陕西马铃薯卷叶病病原的分子生物学鉴定

摘要:
【目的】 对陕西榆林疑似感染马铃薯卷叶病毒(Potato leafroll virus,PLRV)的病株毒源进行分子生物学鉴定,为陕西省马铃薯卷叶病的防治提供参考。【方法】 根据GenBank数据库已报道的马铃薯卷叶病毒外壳蛋白(Coat protein,CP)基因序列设计了1对引物,利用RT-PCR技术成功扩增了陕西PLRV·CP基因,然后进行克隆、测序,并与GenBank中已报道的27个PLRV分离物的CP基因进行同源性分析。【结果】 获得的陕西PLRV·CP基因大小为627 bp,含有1个ORF,编码208个氨基酸的多肽,其与已报道的27个国内外PLRV·CP高度同源。【结论】 陕西榆林的疑似病株确定为感染PLRV的马铃薯病株。

Molecular biological identification of potato leafroll virus from field samples in Shaanxi
Abstract:
【Objective】 The study was to identify samples of potato leafroll diseases from Shaanxi to provide thcoretical reference for the control of potato leafroll diseases.【Method】 Using specific primers designed according to the converted sequences of potato leafroll virus(PLRV) on GenBank,the reaction of RT-PCR was used to obtain virus CP gene,to conduct sequence analysis and homological comparison.【Result】 A 627 bp fragment of CP including an open reading frame coding for 208 amino acids was successfully amplified by this RT-PCR system,and it possessed a very high similarity with previously reported 27 isolates of PLRV·CP.【Conclusion】 Molecular investigation indicates that the potato leafroll diseases from field samples was infected by potato leafroll virus in Shaanxi.
