番茄品种渝粉109的DNA指纹图谱构建

摘    要:用SDS法提取番茄杂一代品种"渝粉109"及其亲本的基因组DNA,通过RAPD-PCR分子标记方法,构建其DNA指纹图谱,用于种子纯度鉴定。利用200个RAPD随机引物,以杂一代品种及其亲本的基因组DNA为模板组进行筛选,结果显示:8个RAPD引物的DNA条带表现出多态性,其中R3能稳定地在亲本之间扩增出互补的特异条带。经定向验证、种子真伪鉴定、纯度鉴定,表明构建的渝粉109指纹图谱有效。

Abstract:Genomic DNAs were extracted from tomato hybrid Yufen 109 and its parents by SDS method.DNA fingerprinting of Yufen 109 was established using random amplified polymorphic DNA(RAPD) assay for purity identification of seeds.In this study,200 random primers were screened to identify Yufen 109 and its parents.Results showed R3 could be used to establish DNA fingerprinting,and the fingerprinting could be use to identify purity of Yufen 109.
