Agreement of SpO2, SaO2 and ScO2 in anesthetized cynomolgus monkeys (Macaca fascicularis)

Objective To assess the agreement between three measurements of arterial oxygen saturation (SpO₂, SaO₂ and ScO₂) in anesthetized cynomolgus monkeys. Study Design Prospective study. Animals Eleven mature, male cynomolgus monkeys (Macaca fasicularis). Methods Monkeys were anesthetized with intramuscular ketamine followed by intravenous propofol. The trachea of each was intubated and the lungs ventilated. Arterial oxygen saturation was measured with a Nonin 8500 V pulse oximeter, using a lingual clip on the cheek. Arterial blood samples were taken from an indwelling catheter. Inspired oxygen concentration was varied from 12 to 20%, and 88 paired arterial blood samples and saturation measurements were taken. Arterial oxygen saturation in the blood samples was measured using a cooximeter. The saturation was also calculated from the arterial oxygen tension using the Adair equation. The results were compared using Bland and Altman's method. Results The pulse oximeter readings were 2.7% higher than that of the cooximeter, with a limit of agreement of ?3.9 to 9.3%. The pulse oximeter readings were 1.8% higher than the calculated saturation, with a limit of agreement of ?6.5% to 10.1%. The cooximeter readings were 0.9% lower than the calculated saturation, with a limit of agreement of ?5.6% to 3.8%. Conclusions The agreement between SpO₂ and other measurements of arterial oxygen saturation in this study is typical for this technique. The bias and limits of agreement are consistent with reports in other species. Clinical relevance The Nonin 8500 V is a useful pulse oximeter for clinical use in primates.

猪2型圆环病毒Cap蛋白在伪狂犬病病毒中的表达

用EcoR Ⅰ酶切伪狂犬病病毒疫苗株Tk-/gE-/LacZ 基因组,与含有猪2型圆环病毒(PCV2)ORF2基因的重组转移载体pIEORF2共转染IBRS-2细胞.收集病变细胞,经空斑纯化和PCR鉴定,获得了新的重组病毒Tk-/gE-/ORF2 .用Southern blot杂交证明该重组病毒构建正确,用间接免疫荧光法和Western blot证日月该重组病毒可表达PCV2的主要免疫原性蛋白Cap蛋白.重组病毒在IBRS-2细胞上连续传15代后,遗传稳定.该重组病毒在IBRS-2细胞上增殖滴度为10-7.1 TCID50/0.1mL,在鸡胚成纤维细胞上增殖滴度为10-4.8 TCID50/0.1mL,在Marc-145细胞上增殖滴度为10-6.8 TCID50/0.1mL.     

猪流感病毒（A／swine／Jiangsu／2／2006（H3N2）NS1基因表达产物分析

采用RT—PCR方法克隆猪流感病毒H3N2亚型NS1全长基因,构建NS1基因原核表达质粒pET-28a—NS1和真核表达质粒p3XFLAG—CMV—NS1,在大肠杆菌和真核细胞内表达NS1基因,制备抗NS1多克隆抗体。用所制备的抗体分析p3xFLAG—CMV—NS1转染表达NS1蛋白和病毒感染细胞内的NS1蛋白。经终浓度为1mmol／L的IPTG诱导后,重组蛋白NS1在大肠杆菌中得到表达。表达蛋白纯化后免疫Wistar大鼠制备抗NS1蛋白多克隆抗体,Western—blot分析表明抗NS1多克隆抗体可以识别大肠杆菌表达的NS1蛋白、转染Veio细胞表达的NS1蛋白和病毒感染细胞内的NS1蛋白。间接免疫荧光发现NS1蛋白主要定位于细胞核。结果显示,本试验成功构建了NS1基因原核和真核表达系统,获得了NS1特异性多克隆抗体,分析了NS1细胞定位,为进一步研究NS1蛋白在病毒复制过程中的生物学功能和猪流感病毒的复制机理奠定基础。

H1N1亚型猪流感病毒A/Swine/GD/2/12基因特征分析

2012年从广东省某猪场的疑似流感猪群采集鼻拭子样品,接种鸡胚并收集尿囊液,通过血凝、血凝抑制和RT-PCR,鉴定出1株H1N1亚型猪流感病毒,命名为A/Swine/GD/2/12。RT-PCR扩增得到全基因8个片段,与GenBank收录的参考毒株比对并构建进化树,发现本毒株可能是H1N1亚型重组株,其8个片段与我国猪源和北美地区1985—1992年间的猪源、禽源(A/turkey/IA/1992)和人源(A/Maryland/12/1991)流感毒株在同一个大分支上,其中与我国猪源参考株同源性在95.8%以上,与北美地区的H1N1参考株同源性在94%以上。HA受体位点分析表明,本毒株既具备结合Saα2,6Gal型人类流感病毒SA受体的特点,也有结合Saα2,3Gal型禽类流感病毒SA受体的可能。提示本毒株可能是由北美地区猪源、禽源和人源的H1N1亚型流感病毒重排形成的。HA蛋白裂解位点分析、NS和PB2蛋白位点分析表明,本毒株具备低致病性毒株的特点。小鼠致病性试验进一步证实本毒株能够引起小鼠运动减少、食欲欠佳、体重减轻等表现,但不会引起咳嗽和死亡等严重反应。     

GI PiCO2: Tissue Specific Monitoring For Improving Patient Outcomes

Improvements in human patient monitoring despite their development in animals, do not always find their way into veterinary clinical use due to financial constraints. Gastrointestinal intraluminal CO₂ partial pressure (Gip₁CO₂) monitoring, however, is not only proving very beneficial in human trauma and critical patient care but is also very likely to become relatively inexpensive. By providing information on the perfusion adequacy of a high risk, critically important tissue, the GI mucosa, GI P₁CO₂ monitoring offers an easily accesible indicator of the efficacy and adequacy of resuscitative interventions. The potential for decreasing morbidity and mortality is enormous. Therefore, the practicing veterinarian should become familiar with GI P₁CO₂ monitoring theory and technology so he or she can be better prepared to incorporate it into practice when in becomes available.     

Low expression of cyclin D2 in G2/M-arrested and transformed proliferating Balb/3T3 cells

At present, the effect of cyclin D2 implicated in cell cycle regulation, differentiation, and oncogenic transformation is not fully confirmed. To better elucidate the role of cyclin D2 in controling the cell proliferation, cyclin D2 expression level was determined at the early initiation and promotion stages during the in vitro two-stage transformation process of Balb/3T3 A31 cells. N-methyl-N'-nitro-N-nitrosoguanidine (MNNG)-induced G2/M-arrested cells expressed low level of cyclin D2 mRNA, while the contact-inhibited nonproliferating cells expressed high level of cyclin D2 mRNA. In the transformed proliferating cells at the promotion stage, cyclin D2 mRNA was not expressed. These data suggest that cyclin D2 expression may be associated with the type of growth arrest and nonproliferating state, but not with the cell proliferation and transformation.
