产絮凝剂黄杆菌的筛选及其絮凝特性研究

目的]从活性污泥中分离絮凝活性较高的菌株，进行初步鉴定，研究其产絮凝剂的最佳培养基组成和絮凝条件。方法]采用常规菌种分离纯化方法获得目的菌株，通过单因子试验研究其培养条件、絮凝条件和絮凝活性。结果]分离到的菌株B2经初步鉴定为黄杆菌属。其产絮凝刺的最佳培养基组成为蔗糖20g／L，（NH4）2SO4 1．2g／L，KH2PO4 4．0g／L。培养基的初始pH值为8．0，装液量为100ml（250ml三角瓶），接种量为2m1。适宜的培养条件：温度为35℃，摇床转速为120r／min，培养时间为48h。用高岭土悬浊液对其絮凝条件进行研究，确定培养液最佳絮凝pH值为7．0，发酵液投加量为2ml，添加Ca^2＋、Zn^2＋能显著提高其絮凝活性。B2菌株在上述最佳条件下的发酵液对0．4％的高岭土悬浊液和生活污水的絮凝率均在85％以上。结论]菌株B2絮凝活性较高，具有良好的应用前景。
