摘    要:目的]建立一种用聚苯乙烯反相色谱柱联用示差检测器高效液相色谱法(HPLC)分析依替米星发酵液的方法。方法]使用MKF—RP—ETI色谱柱(100.0mm×7.8mm,200.0mm×7.8mm,5μm),流动相:O.22mol/L醋酸盐缓冲液(pH=6.0),流速:1ml/min,检测器:示差检测器。结果]MKF—RP—ETI色谱柱和HPLC—RD法可方便、快速用于分析鉴定依替米星发酵液,依替米星保留时间约为8min。结论]该方法可方便、快速、高效地用于依替米星发酵液生产、纯化过程中在线检测的分离分析。

Separation of Etimicin Fermentation Liquor by High Performance Liquid Chromatographic Column Packed with Polymer Resin

Abstract:Objective] The aim was to establish a method for the isolation of Etimicin fermentation liquor by polystyrene reverse phase column(MKF-RP-ETI)with High Performance Liquid Chromatographic(HPLC).Method] The MKF-RP-ETI column was used to separate Etimicin fermentation liquor.The mobile phase was 0.22 mol/L acetate buffer(pH=6.0).The flow velocity was 1 ml/min.The detector was refractive index detector(RD).Result] The MKF-RP-ETI column with HPLC-RD showed satisfactory features in separation properties and the retention time of Etimicin was as long as 8 minutes.Conclusion] This method can be applied for rapid and efficient purification and isolation of Etimicin fermentation liquor.
