摘    要:以佛手的叶片、茎段为材料,建立了其组织培养系统。研究表明,B5比M S更适合作为诱导佛手愈伤组织的基础培养基,诱导愈伤组织效率较高的培养基配方为B5+2.0 m g.L-12,4-D+0.2 m g.L-1ZT和B5+1.5 m g.-L 12,4-D+1.0 m g.L-1NAA+0.4 m g.L-16-BA。在叶片对卡那霉素敏感性的试验中发现,叶片外植体预培养2 d后进行筛选的效果较好,卡那霉素的适宜筛选浓度为25 m g.L-1。

Abstract:The preliminary results of tissue culture of bergamot were reported in this paper.The results showed that B_5 medium was more suitable for callus inducement than MS medium.Leaves and stem segments with axillary buds of bergamot were used as explants,and a tissue culture system were established.B_5+2.0 mg/L 2,4-D +0.2 mg/L ZT and B_5+1.5 mg/L 2,4-D+1.0 mg/L NAA+0.4 mg/L 6-BA media were more suitable for callus inducement.The appropriate concentration of kanamycin for selection of transformed leaf was 25 mg/L.
