新疆刺山柑居群的RAPD多态性研究

摘    要:用CTAB法提取了新疆3个不同居群刺山柑(Capparis spinosa L.)总DNA,使用RAPD技术对其进行多态性的研究。从93种随机引物中筛选出3种扩增较稳定的引物,来扩增3个不同居群刺山柑总DNA,共扩增出25条带,每种引物扩增出6~10条带;多态性条带共13条,多态性比例为52%。利用DPS软件所做的统计分析和聚类分析结果表明,3个群居间相似系数在0.69~0.86之间,平均相似系数为0.76;可聚成2类:阿克陶和乌鲁木齐居群刺山柑可聚为A类;吐鲁番居群刺山柑可聚为B类,他们彼此间的遗传变异较小。

Abstract:The total DNAs were extracted by CTAB method from three different Capparis spinosa L. Population in Xinjiang and its polymorphism was studied based on RAPD method. The three primers, which were more stably amplified, were randomly selected from 93 primers. Total 25 RAPD fragments were amplified,13 of which were polymorphic. Each primer could generate 6 to 10 DNA fragments. The percentage of polymorphic loci was 52%. The results analyzed by DPS version software showed that Jaecard similarity coefficients ranged from 0.69 to 0.86 ,whose mean was 0.76 ,indicating that three the genetic variations existed among the three different Capparis spinosa L. Population. A dendrogram was established,based on Hierarchical cluster analysis could divide three different Capparis spinosa L. Population cuhivars into 2 groups,in which there were smaller genetic relations .
