忍冬茎叶中绿原酸的提取工艺研究

摘    要:以高效液相色谱为检测手段,绿原酸提取率为考察指标,比较了回流法和超声法提取忍冬茎叶中绿原酸的影响;采用单因素和正交试验设计,探讨了回流法提取忍冬茎叶中绿原酸的最佳工艺参数。结果表明,最佳提取条件为乙醇浓度70%,料液比1∶14,提取时间2h,提取温度60℃,提取溶剂最初pH 6,此时绿原酸提取率最高,可达1.37%。

Abstract:Two methods were compared to extract chlorogenic acid from the stems and leaves of Lonicera japonica,alcohol reflux extraction and ultrasound-assisted maceration extraction.The content of chlorogenic acid was used as index,which was measured by high performance liquid chromatography.Single factor and orthogonal design experiments were adopted to select optimal extraction conditions in alcohol reflux extraction.The results showed that under the same concentrations of ethanol,reflux extraction was better than ultrasound-assisted maceration extraction.Optimal reflux extraction conditions were as follows: solvent: 70% ethanol,ratio of material to solvent(W∶V): 1∶14,reflux time: 2 h,extraction temperature: 60℃,and pH 6.Under these conditions,the extraction ratio of chlorogenic acid was up to 1.37%.
