Formalin-inactivated Salmonella enteritidis phage type 4 strain 119/95 (SE) was encapsulated in biodegradable poly (DL-lactide co-glycolic acid) PLGA; (65:35) microspheres by a modified water-in-oil-in-water (w/o/w) double-emulsion solvent extraction/evaporation technique. These SE-loaded microspheres (SE-MS) were porous and spherical in shape with diameters of 0.4-10 microm and 20-80 microm in two preparations. SE-MS were subsequently used to vaccinate specific-pathogen-free chickens in a single dose in order to investigate the potency of a single-dose vaccination in inducing immune responses and protective immunity. In Experiment 1, 4-wk-old chickens that were vaccinated intramuscularly with 20-80-microm SE-MS generated long lasting (over 6 mo) and persistently high serum anti-SE immunoglobulin (Ig)G antibody response. In Experiment 2, 2-wk-old chickens were vaccinated orally with 0.4-10-microm or intramuscularly with 20-80-microm SE-MS and challenged with 10(9) colony-forming units of homologous SE strain at 6 wk postvaccination. When challenged intramuscularly, one each of the orally vaccinated (n = 10) and the intramuscularly vaccinated birds (n = 10) showed clinical signs and death, whereas all of the nonvaccinated control birds (n = 12) were sick and 11 of them were killed. When challenge was via oral route, 26.1% of cloacal swabs and 24.0% of organs (liver, spleen, and cecum) collected from orally vaccinated birds (n = 35) were positive for SE, comparable to 27.9% of feces and 18.7% of organs from the intramuscularly vaccinated birds (n = 35). These figures were significantly lower than those for nonvaccinated birds (n = 30) from which 59.3% of feces and 44.0% of organs tested SE positive (P < 0.05). The humoral immune response was also determined after vaccination with a single dose. The intramuscular vaccination elicited higher serum IgG response than oral administration, but the latter elicited a significant intestinal mucosal IgA antibody response. This is the first evidence that chickens vaccinated with killed SE-loaded PLGA microspheres, intramuscularly and orally in a single dose, developed systematic and local immune responses, thereby conferring protective immunity.  相似文献

Lymphocyte proliferation and interleukin (IL)-2 and IL-6 levels in serum were measured as indicators of cell-mediated immunity after immunization of chickens with a commercial killed Salmonella enteritidis (SE) vaccine or experimental subunit vaccines of crude protein (CP) extract or the outer membrane protein (OMP). Significantly increased proliferative responses to SE flagella, but not lipopolysaccharide, porin, CP, or OMP, were observed at 1 wk postimmunizarion in the three vaccination groups. The responses to flagella were specific because flagella-induced proliferation was not seen in chickens immunized with adjuvant alone. Of the three immunization protocols, use of the killed SE vaccine appeared most effective because it induced higher flagella-stimulated lymphocyte proliferation at 1 and 2 wk postvaccination compared with the CP- and OMP-vaccinated groups. Significantly increased IL-2 and IL-6 levels in serum were seen at 1 wk postimmunization in the three vaccination groups compared with adjuvant alone, but there were no differences between the killed vaccine and the subunit vaccines at this time, and the levels of both lymphokines returned to baseline at 2 wk postimmunization. We conclude that cell-mediated immunity to SE after vaccination with the killed bacterial vaccine or subunit vaccines is transient and mainly limited to flagella.  相似文献

Serum antibody responses and haemolytic complement activity were evaluated in White Leghorn (WLH) and Rhode Island Red (RIR) chickens that were vaccinated with live-attenuated vaccines of Newcastle disease virus, or infectious bronchitis virus, or infectious bursal disease virus by means of ocular challenge at 10 times the normal vaccination dose. Complement titres in non-vaccinated birds were significantly higher in WLH birds compared to RIR birds. The lentogenic viral infection resulted in an immediate stimulation of complement activity, followed by a decrease to initial complement levels within 2 weeks post vaccination, when the antibody response took over immune defence. As compared to WLH chickens, RIR birds mounted a faster and significantly higher antibody response to the vaccine viruses used. In WLH hens, significantly higher haemolytic complement activity post vaccination was found as compared to RIR hens. Possible consequences of the observed differences in immune responsiveness of the two breeds to viral vaccines are discussed.  相似文献

