摘    要:目的]探讨西瓜不定芽的诱导与增殖。方法]在西瓜花药愈伤组织诱导的基础上,研究不定芽的诱导与增殖。结果]MS+2.0mg/L 6-BA+1.0 mg/L KT+0.1 mg/L IAA培养基可以使愈伤组织再分化成不定芽。不定芽在培养基MS+0.5 mg/L 6-BA中进行增殖,随着继代培养次数的增加,玻璃化和黄化现象变得更加严重。采取降低6-BA浓度甚至不添加任何激素、提高琼脂的含量、增加光照时间与强度等措施,可以有效减少西瓜不定芽的玻璃化现象。结论]当培养基中细胞分裂素与生长素的比例为10∶1~100∶1时,由西瓜愈伤组织诱导出不定芽的可能性较大。

Abstract:Objective] The aim of the research was to discuss the induction and proliferation of adventitious buds in watermelon.Method] Based on the induction of anther callus in watermelon,the induction and proliferation of adventitious buds were studied.Result] Medium of MS+2.0 mg/L 6-BA+1.0 mg/L KT+ 0.1 mg/L IAA could make callus redifferentiate into adventitious bud.Adventitious buds were proliferated on the medium of MS+0.5 mg/L 6-BA.Vitrification and etiolation phenomena became more serious with the increasing of subculture frequency.Taking such measures as decreasing the concn.of 6-BA or even not adding any hormone,increasing the content of agar and increasing the time and intensity of illumination could effectively reduce the vitrification phenomenon of adventitious buds in watermelon.Conclusion] When the ratio of cytokinin to auxin in the medium was 10∶1-100∶1,the possibility of inducing adventitious buds by watermelon callus was greater.
