雌核发育鲢RAPD指纹和蛋白质电泳研究

摘    要:用23个随机引物对雌核发育鲢子一代(普通鲢作对照)进行了RAPD—PCR反应,在23个引物中除一个引物无扩增产物外,其它均可得到1~10条DNA带,平均每个引物产生538条带。其中7个引物产生多态现象,占总引物的314%。多态座位达1368%。用Shannon指数对RAPD数据进行遗传多样性(Ho)分析,得出所研究样本的Ho为0175。用血清酯酶和血清蛋白电泳作对比时没有发现在蛋白质水平的多态现象,说明RAPD技术是一种比较灵敏实用的DNA多态检测方法。以RAPD所得数据进行了雌核发育鲢子代和普通鲢个体间的遗传相似性与遗传距离分析,为鲢选育提供了依据。

Abstract:Genetic analysis of gynogenetic silver carp including normal silver carp was conducted by means of Random Amplified Polymorphic DNA(RAPD)and protein electrophoresis.23 10 mer primers were used for RAPD PCR reaction.Only one primer did not produce any band.The other 22 primers produced bands ranging from 1 to 10,and on average ,each primer gave rise 1 to 10 bands.Among primers used 7 primers produced polymorphic bands among individual studied.Total 117 bands were produced by 22 primers,and out of those bands(or loci)16(13 68%)bands are polymorphism.Genetic diversity(Ho)was 0 1475 among sample studied.However,no polymorphism was detected by serum protein and Esterase electrophoresis.According to data obtained by RAPD,the genetic similarity and genetic distance among individuals studied are provided.The application of RAPD for carp population genetics and selective breeding was discussed.
