菥蓂药材中异牡荆苷的分离鉴定及指纹图谱研究

摘    要:采用高效半制备液相色谱法分离,并以13C谱法(13C-NMR)、1H谱法(1H-NMR)、质谱法(Ms)、紫外光谱法(UV)确定结构;采用HPLC梯度洗脱进行色谱分离,对不同产地的11批菥蓂药材指纹图谱的相似度进行比较分析。结果表明,分离得到异牡荆苷纯度>98%(面积归一化法);各批药材共建立了17个共有峰,不同产地的菥蓂药材指纹图谱相似度良好。说明高效半制备液相色谱法制备异牡荆素方法简单、高效,指纹图谱分析方法稳定性、精密度、重现性较好,用于菥蓂的质量评价切实可行。

Abstract:Semi-preparative HPLC was used to isolate and purify the isovitexin from Thlaspi arvense.Its chemical structures was elucidated by 13C-NMR、1H-NMR、Ms、UV;11 batches of Thlaspi arvense samples from different sources were analysed by HPLC gradient elution method.The results showed that the purity of isovitexin reached 98%(by Area normalization method),17 common peaks were selected in chromatograms,and all the common peaks were separated effectively,11 batches of Thlaspi arvense fingerprint had good similarity.The results indicated that semi-preparative HPLC was a simple quick and efficient method to prepare isovitexin.Fingerprint analytical method,being with good precision and repeatability,can be useful in the quality control of Thlaspi arvense.
