摘    要:考察了消毒方式、外植体部位、培养基配方等因素对薄荷(Mentha haplocalyx Brip.)组织培养的影响.结果表明,外植体先用体积分数70%的乙醇处理30 s后,再用1 mg/L HgCl2+2滴吐温80浸泡一定时间,消毒效果好于40 mg/L的漂白粉处理,适宜的HgCl2消毒时间分别为茎段、叶片、叶柄和已萌发的芽12 min;末萌发的芽10 min.茎段的不定芽诱导率和不定芽生长情况好于其他外植体.在薄荷旺盛生长期采集半木质化的茎段作为外植体进行诱导培养,不定芽萌芽早而且生长速度快.MS+0.50 mg/LBA+0.10 mg/L NAA是适合外植体不定芽诱导的培养基配方.

Abstract:The effects of sterilizing methods,explant material,culture medium formula on tissue culture of mint(Mentha haplocalyx Brip.) was studied.The results showed that after explants being immersed in 70% ethanol,1 mg/L HgCl2+Tween 80 had better disinfection effect than 40 mg/L bleaching power.The treatment time of HgCl2 for steam,leaf,leafstalk and geminated bus should be 12 min;10 min for steam,especially steam collected in the vigorous growth period of mint as explants had higher inducing rate than other explants.MS+0.50 mg/L BA+0.10 mg/L NAA was the suitable medium for buds induction of mint explants.
