产絮凝剂黄杆菌的筛选及其絮凝特性研究

摘    要:目的]从活性污泥中分离絮凝活性较高的菌株,进行初步鉴定,研究其产絮凝剂的最佳培养基组成和絮凝条件。方法]采用常规菌种分离纯化方法获得目的菌株,通过单因子试验研究其培养条件、絮凝条件和絮凝活性。结果]分离到的菌株B2经初步鉴定为黄杆菌属。其产絮凝刺的最佳培养基组成为蔗糖20g/L,(NH4)2SO4 1.2g/L,KH2PO4 4.0g/L。培养基的初始pH值为8.0,装液量为100ml(250ml三角瓶),接种量为2m1。适宜的培养条件:温度为35℃,摇床转速为120r/min,培养时间为48h。用高岭土悬浊液对其絮凝条件进行研究,确定培养液最佳絮凝pH值为7.0,发酵液投加量为2ml,添加Ca^2+、Zn^2+能显著提高其絮凝活性。B2菌株在上述最佳条件下的发酵液对0.4%的高岭土悬浊液和生活污水的絮凝率均在85%以上。结论]菌株B2絮凝活性较高,具有良好的应用前景。
