不同培养液对蛹虫草多糖及虫草酸含量的影响

摘    要:目的]明确不同碳氮源培养液对蛹虫草多糖及虫草酸含量的影响。方法]以不同碳氮源培养液振荡培养蛹虫草,用苯酚-硫酸法测定虫草多糖含量,用高碘酸钠法测定虫草酸含量。结果]培养液中多糖含量以米蛹配方最高,为6.48mg/ml,其次为玉米糁蛹的5.87mg/ml;虫草酸含量以玉米糁蛹配方最高,为5.21mg/ml,其次为米蛹的4.94mg/ml;菌丝体中多糖含量以玉米糁蛹配方最高,为68.32mg/g,其次为米蛹的57.91mg/g;虫草酸以麦米蛹配方最高,达186.29mg/g,其次为小麦蛹的180.32mg/g。结论]选择适宜碳氮源进行蛹虫草的液体培养,可获得多糖和虫草酸含量很高的培养物。

Abstract:Objective] The aim of the paper was to ascertain the effect of different liquid culture media with different carbon source and nitrogen source on the contents of polysaccharide and cordyceps acid in Cordyceps militaris. Method] Cordyceps militaris was cultivated vibratingly in different liquid culture media with different carbon and nitrogen sources. The content of polysaccharide was determined by the phenol-sulfuric acid method and that of cordyceps acid was determined by the sodium periodate method. Result] The content of polysaccharide in liquid culture media that compound with rice and pupa was the highest, reached 6.48 mg/ml, the next was that in the liquid culture media that compound with corn grits and pupa, reached 5.87 mg/ml. The content of cordyceps acid in liquid culture media that compound with corn grits and pupa was the highest, reached 5.21 mg/ml, the next was that in liquid culture media that compound with rice and pupa, reached 4.94 mg/ml. The content of polysaccharide in mycelium that cultivated in liquid culture media that compound with corn grits and pupa was the highest, reached 68.32 mg/g, the next was that cultivated in liquid culture media that compound with rice and pupa, reached 57.91 mg/g. The content of cordyceps acid in mycelium that cultivated in liquid culture media that compound with wheat, rice and pupa was the highest, reached 186.29 mg/g, the next was that cultivated in liquid culture media that compound with wheat and pupa, reached 180.32 mg/g. Conclusion] Cordyceps militaris cultivated vibratingly in liquid culture media that compound with suitable carbon and nitrogen sources could get the culture that the contents of polysaccharide and cordyceps acid were higher.
