珠芽魔芋不同外植体诱导愈伤组织研究初报

摘    要:目的]研究珠芽魔芋的快速繁殖技术以扩大珠芽魔芋的种植规模。方法]以珠芽魔芋的珠芽、球茎、叶片、叶脉和叶柄为外植体,以MS为基本培养基,添加不同浓度的6-BA和NAA,研究不同的外植体及接种方式和不同的激素对诱导愈伤组织的影响。结果]5种外植体中以带叶叶脉的诱导率最高(92%)。外植体接种方式对愈伤组织的形成有一定影响,水平接种有利于快速获得大量愈伤组织,背接方式获得的愈伤诱导率远远高于竖插和正接方式的。MS+6-BA 0.5 mg/L+NAA 0.1 mg/L的培养基能快速诱导出质量较高的愈伤组织。结论]珠芽魔芋带有叶片的叶脉是诱导愈伤组织的最佳外植体,且以叶背水平与培养基接触效果比较理想,具有分化时间短、速度快的特点。

Abstract:Objective] The research aimed to studying the rapid propagation technology of Amorphphallus bulbifer so as to enlarge the planting scale of A.bulbifer.Method] With the bulblet,corm,leaf blade,nervation and leaf stalk of A.bulbifer as explants,MS was taken as basic medium to add different concn.of 6-BA and NAA.And the effects of different explant and inoculation ways and different hormone on the callus induction were studied.Result] The induction rate of nervation on the leaf was highest(92%) among 5 kinds of explants.The inoculation ways of explants had certain effects on the callus formation.Horizontal inoculation was favorable for quickly obtaining a great deal of calli.The induction rate of callus obtained by inoculation on the back side was greatly higher than that of vertical intercalation and inoculation on the right side.The optimum medium of MS 0.5 mg/L 6-BA 0.1 mg/L NAA could quickly induce calli with higher quality.Conclusion] Nervations on the leaf in A.bulbifer was the optimum explant for callus induction.The effect of connecting back side of leaf connected horizontally with medium was more ideal and it had the characteristics of short differentiation time and fast differentiation speed.
