羽毛针禾种群遗传多样性分析

摘    要:目的]从分子水平对羽毛针禾居群内、居群间的遗传分化进行研究,揭示其遗传多样性水平。方法]使用11条RAPD引物对生长于古尔班通古特沙漠的优良固沙植物羽毛针禾进行居群遗传变异分析。结果]7个居群76个样品共检测到125个位点,总多态位点百分比为96.8%,居群内平均多态位点百分比为45.3%;羽毛针禾种群Shannon表型多样性指数(I)为0.5151,Nei’s基因多样度指数(h)为0.3471;居群间的基因分化系数为0.5284,即有52.84%的遗传多样性存在于居群间。结论]羽毛针禾居群有较为丰富的遗传变异,且各居群间已有了明显分化。

RAPD Analysis of the Genetic Structure of Stipagrostis pennata Populations

Abstract:Method]The level of genetic diversity inside and among Stipagrostis pennata populations was explored through the research on the study on its genetic differentiation at molecular technique.Method]The genetic variation of good sand-fixating plant-Stipagrostis pennata population in Gurbantunggut Desert was analyzed with 11 RAPD primers.Result]The results fowolled:11 amplifiction random primers used to total of 125 loci in 76 samples from 7 populations were detected and the proportion of polymorphic loci was 96.8%,the proportion of average polymorphic loci inside its population was 45.3%.The diversity index(I) of the Stipagrostis pennata population-Shannon was 0.515 1 and the diversity index(h) Nei gene was 0.347 1.The coefficient of the gene variation among populations was 0.528 4 which revealed that the genetic diversity of 52.84% was existed among populations.Conclusion]There was rich genetic variation in Stipagrostis pennata population and obvious genetic differentiation appeared among populations.
