公农1号紫花苜蓿离体培养中最佳消毒方式和外植体种类的筛选研究

研究了不同消毒方式对公农1号紫花苜蓿种子接种培养基的侵染情况和子叶、叶片、胚轴等不同外植体对愈伤组织诱导率的影响。结果表明：在消毒前,种子经无菌水浸泡24 h（预处理）比不浸泡的消毒效果好,75%乙醇预处理后可大幅降低种子的带菌率,但预处理时间不宜过长;所用消毒剂中0.1%升汞消毒效果最好,种子带菌率为零,三种消毒剂中10%次氯酸钙—无菌水处理的紫花苜蓿种子发芽率最高;三种外植体均容易形成愈伤组织,经过综合比较,胚轴为公农1号紫花苜蓿品种离体培养的最佳外植体。

Screening of Sterilization Methods and Explants for in vitro Culture of Alfalfa Variety Gongnong 1

With the alfalfa variety Gongnong 1 as experiment material,the influences of different disinfection methods on infection situation and germination of seeds were studied and also the effects of different explants(cotyledons,leaves and hypocotyls)on callus induction.The results showed that soaking seeds in aseptic water for 24 hours before sterilization was better than without soaking.The bacteria rate of seeds could be greatly decreased through pretreatment with 75% alcohol,but the pretreatment time should not be too long.0.1% HgCl2 had the best sterilization effect with zero bacteria rate,while the treatment of 10% calcium hypochlorite combined with soaking seeds in aseptic water had the highest germination rate.The callus could be obtained from the three kinds of explants,but hypocotyl was the optimal one.
