摘    要:目的]比较不同浓度乙醇醇析的猪苓多糖对羟自由基(.OH)的清除能力,以进一步提高猪苓的药用价值、经济价值和实用价值。方法]用热水浸提法提取猪苓菌核水溶性多糖,经不同浓度乙醇醇析、纯化获不同等级的猪苓多糖;采用苯酚-硫酸法测定猪苓多糖含量,以多糖含量为评价指标,分析多糖醇析程度;采用邻二氮菲-Fe2+-H2O2体系检测浓度40%、80%和90%乙醇醇析获取的猪苓多糖对.OH的清除作用。结果]猪苓多糖在不同浓度乙醇中醇析效果不同,乙醇浓度为80%时,醇析效果相对较好。不同浓度乙醇醇析获取4种等级猪苓多糖,其中3种对.OH具有清除作用。浓度40%、80%乙醇醇析获取的猪苓多糖在一定浓度范围内与清除.OH的能力呈良好的正相关。结论]研究结果为进一步研究猪苓多糖的结构和功能提供了参考。

Abstract:Objective] The study aimed to compare the scavenging effects of ·OH by polysaccharides of polyporus using different concentrations of alcohol precipitation,to improve the medicinal,economic and practical values of polyporus.Method] Using hot water extraction method,water-soluble polysaccharides from sclerotia of polyporus were isolated,then adopted different concentrations of alcohol precipitation and purification it,different grades of polysaccharides of polyporus were obtained;content of polysaccharides was determined by phenol-sulfuric acid method,and the content of polysaccharides was viewed as the evaluation index to analyze the degree of polysaccharide.Hydroxy free radical scavenging effects by 40%,80% and 90% polysaccharides 0f polyporus were detected using system of phenanthroline-fe2+-H2O2.Result] The result showed that the degree of polysaccharides of alcohol-precipitation was different in different concentrations of alcohol.When the concentration of alcohol was 80%,the effect of alcohol-precipitation was better.Four grades of polysaccharides of polyporus were obtained using different concentrations of alcohol,three of which had scavenging effects on ·OH.Polysaccharides of polyporus based on 40% and 80% alcohol-precipitation had better positive correlation with scavenging effects of ·OH in a certain range.Conclusion] The study provided reference for further studying the structure and function of polysaccharides of polyporus.