An experiment was carried out to study the effects of vaccination with Paracox, a live, attenuated vaccine against avian coccidiosis, on broilers isolated from extraneous Eimeria parasites. The study involved 3200 broiler chickens raised in floor pens similar to commercial conditions, but in an initially Eimeria-free environment. Forty percent of the chickens were vaccinated at 3 days of age and given either a basal unmedicated feed or a feed supplemented with the feed antibiotic virginiamycin. Unvaccinated birds were given either the basal feed or feed supplemented either with virginiamycin or the anticoccidial ionophore narasin. At slaughter at 36 days of age vaccinated birds had a lower live weight than non-vaccinated birds. The difference was 4.6% in unmedicated, and 6.0% in virginiamycin medicated chickens. Feed conversion ratio at slaughter was 2.5% higher for unmedicated vaccinated birds, and 1.3% higher for virginiamycin medicated vaccinated birds, compared to respective non-vaccinated groups. There was no significant difference in overall performance of unvaccinated birds given narasin as compared to virginiamycin. At 10 days post vaccination vaccinated birds had a higher number of Clostridium perfringens in the caeca, but there was no difference thereafter. Throughout the experiment, caecal clostridial counts were considerably higher in vaccinated unmedicated birds than in unvaccinated birds given narasin. The number of oocysts shed in the vaccinated groups was very low, but during a subsequent challenge with E. maxima and E. tenella the birds' immunity was found to be satisfactory.  相似文献

A number of studies have demonstrated significant protection of cattle against bovine tuberculosis following vaccination with the Pasteur strain of Mycobacterium bovis bacille Calmete-Guerin (BCG). However, it is unclear if other daughter strains of BCG are as effective, which is an important issue to resolve for a variety of regulatory compliance issues. This study compared the protective immune responses to bovine tuberculosis induced in cattle vaccinated with BCG Danish with those induced by BCG Pasteur. Groups of calves (n=10) were vaccinated with 10(6) colony forming units (CFU) BCG Pasteur prepared from a fresh liquid culture, 10(6) CFU BCG Danish prepared from a fresh liquid culture or 0.4 mg of reconstituted freeze-dried culture of BCG Danish. Another group (n=10) served as non-vaccinated controls. BCG Pasteur induced significantly higher and more sustained levels of bovine purified protein derivative (PPD)-specific gamma interferon (IFN-gamma) in whole-blood cultures following vaccination compared to either fresh culture BCG Danish or freeze-dried BCG Danish. Vaccination with a fresh culture of BCG Pasteur, fresh culture BCG Danish and freeze-dried BCG Danish gave a significant enhancement in three, four and three pathological and microbiological parameters of protection, respectively, compared to the non-vaccinated group. These results demonstrate the Danish strain of BCG is a viable alternative to BCG Pasteur for vaccination of cattle as both strains had similar efficacy and there was little difference between freshly cultured and freeze-dried formulation of BCG Danish. The results also show that post-vaccination antigen-specific IFN-gamma levels in whole blood is not always a reliable indicator of protection against a subsequent virulent challenge.  相似文献

Two experiments were conducted to evaluate the virulence and vaccination efficacy of a Mycoplasma gallisepticum (MG) isolate designated MG Intervet 6/85. Virulence of the strain was determined by evaluation of airsacculitis scores following aerosol exposure to the isolate before and after 10 sequential passes in either commercial broiler chickens or commercial turkeys. Two-week-old specific-pathogen-free chickens were vaccinated by aerosol exposure. The birds were challenged with the R' strain of MG at either 4 or 8 weeks post-vaccination. Efficacy was evaluated by airsacculitis scores determined 21 days after challenge. Ten repetitive back-passes of the isolate in chickens and turkeys did not substantially increase the virulence. Virulence for both chickens and turkeys was minimal, while protection elicited by aerosol vaccination in young chickens against virulent R' strain was significant (P less than or equal to 0.05) compared with unvaccinated controls.  相似文献

Immunization against Angara disease virus (ADV), a serotype 4 avian adenovirus, and Newcastle disease virus (NDV), an avian paramyxovirus serotype 1, is the mainstay of a broiler vaccination programme, while polyether ionophores usually form an essential component of a broiler medication programme in most parts of India and Pakistan. The role of polyether ionophores in the protective immune responses of broiler chickens vaccinated and challenged with ADV and NDV was investigated. A total of 1600 birds were divided into eight groups of 200 birds each. First four groups were vaccinated against NDV and ADV, while the remaining four served as unvaccinated controls. The first 3 groups of birds were administered salinomycin, monensin and cyclophosphamide (CYP), respectively. The last group served as an untreated control. The same treatment schedule was also followed for the next four unvaccinated groups. The post-vaccination and post-challenge serological responses to NDV and ADV, body and lymphoid organ weight gains, post-challenge survival rate and detection of NDV and ADV in the tissues of infected birds were evaluated. Birds administered salinomycin showed a significant stimulation of protective immune responses against both NDV and ADV as compared to the untreated and CYP-treated birds. Monensin also enhanced the protective immune responses against both viruses but the effect was not statistically significant. Thus, it is concluded that monensin and salinomycin augment the anti-NDV and anti-ADV immune responses in broiler chickens, which supports their use in poultry flocks.  相似文献

