茉莉离体微繁及无糖生根技术

摘    要:为解决茉莉长期无性繁殖导致的种性退化、抗逆性下降、花朵产量逐年下降等问题,进行了茉莉离体微繁及无糖生根技术的研究.结果表明:茉莉腋芽萌发的适宜培养基为MS 6-BA 2.0 mg/L NAA 0.1 mg/L;继代增殖的适宜培养基为MS 6-BA 1.5 mg/L NAA 0.3 mg/L GA3 0.5 mg/L;无糖生根的适宜培养基为1/2 MS NAA 1.2 mg/L;适宜培养条件为光强5 000~5 500 lx、CO2浓度1 500 mg/L、通气孔数为3个.无糖试管苗移栽后长势健壮,成活率可达95%.

Abstract:Due to low seed-setting rate in the nature,cuttage is widely used for the reproduction of jasmine.However,long term cuttage causes varietal degeneration,resistance weakness,and declining of flower produciton.Tissue culture and micropropagation is an effective way to avoid this.The optimum media and condition for tissue culture and micropropagation of jasmine were developed.The media favoring axillary bud germination,axillary bud reproduction and sugar-free rooting were MS with the combination of BA(2.0 mg/L) and NAA(0.1 mg/L),MS with the combination of BA(1.5 mg/L),NAA(0.3 mg/L) and GA3(0.5 mg/L),and 1/2 MS with NAA(1.2 mg/L),respectively.The condition suitable for test-tube plantlet growth were light density 5 000-5 500 lx,CO2 concentration of 1 500 mg/L,and three ventholes.The sugar-free cultured plantlet grew strongly,the survival rate reaching 95%.
