西瓜RAPD-PCR体系的正交优化研究

摘    要:采用正交试验设计的方法,建立了西瓜RAPD分析的优化反应体系,即20μL反应体系中含有1×buffer,dNTP250μmol/L、Primer0.4μmol/L、MgCl22.0mmol/L、Taq酶1U和模板DNA20~40ng。适宜的扩增条件为94℃5min;94℃30s,37℃45s,72℃90s,45个循环;72℃5min;4℃保存。

Abstract:The optimal RAPD-PCR system in watermelon was established with orthogonal design. In a total volume of 20 L RAPD-PCR system, it contains 1buffer,250 mol/L dNTP,0.4 mol/L Primer,2.0 mmol/L MgCl_2,1 U Taq DNA polymerase and 2040 ng template DNA. The suitable PCR procedure is one cycle denaturing at 94 for 5 min;45 cycles which involves denaturing at 94 for 30 s,annealing at 37 for 45 s and extending at 72 for 90 s, one cycle extending at 72 for 5 min, and then kept at 4.
