Serum biochemical analysis was undertaken to study the pathophysiological details of emaciation disease of the tiger puffer fish Takifugu rubripes (Temminck and Schlegel). Serum parameters were measured by biochemical analysis using automated dry chemistry and gas chromatography/mass spectrometry (GC/MS). Serum concentrations of albumin, amylase, calcium, creatinine, glucose and total protein were significantly lower in the emaciated fish when compared with those of normal fish. Regression analyses found close correlation between concentrations of total protein, albumin, amylase, glucose and progress of the disease. In contrast, serum alanine aminotransferase increased significantly in emaciated fish indicating liver function disorder. Further, GC/MS metabolic profiling of the puffer serum showed that the profile of the emaciated fish was distinct to that of non‐infected control. The serum content of amino acids including glycine, 5‐oxo‐proline and proline, and ascorbic acid, fumaric acid and glycerol increased significantly in serum in moderately emaciated fish. The serum glucose, linolenic acid and tyrosine level decreased significantly in the late phase of the disease. Our results clearly show that prolonged intestinal damage caused by myxosporean infection impairs absorption of nutrients, resulting in extreme emaciation.  相似文献

Although jellyfish blooms are serious nuisances for fisheries and other industries, the utilization and nutritional value of by‐catch jellyfish has drawn public attention. Here, we evaluated the efficacy of feeding tiger puffer Takifugu rubripes with moon jellyfish Aurelia sp. using 20‐day rearing experiments. Feeding on jellyfish had no positive effect on growth performance, although it significantly reduced neutral lipids and increased proportions of polar lipids, n?3 and n?6 highly unsaturated fatty acids (HUFAs), especially arachidonic and docosahexaenoic acids, and taurine. Supplemental feeding on jellyfish plus pellets elevated activity and responsiveness in the fish, with no effect on burst swimming speed or prevention of aggressive biting by siblings. The present research suggests that feeding on jellyfish improves body composition and some behaviours in tiger puffers. As cultured fish tend to accumulate excessive amounts of lipids from lipid‐rich pellets that can induce an unhealthy condition, and behavioural deficits of hatchery‐reared fish reduce survival after release in the wild, we propose the use of jellyfish as a supplemental diet in the nursery production of tiger puffer. Moreover, feeding on jellyfish can overcome deficiencies in alternative soy‐material feeds by supplying phospholipid, HUFAs, or taurine.  相似文献

In this study, the phenotypic and pathogenic properties in freshwater host were characterized in 14 strains of halophilic Vibrio harveyi isolated from infected marine black tiger shrimp, Penaeus monodon. The lysogenic phenotype was assayed via prophage excision and mitomycin C induction. The bacteria were grouped into two types, corresponding to lysogenic and non‐lysogenic strains. The pathogenicity was determined via direct injection of bacterial cultures into post‐larval juvenile giant freshwater prawns, Macrobrachium rosenbergii De Man. All of the infected prawns showed similar symptoms and inflamed hepatopancreas. The V. harveyi isolates derived from the first‐injected infected prawns were re‐isolated and re‐injected into healthy giant freshwater prawns, in which they retained similar infectivity. Both lysogenic and non‐lysogenic Vibrio spp. showed identical virulence associated with 100% mortality within one day post‐injection. TEM micrographs showed hepatopancreatic nuclear deformation and lipid breakdown caused by lysogenic γ‐hemolytic VL19 and non‐lysogenic β‐hemolytic V33. However, the V33 strain was associated with severely disrupted mitochondria. None of the V. harveyi strains was able to produce a biofilm. Together, our findings indicate that the lysogenic and non‐lysogenic halophilic V. harveyi isolated from marine shrimps may use different virulence factors that are responsible for their pathogenicity in freshwater prawns.  相似文献

