黄芩苷微生物转化菌的筛选鉴定及发酵条件优化

摘    要:通过罗尔夫青霉(Penicillium rolfsii)发酵培养基和发酵条件优化,来提升黄芩苷转化率。采用单因素方法对氮源、发酵接种量、初始pH发酵时间、发酵温度等因素进行考查,在此基础上进行L_(16)(4~5)正交试验优化。得到的优化发酵条件是黄芩粉20 g/L,(NH_4)_2SO_4 0.6 g/L,KH_2PO_4 0.2 g/L,醋酸钠0.1 g/L,维生素C 0.1 g/L,MgSO_4 0.1 g/L,pH 5.0,料液比为1∶30,接种量10%,装液量80 mL/250 mL,发酵温度35℃,发酵时间6 d,转速180 r/min。在最优条件下,黄芩素含量达到1.83 mg/mL。

Abstract:The Penicillium rolfsii fermentation medium and conditions were optimized to increase the baicalin yield. Firstly, us-ing single-factor test, the effects of these 5 parameters, i.e., nitrogen source, the inoculation amount, initial pH of medium, fermentation time, fermentation temperature on the baicalin yield were studied. Then the key parameters of fermentation condi-tions were statistically optimized through orthogonal array experiments with 5 factors at 4 levels. The optimal conditions were Scutellaria baicalensis Georgi. power 20 g/L,(NH4)2SO40.6 g/L, KH2PO40.2 g/L, sodium salt 0.1 g/L, MgSO40.1 g/L, vitamin C 0.1 g/L, initial pH 5.0, solid liquid ratio 1:30, rotation speed 180 r/min, 10% inoculum size in 80 mL working volume(in 250 mL flasks) at 35℃ for 6 d. Using these conditions, the baicalein yield was 1.83 mg/mL.
