四川省不同地区梁山慈竹RAPD与ISSR遗传多样性研究

摘    要:应用随机扩增多态性DNA(RAPD)和简单重复序列区间(ISSR)对四川省6个不同地区梁山慈竹进行遗传多样性分析。研究表明,筛选出的6个RAPD引物扩增出多态性条带57条,多态性高达95%;筛选出的6个ISSR引物扩增出多态性条带34条,多态性为58.64%。利用Popgen32软件进行聚类分析,结果表明,聚类结果可区分不同地区的梁山慈竹,其遗传距离分别为0.2657-0.9589和0.1092-0.5639,且树状聚类图分类总体趋势一致,表明RAPD和ISSR分子标记结合是一种在DNA水平上有效地检测梁山慈竹遗传结构的优良方法。

Abstract:The genetic diversity and genetic relationships in Dendrocalamus farinosus from 6 different regions in Sichuan Province were studied using random amplified polymorphic DNA(RAPD) and inter-simple sequence repeat(ISSR) amplification markers.The results showed that 6 RAPD and 6 ISSR primers generated 57 and 34 bands,of which the percentage of polymorphic bands(PPB) was 95% for RAPDs and 58.64% for ISSRs.The UPGMA analysis of Popgen 32 program showed the following results: the genetic distance ranged from 0.265 7 to 0.916 3 for RAPDs and 0.109 2 to 0.563 9 for ISSRs.The trends in classification of dendrogram seems to be same,it has proved that RAPD and ISSR molecular markers were a good method to detecte effectively the genetic structure of D.farinosus in DNA level.