Effects of daily feeding frequency, water temperature, and stocking density on the growth of tiger puffer, Takifugu rubripes, fry were examined to develop effective techniques to produce tiger puffer in a closed recirculation system. Fish of 4, 14, and 180 g in initial body weight were fed commercial pellet diets once to five times a day to apparent satiation each by hand for 8 or 12 wk at 20 C. Daily feeding frequency did not affect the growth of 14‐ and 180‐g‐size fish. However, the daily feed consumption and weight gain of the 4‐g‐size fish fed three and five times daily were significantly higher than those of fish fed once daily (P < 0.05). Fish of 4 and 50 g in initial body weight were reared with the pellet diet at 15–30 C for 8 wk. The weight gain of fish increased with increasing water temperature up to 25 C and decreased drastically at 30 C for both sizes. Similar trends were observed for feed efficiency, although 4‐g fish had highest efficiency at 20 C. Effects of stocking density on growth were examined with fish of 8, 13, and 100 g in initial body weight. Fish were reared with the pellet diet for 8 or 16 wk at 20 C. Fish were placed in floating net cages in the culture tank, and the stocking density was determined based on the total weight of fish and volume of the net cage. Fish of 8 g in body weight grew up to 35–36 g during the 8‐wk rearing period independent of the stocking density of 8, 15, and 31 kg/m3 at the end of rearing. Final biomass per cage reached 32, 60, and 115 kg/m3 for 13‐g‐size fish, and 10, 18, and 35 kg/m3 for 100‐g‐size fish, and the growth of the fish tended to decrease with increasing stocking density for both sizes.  相似文献

The effects of water temperature on the development of the enteric myxosporeans, Enteromyxum fugu and Enteromyxum leei, were investigated in experimentally infected tiger puffer, Takifugu rubripes. After naïve tiger puffer were fed gut tissue infected with both E. fugu and E. leei, they were divided into separate tanks and kept at different constant temperature regimes between 10 and 25 °C. Regardless of the water temperature tested, E. fugu was consistently detected with a high prevalence of infection (60–100%), although no sporulation occurred at 10 and 15 °C. Development of E. leei and the onset of disease were suppressed by low water temperatures (<15 °C). However, a temperature increase to 20 °C promoted the development of E. leei, followed by an increase of disease rate in the fish. The present study demonstrates that water temperatures below 15 °C have an inhibitory effect on the development of E. fugu and E. leei, resulting in suppression of enteromyxosis at low temperatures.  相似文献

The lipid‐regulating effects of astaxanthin (ASTX) have been widely reported in terrestrial animals. However, little relevant information has been available in fish although ASTX has been used as exogenous pigment and antioxidant in fish feed. A 74‐day feeding study was conducted to investigate the effects of dietary ASTX on lipid accumulation in the marine teleost tiger puffer. Four experimental diets differing only in ASTX supplementation, that is, 0 (control), 50 (ASTX50), 100 (ASTX100) and 500 (ASTX500) mg kg‐1, were randomly assigned to 12 tanks of juvenile tiger puffer. Compared to control, the liver lipid content in group ASTX50 was significantly higher, while those in groups ASTX100 and ASTX500 were lower. The muscle lipid contents in group ASTX500 were significantly higher compared to control. Group ASTX50 had the best growth performances, while diet ASTX500 seemed to have adverse effects. In the liver, compared to control, groups ASTX50 and ASTX100 showed significantly lower mRNA expressions of genes related to triglycerol synthesis and fatty acid synthesis, transport and uptake, but higher expressions of genes related to β‐oxidation and monoglycerol hydrolysis. In the muscle, compared to control, ASTX100 showed higher expressions of genes related to β‐oxidation. ASTX50 resulted in higher contents of saturated and monounsaturated fatty acids but lower contents of n‐3 polyunsaturated fatty acids in fish. In conclusion, astaxanthin in diets for tiger puffer differentially regulated the lipid accumulation in the liver and muscle, both in dose‐dependent manners. ABSTRACT:   Rearing experiments were conducted to investigate the essential fatty acid requirements in the early developmental stages of river puffer Takifugu obscurus and tiger puffer T. rubripes using two n-3 series unsaturated fatty acids, α-linolenic acid (18:3n-3, α-LNA) and docosahexaenoic acid (22:6n-3, DHA), under salinity of 30 and 18.5–20.3°C. River and tiger puffer larvae used in this study were 15 and 14 days old after hatching, and their average body weights were 30.1 and 20.8 mg, respectively. The results on fatty acid requirements of these two species were evaluated from fish growth, survival, fatty acid composition of the fish body and activity test results. The DHA groups of both river and tiger puffer exhibited better survival and weight gain. However, there was no difference in the mean final body weights of river puffer between two dietary groups. Also, the DHA group of tiger puffer showed better results in the recovery test from anesthetic condition than that obtained in the LNA group. In an examination of the fatty acid compositions of the whole body, the LNA group containing no dietary DHA resulted in 0.5% DHA in tiger puffer and 1.1% DHA in river puffer . These results suggest that α-LNA from Artemia converted to eicosapentaenoic acid (20:5n-3, EPA) and to DHA successively by their fatty acid metabolism. Symptoms following essential fatty acid deficiency were not observed in any experimental groups. As river puffer did not represent a significant difference in the dietary effects between α-LNA and DHA treatment groups, its essential fatty acid requirement was assumed to be somewhat closer to that of the freshwater fishes in comparison with that of marine fishes, including tiger puffer.  相似文献

