摘    要:研究了6-BA、NAA和IBA对怀山药不同外植体分化和植株再生的影响。结果表明:以节为外植体,改良的MS+100 g/L香蕉+0.5mg/L 6-BA+1.00 mg/L NAA+0.5%琼脂+0.5%活性炭+3%蔗糖培养基为最优组合,其分化率达100%;最适根块分化培养基是,改良的MS+100 g/L香蕉+0.5 mg/L 6-BA+0.25 mg/L NAA+0.4 mg/L IBA+0.5%琼脂+0.5%活性炭+3%蔗糖;最适无菌茎段分化培养基是,改良的MS+100 g/L香蕉+0.5 mg/L 6-BA+0.50 mg/L NAA+0.8 mg/L IBA+0.5%琼脂+0.5%活性炭+3%蔗糖;最适无菌叶柄分化培养基是,改良MS+100 g/L香蕉+1.5 mg/L 6-BA+0.25 mg/L NAA+0.5%琼脂+0.5%活性炭+3%蔗糖。

Abstract:In this paper the the influence of three primary external hormone(6-BA,NAA,IBA) on direct differentiation and plant regeneration through different explant(burls,aseptic shoots,petioles and root tubers)of the Tiegun Dioscorea Thund was discussed.The result showed:①with burls as explant,modified MS medium + banana 100 g/L +0.5 mg/L 6-BA+1.00 mg/L NAA+0.5% agaragar+0.5%active carbon+3%sucrose was optimal medium culture,the differentiation rate was completely 100% though the experiment of orthogonal experiment L9(34).②Modified MS+banana 100 g/L+0.5 mg/L 6-BA+0.25 mg/L NAA+0.4 mg IBA +0.5% agaragar+0.5%active carbon+3%sucrose was the most suitable culture medium for germfree boot tuber differentiation,modified MS+banana 100 g/L+0.5 mg/L6-BA+0.50 mg/L NAA+0.8 mg/L IBA +0.5% agaragar+0.5%active carbon+3%sucrose was used for germfree shoot differentiation,and the most suitable medium culture for germfree petiole differentiation was modified MS+banana 100 g/L+1.5 mg/L 6-BA+0.25 mg/L NAA+0.5% agaragar+0.5%active carbon+3%sucrose.
