不同来源的超氧化物歧化酶部分理化性质比较研究

摘    要:对4种不同来源的超氧化物歧化酶的部分理化性质进行了比较研究。结果表明:牛血Cu,Zn-SOD和rhCu,Zn-SOD对一些化学试剂的敏感性方面基本相似,rhCu,Zn-SOD的热稳定性最好,80℃水浴60 min后活力仍有56%;在pH 5.0~11.0的范围内,Cu,Zn-SOD的活力比较稳定,Mn-SOD在pH 5.0以下容易失活,在碱性条件下较稳定。螺旋藻Fe-SOD和rhMn-SOD热、酸碱的稳定性较低;Cu,Zn-SOD、Fe-SOD和rhMn-SOD对一些化学试剂和不同变性剂的敏感性有明显的差别。rhMn-SOD稳定性,对化学试剂的敏感性方面与天然原料来源的Mn-SOD基本一致。Cu,Zn-SOD和Mn-SOD、Fe-SOD相比较,理化性质有明显的差别。

Abstract:In this paper,the physico-chemical characteristics of superoxide dismutase from different sources was studied.The results indicated:bovine Cu,Zn-SOD was similar to rhCu and Zn-SOD in sensitivity to some chemical reagents.The rhCu and Zn-SOD thermostability was best,after 80 ℃ water baths 60 min its vigor still had 56%.In pH 5.0~11.0 scopes,Cu and Zn-SOD vigor was quite stable;rhMn-SOD below pH 5.0 easy deactivation,was stable under the alkalinity condition.The stability of Spiral algae Fe-SOD and rhMn-SOD at heat,acid and alkali was lower.There was obvious difference in sensitivity to some chemical reagents and the different denaturant sensitivity for Cu and Zn-SOD,FeSOD and rhMn-SOD.The rhMn-SOD stability,to the chemical reagent sensitive was consistent to Mn-SOD which originated with the natural raw material.The physico-chemical characteristics of Cu,Zn-SOD and Mn-SOD,Fe-SOD had the obvious difference.
