摘    要:构建了包含RSA14-44cDNA的pAS2-1-14诱饵质粒,并进行了人睾丸cDNA文库的酵母双杂交筛选,获得了一条编码人20S蛋白酶体蛋白PSMB5的cDNA片段;用所获得的序列编码该蛋白3’端编码框内全部氨基酸(191个氨基酸);首次证明RSA14-44与PSMB5在酵母细胞中存在相互作用。

Abstract:RSA14-44 is a new gene located on the fourth chromosome of rat and obtained from the rat testis tissue. To obtain the protein interacting with RSA14-44, the recombinant bait plasmid pAS2-1-14 was constructed and transformed into yeast strain Y190 to test pAS2-1-14 for no autonomous activation. One true positive, which obtained by screening the Human testis cDNA libraries, was confirmed to be PSMB5 by sequencing and protein BLAST. It concluded that RSA14-44 could interact with PSMB5.