The impact of live and killed Salmonella vaccines on cell-mediated immunity (CMI) was investigated in 18- and 32-week-old White Leghorn chickens, by assessing splenic lymphocyte proliferation, expression of IL-2 mRNA in concanavalin A (Con A) stimulated cells and flow cytometric analysis of cell subpopulations. Con A and Salmonella enteritidis (SE) flagella induced proliferation of splenocytes were enhanced in the 18- and 32-week-old chickens treated with live vaccine, compared to the corresponding control chickens. Among the killed vaccine treated birds, Con A-mediated response was higher in the 18-week-old chickens compared to the corresponding control birds. Increased proliferation was accompanied by increased CD4 and reduced CD8 and gammadelta T-lymphocytes in the 18-week-old live vaccine treated chickens. Relative expression of IL-2 mRNA in Con A-stimulated splenocytes from 18-week-old birds was not affected by vaccine treatment. Overall, live vaccine was more effective in increasing the lymphocyte proliferation to Con A as well as SE antigen. This enhanced CMI may prove beneficial in protecting chickens against SE infection.  相似文献

Broiler chickens, in groups of 10, received a single vaccination by the stick-wing route at 1, 2, 3, 4, 5, 6, or 11 weeks of age with live Clemson University strain of Pasteurella multocida. Twenty non-vaccinates kept in isolation served as controls. Cholera serum antibody titers in all chickens were determined by enzyme-linked immunosorbent assay at weekly intervals. Chickens vaccinated once at 1, 2, 3, 4, 5, and 6 weeks, respectively, attained 25.2%, 28.7%, 34.7%, 46.2%, 51.8%, and 64.6% of the titers of those vaccinated once at 11 weeks of age. Variation in antibody response was greatest in chickens vaccinated at 1 or 2 weeks of age. Additionally, chickens vaccinated at 1 or 2 weeks of age showed the longest response time (5 weeks) to reach maximum antibody titers after a single vaccination. When the original vaccinates were revaccinated at 11 weeks of age, all showed a secondary response equal to or greater than that seen in chickens vaccinated once at 11 weeks of age. Age of the chickens at the time of vaccination and antibody titer were positively correlated (r = 0.997). Overall antibody responses to vaccination were higher and much more uniform as birds increased in age.  相似文献

应用间接ELISA法,检测了传染性法氏囊病疫苗免疫雏鸡4种局部体液(泪液、气管液、胆汁、肠液)免疫球蛋白IgG、IgM、IgA含量的动态变化。结果发现免疫雏鸡局部体液3种免疫球蛋白含量较未免疫对照雏鸡均有不同程度地升高;IBD超强毒株攻击后,对照雏鸡上述指标均明显低于疫苗免疫雏鸡,发生典型IBD,并全部死亡,而疫苗免疫雏鸡免疫保护率达60%,表明疫苗免疫后,可增强雏鸡的局部体液免疫功能。  相似文献

Prevalence studies have shown that almost 100% of free-range chickens are infected with a wide range of parasites. The infections are mostly subclinical in nature, resulting in production losses and occasionally mortality. Newcastle disease (ND), on the other hand, results in high mortality rates during epidemics. ND is a limiting factor for increasing poultry production in many tropical countries, where frequent reports indicate vaccination failures. The aim of our study was to investigate the influence of helminths on the antibody response after vaccination against Newcastle disease of free-range chickens naturally infected with parasites. Sixty chickens were divided into six groups, of which three were vaccinated against ND with a live De Soto vaccine, while the other three remained non-vaccinated. One group within the vaccinated groups and the one within the non-vaccinated group was kept naturally infected with helminth parasites, while the other two groups in each set were dewormed with fenbendazole and niclosamide, and one of each of these groups was subsequently infected with Ascaridia galli. After vaccination, all the groups were followed for 5 weeks and their antibody titres were determined weekly using a HI test. All the birds were finally challenged 4 weeks after vaccination with a virulent velogenic ND virus obtained from a field outbreak. All the vaccinated chickens seroconverted and had high antibody levels after 3 weeks, but these dropped to low levels at 4 weeks after vaccination. After challenge, the antibody titres rose in the dewormed groups but not in the parasite-infected groups. After 5 weeks, all the parasite-infected animals had significantly lower antibody titres than the dewormed animals. All the vaccinated chickens survived the challenge infection, emphasizing the importance of the cellular immune response. Further studies are needed to examine the effects of the parasitic infection on protection against ND over a longer period.  相似文献

