摘    要:目的]研究接种甘蔗宿根矮化病(Ratoon stuning disease,RSD)菌后甘蔗防御酶活性的变化,为进一步探明甘蔗与RSD病菌的互作机制提供理论依据.方法]以温汤脱毒后的甘蔗品种桂糖1 1号(GT11)为材料,设RSD菌液浸种为处理,ddH2O浸种为对照,测定甘蔗茎、叶不同时期超氧化物歧化酶(SOD)、过氧化物酶(POD)、过氧化氢酶(CAT)、抗坏血酸过氧化物酶(APX)、谷胱甘肽还原酶(GR)等5种防御酶活性变化.结果]在RSD病菌胁迫下,甘蔗茎和叶片中相关防御酶活性均有所变化,茎中除APX活性一直高于对照外,其他酶活性均表现为在90 d时低于对照,180d时高于对照;叶中除POD活性一直高于对照外,其他酶活性在90 d时高于对照,180 d时低于对照.结论]在RSD病菌胁迫下,甘蔗茎和叶片中CAT、SOD、POD、APX及GR活性均有所提高,随着胁迫时间的延长在茎中出现逐渐高于对照的趋势,叶片则与此相反.

Abstract:Objective]The present study was carried out to assess the changes of defense-related enzyme activi- ties in sugarcane under ratoon stunting disease (RSD), in order to provide references for illustrating the mechanism of interaction between sugarcane and the bacteria of RSD. Method]Sugarcane variety of GTll was selected as the plant materials. Seedcanes were treated at 52 ℃ for 30 min for pathogen free as a control and inoculated with Leifsonia xyli subsp. Xyli(Lxx)as RSD-infected sugarcane. The healthy and RSD-infected sugarcane was confirmed by PCR and the change of defense-related enzyme activity in sugarcane was analyzed in both treatments. Result]The activities of CAT, SOD, POD and GR in stem were lower in RSD-infected treatment than those in control on the 90th day after transplant, while they were higher in treatment on the 180th day after transplant as comparison with the control except the activity of APX which was higher in treatment than this in control during sugarcane growth period. In contrast to stem, the activities of CAT, SOD, APX and GR in treatment was higher on the 90th day and lower on the 180th day as comparison to control, while the activity of POD was higher than that of control from 90 days to 180 days after transplant. Conclusion ]The activities of defense enzymes including CAT, SOD, POD, APX and GR increase more or less in both sugarcane leaf and stem under RSD stress. However,along with longer stress time, they're higher in stem than those of the control. Leaf showed the contrary tide.
