红茄子叶的不定芽诱导及植株再生

摘    要:【目的】研究不同激素浓度和组合对红茄不定芽及根诱导的影响,建立红茄的高频再生体系,为开展茄子的遗传转化研究奠定基础。【方法】以红茄子叶为外植体,采用0.5~2.0mg/L6-BA、0.1~0.5mg/LIAA激素组合对不定芽进行诱导,采用0~0.5mg/LIAA对根进行诱导,筛选出最佳的分化培养基和生根培养基。【结果】在不定芽诱导培养基中,当6-BA浓度在0.5~2.0mg/L时,随着浓度增加,不定芽分化率逐渐升高;当IAA浓度在0.1,-0.5mg/L时,随着IAA浓度的增加,不定芽萌发率呈先增加后降低趋势,其中以添加6.BA2.0mg/L、IAA0.3mg/L的诱导效果最好,分化率高达86%。在根诱导培养基中,随着IAA浓度的增加,根诱导率呈先降低后升高趋势,诱导率可达100.0%。【结论】红茄子叶不定芽诱导最佳培养基为MS+2.0mg/L6-BA+0-3mg/LIAA;不定芽生根最佳培养基为MS+0.1mg/LIAA。

Abstract:Objective]The present experiment was performed to study the effects of different hormone concentrations and combinations on induction of adventitious bud and root with the aim of establishing a highly efficient regeneration system of S.integrifolium to provide references for the research of eggplant genetic transformation. Method]With the cotyledon of S. in- tegrifolium as explauts, the different hormone combinations (0.5-2.0 mg/L 6-BA, 0.1-0.5 mg/L IAA and 0-0.5 mg/L IAA) were added in MS medium to induce the adventitious bud and root, respectively. Subsequently the optimal differential medium and rooting medium were screened. Result]The germination rate of adventitious buds increased with the increase in 6-BA concentration from 0.5 mg/L to 2.0 mg/L in MS medium. The germination rate of adventitious buds increased initially with IAA concentration increasing from 0.1 mg/L to 0.5 mg/L in MS medium, followed by a decrease. However, 2.0 mg/L 6-BA plus 0.3 mg/L IAA showed ideal induction effects on adventitious buds with 86% germination rate. As the IAA concentra- tion in MS medium increased, the rooting rate decreased at first, then increased. The highest germination rate reached 100%. Conclusion]The best medium for initiation of adventitious bud of cotyledon was MS+2.0 mg/L 6-BA + 0.3 mg/L I- AA, and the optimal rootinz medium was MS+0.1 mJL IAA.