为研究分析重组大肠杆菌不耐热肠毒素(labile enterotoxin,LT)作为粘膜佐剂,配伍鸡新城疫疫苗的冻干样品对免疫鸡非特异性免疫应答的影响,在前期已经构建的LTR72/G192基础上,选择2、4μg LTRG蛋白作为每羽份疫苗添加量,及降低新城疫病毒(Newcastle disease virus,NDV)50%抗原量等配伍条件。按照生产条件制备冻干疫苗,进行雏鸡免疫试验,免疫后24、48、72 h采集脾脏、胸腺淋巴结,检测几种非特异性细胞因子表达水平。结果表明:IL-6、INF-γ水平在各疫苗免疫后24h即升高,48 h达最高值;添加2、4μg LTRG蛋白的NDV疫苗组IFN-β、TNF-α水平在48 h左右显著升高,其中含4μg LTRG疫苗组与其他组差异具有极显著统计学意义(P〈0.01);IL-6、IFN-γ转录水平有所增强,但各组间差异不具有显著统计学意义(P〉0.05),72 h左右几种细胞因子水平均迅速下降,与NDV疫苗接种组相近。由此可见,LTRG能辅助抗原诱导免疫雏鸡的非特异性免疫应答,有利于对感染病毒的清除。  相似文献

The present study was designed to assess and compare three different formulations of the new Onderstepoort infectious coryza (IC) quadrivalent vaccine, which contain an NAD-independent strain of Avibacterium paragallinarum (previously known as Haemophilus paragallinarum), and a commercial IC vaccine, not containing an NAD-independent strain, for their safety and ability to protect chickens of varying ages against virulent challenges with four different serovars of A. paragallinarum, including the NAD-independent strain of the C-3 serovar. Four groups of 140 chickens each were vaccinated at the age of 17 weeks and revaccinated at the age of 19 weeks with each of the four vaccine formulations. A similar sized group of non-vaccinated chickens was used as control. Two rounds of challenge were conducted: a group of chicken in each vaccination group was challenged between 31 and 35 weeks of age, while another group was challenged between 51 and 55 weeks of age. The "in-contact" challenge model was used in this experiment. For each vaccination group, the four challenge strains representing four local serovars were used in each challenge round. The efficacy of the vaccines was compared based on overall protection levels obtained and the duration of protection. The safety of the different vaccines was determined by the severity of post-vaccination reactions. The need for the incorporation of the NAD-independent strain in the vaccine was evidenced by the low protection level against NAD-independent challenge recorded in the group of birds vaccinated with the commercial vaccine. The results obtained confirmed not only the variation in virulence of different South African serovars, with serovar C-3 being the most virulent and serovar B having almost no virulence but also the age related increase in susceptibility. The importance of a suitable formulation of the vaccine is discussed.  相似文献

Serpulina hyodysenteriae infection of pigs, swine dysentery, causes a mucohemorrhagic diarrhoea resulting in significant economic losses to swine producers. The pathogenesis of this disease is poorly understood. Regardless, commercial vaccines have been developed and are in use. Thus, the present study was designed to examine cellular immune responses induced by parenteral S. hyodysenteriae vaccination. Significant antigen-specific interferon-gamma (IFN-gamma) and blastogenic responses were detected from peripheral blood lymphocytes isolated from vaccinated pigs. However, poor IFN-gamma responses were detected from colonic lymph node lymphocytes from these same pigs despite significant antigen-specific blastogenic responses. In addition, peripheral blood IFN-gamma responses were diminished by either in vitro depletion of CD4 expressing cells or by in vitro treatment with porcine IL-10. Colonic lymph node IFN-gamma responses were not inhibited by treatment with porcine IL-10. Vaccination also resulted in increased percentages of both mucosal and peripheral blood CD8 single positive cells with concurrent decreases in percentages of CD4 single positive cells as compared to percentages of these same populations from non-vaccinated pigs. In conclusion, these studies show that parenteral S. hyodysenteriae vaccination results in cellular immune responses detectable both peripherally (systemic immunity) as well as at the site of infection (mucosal immunity). However, it appears that regulatory mechanisms affecting IFN-gamma production in response to S. hyodysenteriae antigen differ between peripheral and colonic compartments.  相似文献

