摘    要:为从烟草中克隆到与植物抗逆相关的肌醇半乳糖苷合成酶基因GolS,以辣椒GolS基因为探针,通过电子克隆法从烟草栽培品种K326中分离出一条编码GolS基因的cDNA序列,暂被命名为NtGolS1.NtGolS1包含一个完整的开放读码框,编码343个氨基酸,具备糖基转移酶保守结构域;序列比对结果表明,NtGolS1蛋白与其他植物的GolS蛋白具有很高的相似性;系统进化树分析表明,NtGolS1与耐旱植物维柯萨的Gols亲缘关系最近.这些结果为进一步研究NtGolS1的性质和功能,并为植物抗逆基因工程研究提供理论基础

Abstract:In silicon cloning,a new galactinol synthase(GolS) gene was obtained successfully from tobacco cultivar K326 by using pepper CaGolS gene sequence(EF566470) as probe.The gene was tentatively designated as NtGolS1,whose open reading frame was 1 032 bp in length and encoded 343 amino acids containing a conserved glycosyltransferase domain.NtGolS1 protein was highly homologous to other GolS protein in different species and was more related to that from Xerophyta viscose.Those results were of potential importance not only for further study on the nature and function of tobacco GolS,but also for the genetic engineering improvements of stress-resistance in plants.
