激素和表皮生长因子对牛卵母细胞体外受精的影响

目的]研究激素和表皮生长因子(EGF)对牛卵母细胞体外受精的影响。方法]通过对牛进行体外受精和受精卵体外培养,研究了不同血清、激素及EGF对卵母细胞体外成熟和胚胎体外发育的影响。结果]单独添加ECS组卵母细胞的成熟率、卵裂率、桑囊率均最高,分别为81.39%5、6.05%、30.94%,且卵裂率、桑囊率与添加NBS组的差异显著。在培养液中添加EGF和激素能促进卵母细胞的成熟和卵裂的发生,其成熟率、卵裂率分别为82.50%、83.33%和61.43%、62.21%,均比未添加EGF组的高,差异显著,并且EGF与激素间有协同作用。结论]单独添加10%ECS对卵母细胞的作用最理想,EGF对牛卵母细胞体外成熟及卵裂有良好的促进作用。

Effect of EGF and Hormone on IVF of Oocyte in Bovine

The experiment in the effect of the different serum,hormone and EGF on the IVM,IVD of oocytes in bovine was conducted.The results showed that single addition of hormone obtained higher maturation rate(81.39%),cleavage rate(56.05%),development rate of morula or blastocyst(30.94%);Compared with the addition of NBS,the cleavage rate(51.08%),the rate of morula or blastocyst(25.42%) was significant different(P<0.05).Addition of EGF and hormone in culture medium can promote the maturation rate and cleavage of oocytes,and the maturation rate(82.50%,82.33%)and cleavage rate(61.43%,62.21%)was higher than the group of no addition of EGF(79.49%,56.81%),with significant difference(P<0.05).Furthermore,EGF and hormone had cooperation function.