The immunity state after vaccination against Marek's disease (MD) was studied in three multiplier flocks of laying fowl (MFL), five multiplier flocks of broiler fowl (MFB), and one commercial layer flock (CLF). The occurrence and average titres of post-vaccination viraemia in the selected sets of chickens from these flocks, examined at the age of three or twenty weeks, were used as the immunity criterion. The development of post-vaccination viraemia, following the administration of the MARVAK vaccine at the doses of 100 and 1000 PFU per bird in the HX-SL and Shaver layer hybrids, was examined under laboratory conditions at the same time. In the group of birds examined in the third week of age and coming from the MFL vaccinated with the recommended MARVAK vaccine (dose (100 PFU per bird), 64.3% of the chicks were viraemic, the average viraemia titre being 12 PFU/10(7) leucocytes. After the administration of a four-fold vaccine dose, 57.1% of the chicks were viraemic, the average titre being 3.2 PFU/10(7) leucocytes. After the administration of a ten-fold dose of MARVAK vaccine, 80% of the chicks were viraemic and the average titre of viraemia was 8 PFU/10(7) leucocytes. In the MFB vaccinated with the recommended dose of the MARVAK vaccine, the percentage of viraemic chickens was 48.3% and the average titre of viraemia was 6.5 PFU/10(7) leucocytes. In the pullets examined at the age of 20 weeks the number of viraemic birds ranged from 20 to 50% and the average viraemia titres were from 3.8 to 13.1 PFU/10(7) leucocytes. In the flock affected by acute MD, no post-vaccination viraemia was found in the clinically diseases pullets. In the chickens of the HX-SL line vaccinated with the recommended MARVAK vaccine dose, viraemia culminated in the third week after vaccination (31.5 PFU/10(7) leucocytes), and after the use of the dose ten times as high as the recommended one the culmination came a week later (47 PFU/10(7) leucocytes). In the Shaver chicks vaccinated with the recommended dose or with the ten-fold dose of the MARVAK vaccine, the post-vaccination viraemia culminated in the fourth week after vaccination (94.9 and 116.8 PFU/10(7) leucocytes). The post-vaccination precipitation antibodies were first detected in the eighth week after vaccination.  相似文献

FliC, the flagellin antigen of Salmonella Enteritidis, was tested as a vaccine candidate for protective effect against a homologous challenge in chickens. After immunization with recombinant FliC (rFliC) or administration of phosphate-buffered saline (PBS) at 56 days old, the chickens were challenged with 10(9) colony-forming units of Salmonella Enteritidis at 76 days old. The vaccinated birds showed significantly decreased bacterial counts in the liver and cecal contents compared to those administered PBS at 7 days postchallenge, but the protection was partial. The replication experiment also showed a similar result. In both experiments, vaccination induced an increased level of serum anti-rFliC IgG, which was also reactive to the native flagella. The intestinal IgA level was slightly higher in the vaccinated birds than in the control. However, neither the proliferative response nor interferon-gamma secretion of splenic cells upon stimulation with rFliC was induced. Therefore, the effect of rFliC as a vaccine is limited, and further improvement is needed.  相似文献

Eimeria necatrix-specific ELISA, using a recombinant antigen (the cDNA-clone NP19 expressing protein), was utilized to detect antibodies against E. necatrix in breeder pullet flocks that had previously received an attenuated live vaccine to E. necatrix. Vaccinated flocks were discriminated significantly from non-vaccinated flocks by their antibody titers and antibody positive rates at 30-55 days post-vaccination. In addition, E. necatrix-oocysts were confirmed in fecal samples of vaccinated flocks using PCR in the case where the antibody positive rates rose. These findings implied that the vaccination prompted repeated infections, and consequently the chickens generated antibodies and secured their protection against virulent field-E. necatrix. Therefore, the ELISA was suggested to be a useful tool to estimate the immune state of chickens as a result of vaccination with a live E. necatrix-vaccine.  相似文献

雏鸡1日龄感染鸡贫血病毒,8日龄接种Lasota疫苗,以未感染免疫雏鸡为对照,于免疫后7、14、28d检测血清免疫球蛋白IgG、IgM、IgA,在凝抑制抗体(HI)滴度;胸腺、法氏囊、脾脏T细胞、IgG^ 、IgM^ 、IgA^ ,抗体生成细胞数量及T、B细胞增殖反应;胸腺、脾脏细胞因子IL-2、IFN活性的变化。结果发现,感染CAV雏鸡Lasota疫苗免疫后,其血清IgG、IgM、IgA免疫球蛋白含量明显减少,HI抗体滴度降低;胸腺、法氏囊、脾脏T细胞、抗体生成细胞数量降低及T、B细胞增殖反应减弱,胸腺、脾脏IL-2及TNF诱生活性降低,表明其细胞免疫和体流免疫功能以及细胞因子免疫调节作用均未感染免疫雏鸡明显减弱。  相似文献