In vitro accumulation of tetrodotoxin (TTX) and paralytic shellfish toxin (PST) in tiger puffer fish Takifugu rubripes was investigated using liver tissue slices. When T. rubripes liver slices were incubated with Leibovitz’s L-15 medium containing 0.13 mM TTX at 20 °C in air with saturated humidity, they accumulated 21.5 ± 7.3 μg TTX g−1 liver after the incubation for 12 h and increased to 55.3 ± 8.2 μg TTX g−1 liver at 48 h. In the incubation of T. rubripes liver slices with 0.13 mM PST-containing medium, PST was detected 6.3 ± 0.9 μg g−1 liver at 12 h and reached a plateau thereafter. These results reveal the difference between TTX and PST in accumulation in T. rubripes liver tissue slices. To examine the variation in PST accumulation among fish species, the liver tissue slices from tiger puffer fish T. rubripes, parrot-bass Oplegnathus fasciatus and green ling Hexagrammos otakii were incubated at a concentration of 0.027 mM PST. The toxin contents of 3.0 μg g−1 liver were observed at 8 h regardless of fish species but were not increased subsequently, showing no variety among these three species as to accumulation patterns of PST. Two 12‐wk rearing experiments were conducted to examine the effect of rearing salinities of 10–35 ppt on the growth of 3‐ and 170‐g‐size tiger puffer, Takifugu rubripes. Fish were reared in a closed recirculation system without introducing fresh culture water at 23 C and were fed commercial pellet diet for tiger puffer twice or three times daily to apparent satiation each, almost everyday. Growth of 3‐g‐size fish seemed to increase with decreasing salinity; however, there were no significant differences in the specific growth rate and weight gain among treatments because of differences in initial body weight. Final body weight and length of fish reared at 10 ppt were significantly higher than those for fish reared at 30 ppt although initial sizes were similar. Differences were not found for the feed efficiency (FE) and daily feed consumption. Apparent relationships were not observed between salinity and blood characteristics or proximate compositions of muscle of the cultured fish. Differing from smaller fish, growth of 170‐g‐size fish tended to decrease with decreasing salinity from 30 to 10 ppt and with increasing salinity from 30 to 35 ppt. Vibrio harveyi causes vibriosis in various marine aquaculture fish species, especially when they are young. The infection subsequently leads to significant economic losses for aquaculture farms. Vaccination is recommended to control this disease. This study describes the efficacy of a live attenuated V. harveyi strain MVh_vhs (LAVh) as a vaccine candidate in controlling infection by wild‐type V. harveyi (WTVh) in Lates calcarifer. A total of 240 fingerlings were divided into four groups. Group 1 was not vaccinated and was not challenged, Group 2 was vaccinated with a formalin‐killed V. harveyi (FKVh), Group 3 was vaccinated with the LAVh before challenge and Group 4 was not vaccinated and was challenged. Bath vaccination was employed for one hour before the LAVh distribution was determined in the fish mucus, gill, liver, gut, kidney and spleen. The gills, livers, kidneys and skins were also sampled for gene expression analysis. To challenge the fish, skin abrasion was conducted before the fish were challenged by immersion with WTVh. The results revealed an extensive distribution of the LAVh in the liver and kidneys of the fish in Group 3 for the first 12 hr, resulting in mild lesions compared with Group 1. Similarly, there were significantly (p < .05) higher expressions of the Chemokine ligand 4 and major histocompatibility complex I genes in the skin and liver of the fish in Group 3 in comparison with other groups. Vaccination with LAVh resulted in a significantly high rate of survival (68%) of the fingerlings after being challenged with WTVh.  相似文献

