摘    要:目的]建立牛皮杜鹃RAPD反应体系。方法]以野生牛皮杜鹃的叶片为材料,采用改进CTAB法提取基因组DNA,并对影响RAPD扩增反应的各因素进行优化。结果]适合牛皮杜鹃的RAPD反应体系为:在20μlPCR反应体积中加模板DNA10ng,Mg2+浓度1.5mmol/L,引物OPC0515pmol/μl,dNTP0.15mmol/L,TaqDNA聚合酶1.0U。结论]该研究为牛皮杜鹃的遗传多样性分析提供了有益的参考。

Optimization for RAPD Reaction System of Rhododendron chrysanthum Pall

Abstract:Objective] To establish a RAPD reaction system of Rhododendron chrysanthum Pall.Method] leaves of Rhododendron chrysanthum Pall were used as experimental materials.Genomic DNA was extracted by the improved CTAB method,and the various factors affecting the RAPD amplification were also optimized.Results] The RAPD program of Rhododendron Chrysanthum Pall in the 20 μl PCR reaction solution were as follows:template DNA 10 ng,Mg2+ 1.5 mmol/L,OPC05 primer 15 pmol/μl,dNTP 0.15 mmol/L,Taq DNA polymerase 1.0 U.Conclusion] This study provides a useful reference for analyzing the genetic diversity of Rhododendron chrysanthum Pall.
