伪狂犬病基因缺失疫苗株(SA215)某些生物学特性研究

本试验测定了伪狂犬病ｇＥ－／ｇＩ－／ ＴＫ－／ ＬａｃＺ＋基因缺失疫苗株（ＳＡ２１５）的致细胞病变效应、安全性、免疫原性和免疫期等生物学特性。试验结果显示，该疫苗株能在Ｖｅｒｏ细胞上适应生长，并形成典型的蚀斑。其对１日龄仔猪、怀孕母猪、牛、羊以及家兔安全，无不良接种反应，接种动物不向体外散毒。ＳＡ２１５疫苗接种猪能抵御高剂量（１０７ＰＦＵ）Ｆａ株强毒感染，攻毒后试验猪的发热期、增重受阻天数、散毒滴度均低于Ｂａｒｔｈａ株疫苗接种猪，远远低于对照组猪。ＳＡ２１５接种猪能维持长时间的高水平中和抗体滴度，免疫期可达半年以上。试验结果表明，ＳＡ２１５株是一株安全、免疫原性好、免疫期长的疫苗株。

The Biological Characteristics of Pseudorabies Virus Gene-deleted Vaccine Strain(SA215)

We tested the culture, safety, efficacy and antibody duration of a gene-deleted pseudorabies virus vaccine strain(SA215), which lacked glycoprotein gE,gI and thymidine kinase genes,but can express LacZ gene in this paper. SA215 could propagate well, but it was severely impaired in plaque formation , inducing tiny plaques on Vero cells. It was safe to 1-day-old piglets, pregnant sows, cattle, sheep, goats and rabbits,The vaccinated animals with SA215 strain were less clinically affected and the SA215 strain couldn' t be recovered from vaccinated animals.Pigs vaccinated i. m with SA215 could be protected from being affected after oronasal challenge-exposure with 107 P. F. U. high-dose virulent pseudorabies virus Fa strain.The periods of fever and growth arrest and titer of exertion virus of the pigs vaccinated with SA215 were lower than that of the pigs vaccinated with Bartha strain, but were much lower than than of the unvaccinated pigs. The vaccinated pigs with SA215 strain produced high-level neutralizating antibodies for pseudorabies virus that could maintain for over half a year.The result showed that pseudorabies virus SA215 stain was a safe vaccine that had high efficacy and long duration of anti-PRV antibody.