Pyridinoline (Pyr), one of the mature crosslinks of collagen, was determined in muscular collagen of three species of fish. The amounts of muscular Pyr in red sea bream, yellowtail, and tiger puffer were 3.4, 8.8, and 50.3 mmol/mol collagen, respectively, indicating that the Pyr concentration in muscular collagen differs greatly among fish species. The Pyr concentration in tiger puffer muscular collagen was the greatest, but it was only one-fourth that in rabbit muscle. As in mammalian skin collagen, Pyr was not detected in skin collagen of red sea bream and yellowtail. However, tiger puffer skin contained Pyr (3.75 mmol/mol collagen). The presence of Pyr would have a relationship to some features of tiger puffer skin, such as mechanical strength and thickness. Pyr concentrations in acid-soluble collagen (ASC), pepsin-solubilized collagen (PSC), and insoluble collagen (ISC) in muscles of three species of fish were determined. Pyr was found in ISC > PSC > ASC, from the highest to the lowest concentration, and the concentration in ISC was 45–200 times that in ASC. Therefore, Pyr crosslinks that are formed between collagen molecules would have a close relationship to collagen solubility.  相似文献

Inclusion of the water-soluble fraction of blue mussels Mytilus galloprovincialis as a feed additive was examined with tiger puffer Takifugu rupbripes. The control diet mainly consisted of fish meal, potato starch, and pollack liver oil. Experimental diets were formulated to replace 30% and 40% of the fish meal protein with defatted soybean meal (SBM), and were supplemented with 0–20% mussel extracts. Fish of 18 g initial body weight were fed each diet to satiation, twice daily, 6 days per week for 7 weeks at 20°C. Weight gain and protein efficiency ratio of fish fed the diet containing SBM without the extract were significantly lower than those of the control at the 30% replacement level. Addition of 5% or 10% extract improved the growth and feed utilization to levels comparable to those of the control. At 40% substitution, statistically identical growth and feed performance to the control were obtained for diets supplemented with 10% or 20% extract; however, performance for 5% addition was significantly lower. Thus, the water-soluble fraction of blue mussels would be an effective feeding stimulant in aquaculture feed for tiger puffer that makes it possible to replace 30–40% of fish meal protein with SBM.  相似文献

