草珊瑚试管苗生根培养研究

摘    要:目的]建立有效的草珊瑚Sarcandra glabra(Thunb.)Nakai]试管苗生根系统,以完善组培体系。方法]以草珊瑚组培再生芽为供试材料,研究了不同生长素种类及浓度、不同无机盐和蔗糖浓度对其不定根形成的影响。结果]NAA不利于草珊瑚的生根诱导,IBA可促进生根,提高生根速度和根系质量,浓度以0.20mg/L最佳;较低浓度无机盐更有利于草珊瑚生根培养,如1/2MS或1/4MS;蔗糖浓度在30g/L时诱导草珊瑚生根效果最好,过高或过低都会抑制其生根。结论]草珊瑚试管苗生根培养的适宜培养基为1/2MS+IBA0.20mg/L+蔗糖30g/L或1/4MS+IBA0.20mg/L+蔗糖30g/L。

Study on Rooting Cultre of Sarcardra glabra (Thunb.)Nakai in vitro

Abstract:Objective]The research aimed to establish an efficient rooting protocol in vitro for perfecting the tissue culture system of Sarcandra glabra (Thunb.) Nakai.Method]With the regenerated buds of S.glabra (Thunb.) Nakai as tested materials,the effects of auxin,mineral strength and sucrose concentration on adventitious root formation were studied.Result]NAA went against the rooting induction of S.glabra (Thunb.) Nakai.IBA could promote in vitro rooting for improving the rooting speed and the root system quality.The optimum concentration was 0.20 mg/L.Lower mineral strength such as 1/2MS or 1/4MS was much better for rooting of S.glabra (Thunb.) Nakai.The best induction results were obtained at the concentration of 30 g/L.Too high or too low sucrose concentration had some restrainable effect on root formation.Conclusion]The optimal rooting media of S.glabra (Thunb.) Nakai was 1/2MS+0.20 mg/L IBA+30 g/L sucrose and 1/4 MS+0.20 mg/L IBA + 30 g/L sucrose.
