菊花对潮霉素敏感性的研究

摘    要:目的]探讨菊花对潮霉素的敏感性,为菊花的遗传转化提供理论依据。方法]以菊花品种45、34、30、44、20、35的愈伤组织为材料,采用MS培养基附加6-BA、IAA、KT,以不同浓度的潮霉素进行处理。结果]随着潮霉素浓度的增加,全部试材的褐化率均增大,体积增殖均减小。菊花35对潮霉素最敏感,在潮霉素浓度为10 mg/L时,其褐化率为56.67%。在潮霉素浓度小于20 mg/L时,菊花愈伤组织的体积增殖平均为3 mm;在潮霉素浓度大于20 mg/L时,其体积增殖平均为0.5 mm,且其褐化率达70%~80%。作为菊花筛选标记时,潮霉素的适宜浓度为20 mg/L。结论]菊花对潮霉素的敏感性在基因型间存在差异。来自同一基因型菊花不同部位的外植体对抗生素的反应不同。

Abstract:Objective] The purpose was to discuss the sensitivity of chrysanthemum to hygromycin so as to supply theoretical basis for its genetic transformation.Method] With calli of chrysanthemum varieties 45,34,30,44,20 and 35 as tested materials,they were treated by hygromycin with different concn.on MS medium added 6-BA,IAA and KT.Result] Along with the increment of hygromycin concn.,the browning rate of all the tested materials increased and their volume proliferation decreased.Chrysanthemum 35 was most sensitive to hygromycin,when hygromycin was 10 mg/L,its browning rate was 56.67%.The average volume proliferation of chrysanthemum calli was 3 mm when hygromycin was lower than 20 mg/L,and that was 0.5 mm and their browning rate reached 70%~80% when hygromycin was more than 20 mg/L.As the screening marker of chrysanthemum,the suitable concn.of hygromycin was 20 mg/L.Conclusion] The sensitivity of chrysanthemum to hygromycin had difference among genotypes.The explants from different parts of the chrysanthemum with the same genotype had different responses to antibiotics.
