兔病毒性出血症病毒中国株(JX/CHA/97)全基因组分子克隆及序列分析

摘    要:本研究首次完成了免病毒性出血症病毒(Rabbit hemorrhagic disease virus,RHDV)中国分离株JX/CHA/97全基因组序列的测定与分析。研究结果表明,JX/CHA/97株的全基因组由7437个核苷酸组成,其基因组构成为:5'端有一个长度仅为9nt的非编码区,随后有2个阅读框架(ORFs).分别编码一个聚合蛋白和一个结构蛋白。在基因组的3'端也有一个非编码区,长度为59nt。另外,采用RACE方法获得了RHDV的poly(A)尾序,证明该结构至少含有27个A。根据RHDV衣壳蛋白的基因序列,将JX/CHA/97株与来自世界各地的22株RHDV分离株的基因组序列进行了比较与分析,并绘制了系统发生树。结果表明,RHDV各分离株之间存在较高的序列同源性,可以将它们至少分为6个拓朴群,提示RHDV有朝不同方向发生进化的趋势。

Abstract:The complete nucleotide sequence of the China strain of rabbit hemorrhagic disease virus(RHDV)was determined to be 7 437 nucleotides in length with a Y-terminal non-coding region of 9 nucleotides and a 3'-terminal non-coding region of 59 nucleotides.Two open reading frames(ORFs)were found within this sequence coding for a polymerize protein and a structural protein.Besides,our data showed that the poly(A)tail of RHDV had 27 A at least.Sequence comparison studies revealed significant homology between nonstructural proteins or structural proteins of JX/CHA/97 strain and the other strains. However,the results also showed that groups of RHDV strains appeared to have evolved to different extents.