Scale drop and muscle necrosis disease with high mortality widely occurred recently in the hybrid grouper (Epinephelus fuscoguttatus × E. lanceolatus ♂), a crucial cultured marine fish species in China. In this study, 30 Harveyi clade isolates of 27 Vibrio harveyi strains were isolated from diseased hybrid groupers in the south‐east and north‐east coastal areas of China. A total of 22 V. harveyi strains were determined to be pathogenic, and most challenged fish died within 2 days of infection; surviving individuals exhibited scale drop and deep dermal lesions as naturally diseased fish. Although five typical virulence genes, including luxR, toxRVh, chiA, serine protease and vhh widely existed in V. harveyi, no obvious correlation was established between virulent strains and virulence genes harboured in them. Furthermore, multiple antibiotic resistance was widely exhibited in Harveyi clade strains, particularly for penicillins, polypeptides, lincomycins, acetylspiramycin, streptomycin, metronidazole and bacitracin. And the multiple antibiotic resistance indices were gradually decreased from southern to northern areas of China. This study demonstrated that the pathogenic V. harveyi with multiple antibiotic resistance is highly prevalent in hybrid grouper in China, which requires particular attention.  相似文献

采用2216E和TCBS培养基分别从患溃疡病半滑舌鳎的肠道和体表病灶分离获得菌株40株,经血平板检验分离菌株的溶血活性,体外筛选并鉴定出3株潜在致病性菌株,即哈维氏弧菌、溶藻弧菌以及副溶血性弧菌。通过将3株潜在致病菌与健康半滑舌鳎的肠道上皮细胞共培养,检测病原菌刺激后半滑舌鳎肠道上皮细胞相关免疫基因的相对表达量及共培养后肠道上皮细胞的凋亡率,对病原菌的致病性进行细胞水平的筛选。结果显示,经哈维氏弧菌刺激后肠道上皮细胞白介素10基因(IL-10)表达量极显著上调,说明哈维氏弧菌引起的共培养上皮细胞免疫反应最为剧烈,且哈维氏弧菌与肠道上皮细胞共培养后导致细胞的凋亡率高达48.3%,其次为溶藻弧菌(36%)和副溶血性弧菌(34.5%),推测哈维氏弧菌为半滑舌鳎溃疡病高致病性弧菌。通过对半滑舌鳎进行浸浴回感实验,结果发现,哈维氏弧菌、溶藻弧菌和副溶血性弧菌人工回感后第6天半滑舌鳎的累计死亡率分别为100%、95%和75%,与细胞水平检测结果相吻合。实验表明,所筛选到的3株弧菌均具有较强的毒性和致病性,尤其以哈维氏弧菌致病性最强,并由此推测半滑舌鳎溃疡病可能由多种致病菌共同感染所致。  相似文献

In this study, susceptibility and potential carrier status of burbot, Lota lota, were assessed for five important fish pathogens. Burbot demonstrated susceptibility and elevated mortality following challenge with infectious haematopoietic necrosis virus (IHNV) by immersion and to Aeromonas salmonicida by intraperitoneal (i.p.) injection. IHNV persisted in fish for at least 28 days, whereas A. salmonicida was not re-isolated beyond 17 days post-challenge. In contrast, burbot appeared refractory to Flavobacterium psychrophilum following intramuscular (i.m.) injection and to infectious pancreatic necrosis virus (IPNV) by immersion. However, i.p injection of IPNV resulted in re-isolation of virus from fish for the duration of the 28 day challenge. Renibacterium salmoninarum appeared to induce an asymptomatic carrier state in burbot following i.p. injection, but overt manifestation of disease was not apparent. Viable bacteria persisted in fish for at least 41 days, and bacterial DNA isolated by diagnostic polymerase chain reaction was detected from burbot kidney tissue 90 days after initial exposure. This study is the first to investigate susceptibility of burbot to selected fish pathogens, and this information will aid in efforts to culture and manage this species.  相似文献

