摘    要:以哺乳动物的新鲜毛发和在冷冻条件下保存4个月的毛发作为材料。从毛囊中提取DNA,应用PCR方法扩增SRY基因片段,ZFX/ZFY基因片段,探讨哺乳动物性别鉴定的最优化PCR方案。结果表明,毛发DNA和血液DNA均可获得相同的扩增结果。证实了毛发作为PCR扩增材料是可靠的。对不同材料量、不同模板量、不同提取方法进行比较,优化实验方案,达到1根毛发用Chelex-100提取得到的DNA即可以成功地进行

Abstract:In this paper fresh hairs and hairs frozen for 4 months sampled from living animals were taken as materials. The genomic DNA was isolated from hair roots by different methods and then used in the polymerase chain reaction (PCR) to amplify the locus SYR (Sex-determining region of Y chromosome) and locus ZFX (X lined Zinc-finger containing protein)/ZFY (Y lined Zinc-finger containing protein) for optimizing mammalian gender identification protocols. The results indicate that DNA of hair roots has the same result as that of blood. suggesting that rooted hair could be reliable material for gender identification by PCR. By comparing the results of various sample (hair) amount. various isolating methods and various template DNA amount, it was achieved that 1 single hair extracted by Chelex-100 can be enough for identifying the gender. By this means. the sexes of animals of & species were correctly determined. Taking hair as sample is not invasive for the animals. At the mean time. amplifying the loci SRY and ZFX/ZFY by PCR to identify the sex is not only rapid. simple. and accurate. but also common to various species. Therefore. it can be widely applied to identifying mammalian gender.
