莲雾组织培养和快速繁殖技术

摘    要:以莲雾未萌发带顶芽茎段为材料研究其组织培养和快速繁殖技术。结果表明:莲雾组织培养的基本培养基为WPM;带顶芽茎段诱导芽的适宜培养基为WPM+0.5 mg.L-16-BA+0.02 mg.L-1NAA;通过腋芽增殖途径的适宜培养基为WPM+0.8 mg.L-16-BA+0.4 mg.L-1IBA;壮苗适宜培养基为WPM+0.2 mg.L-16-BA+0.4 mg.L-1IBA;生根适宜培养基为1/2WPM+0.5 mg.L-1NAA+0.2 mg.L-1IAA。组培苗驯化移栽成活率达90%以上。剪取成活组培苗顶部枝梢进行嫩枝(幼态)扦插生根成苗,能提高繁殖率,降低成本。

Abstract:A study was conducted on the technique of tissue culture and rapid propagation with the stem segments of wax jumbu as material.The results showed that WPM medium was its basic one,WPM+0.5 mg·L~(-1) 6-BA + 0.02 mg·L~(-1) NAA was suitable medium for generation of induction bud from stem segments with terminal bud,WPM+0.8 mg·L~(-1) 6-BA+0.4 mg·L~(-1) IBA for multiplication of the buds with germination of axillary buds,WPM+0.2 mg·L~(-1) 6-BA+0.4 mg·L~(-1) IBA for sound seedling,(1/2)WPM+0.5 mg·L~(-1) NAA+0.2 mg·L~(-1) IAA for rooting of shootings.The rate of survival after habituation was over 90%.The method which is to cut the tip of a branch from tissue cultured seedlings survival in transplant for cuttage,could enhance rate of multiplication and reduce cost.