Tiger puffer, Takifugu rubripes, juveniles were fed with four semi-purified experimental diets containing 0.2% Ca from Ca-lactate (diet 1), no supplemental Ca (diet 2) and 0.2% and 2.5% Ca from tricalcium phosphate, TCP (diets 3 and 4), respectively. After a 10 week rearing period, growth and feed utilization were significantly lower in the fish group fed on diet 2 than in the fish group fed on diet 1. Fish groups fed on diets 3 and 4 also showed poor growth performances compared with group 1. It appears that Ca intake from seawater is not sufficient for the normal growth of tiger puffer. Furthermore, Ca in dietary TCP appeared to be unavailable to this species. Dietary TCP strongly inhibited the bone mineralization of Zn and Mn. The findings indicate that easily digestible Ca supplementation is indispensable in a diet of tiger puffer for normal growth, feed utilization and bone mineralization. © Rapid Science Ltd. 1998  相似文献

ABSTRACT

Obscure puffer fish (Takifugu obscurus) flesh, often eaten as raw fish fillet, is susceptible to softening. To investigate the mechanisms for the softening of flesh and changes in firmness, the proliferation of total microbial flora, Enterobacteriaceae, Pseudomonas spp., and lactic acid bacteria (LAB), total volatile basic nitrogen (TVBN), trimethylamine (TMA), and pH were periodically assayed during storage at 0, 4, and 8ºC. Myofibril extraction and separation of puffer fish flesh was carried out and the length of the separated myofibrils was measured periodically using an optical microscope during the entire storage time. Transmission electron microscopy was also used to inspect the changes in myofibrillar characteristics of the fish flesh at the beginning and end of the storage period. The results indicated that lower chilled temperature can contribute to reduced proliferation of microbial flora and less variation in TVBN, TMA, and pH and further maintain the integrity of myofibrils and retain firmness of puffer fish flesh during storage.  相似文献

The hybrid grouper, Epinephelus fuscoguttatus (♀) × Epinephelus lanceolatus (♂), is a newly bred cultivated marine fish species of high economic value. However, a skin ulcer disease with high mortality has occurred, and the responsible pathogen remains unknown. In this study, we summarized the epidemic status and external signs of this disease. We screened potential pathogens and finally isolated one bacterial strain ML01 from affected fish. We subjected healthy juvenile hybrid groupers to bacterial challenge tests with the isolate by immersion, immersion after dermal abrasion and intraperitoneal injection, respectively. Within 14 days post‐infection, the isolate ML01 caused mass mortality of juveniles infected via immersion after dermal abrasion or intraperitoneal injection. Diseased juveniles displayed obvious signs of skin ulcers. The median lethal dose of ML01 by intraperitoneal injection was 1.10 × 105 colony‐forming units. ML01 was identified as Vibrio harveyi by bacterial morphology, analytical profile index identification, 16S rDNA sequencing and multilocus sequence analysis. Antibiotic susceptibility tests showed that ML01 was sensitive to ceftriaxone, doxycycline and minocycline. The results of this study suggest that V. harveyi is the causal agent of skin ulcer disease in juvenile hybrid groupers, thus providing a basis for effective control and prevention of this disease.  相似文献

Effects of dietary fatty acid composition on the growth of the tiger puffer Takifugu rubripes were examined. Eight experimental diets were formulated with fish meal and casein as the major ingredients, providing 45.0–48.2% crude protein. Pollack and squid liver oils were used for the control diet while experimental diets contained three levels of EPA-DHA concentrated (C-HUFA) oils, soybean oil, linseed oil, and combinations of them, providing 0.5–5.5% n-3 HUFA and 5.0–11.1% crude lipid. Triplicate groups of fish with a mean body weight of 18.7 g per dietary treatment were fed the diets twice daily to apparent satiation, 6 days per week for 8 weeks. The final body weight and weight gain of the fish fed the soybean oil diet without C-HUFA oils were significantly lower than those fed the control diet. Both parameters were not statistically different among the other dietary groups, although the values obtained for the linseed oil diet without C-HUFA oils were lower. Similar trends were seen for SGR and feed efficiency. However, significantly lower PER values compared to the control were observed for the lower C-HUFA and the vegetable oil without C-HUFA oil groups. These results indicate that tiger puffer can be produced successfully with diets containing more than 1.5% n-3 HUFA.  相似文献
