个QTL为99%染色体水平显著性,其余3个QTL均为95%染色体水平显著性;5个与体长相关的QTL中有4个与体质量QTL区间重叠,其中,1个QTL为99%基因组水平显著性,1个QTL为99%染色体水平显著性,其余3个QTL均为95%显著性染色体水平。结果表明,在LG26上,存在着与体质量和体长都显著相关的QTL区间,且均达到基因组显著性水平,最小置信区间为3 cM。此QTL结果可以应用于鲤鱼分子标记辅助育种。  相似文献
