文心兰组织培养初步研究

摘    要:以文心兰花梗为材料,采用不同激素配比、光照强度、培养基分别进行愈伤组织诱导、芽分化诱导和幼苗形成试验。结果表明,在添JJIINAA1.Omg/L+6-BA0.2mg/L+TDZ0.3mg/L+琼脂8g/L+蔗糖30g/L的培养基上,愈伤组织的诱导率最高,可达70.O%;在250lx的散射光条件下,文心兰花梗愈伤诱导率达到最高;在6-BA0.4mg/L+NAAO.2mg/L激素组合条件下,从愈伤组织分化出幼苗的形成率和商品化率可达到最佳效果;中、低无机盐浓度且全量的基本培养基较利于幼苗分化;当分化形成的幼苗株高为1-2cm时利于壮苗生根,生根苗在适宜条件下移栽,成活率可达99.O%以上。因此,采用适合的激素配比、光照强度、基本培养基对花梗进行愈伤组织诱导、分化芽和幼苗,同样可以实现文心兰的大量繁殖。

Abstract:The pedicel of Oncidium was used as material,and different hormone combinations,light intensity and medium were applied to induce callus,bud and plantlet of pedicel.The results showed that the induction rate of callus was the highest on medium supplemented with NAA 1.0mg/L +6-BA 0.2mg/L +TDZ 0.3mg/L + agar 8g/L + sugar 30g/L,it reached 70.0%.When the light intensity was 250lx,the induction rate of callus reached the highest.The formation rate and commercialization rate of plantlets derived from callus were ...
