摘    要:陆地棉Ligon lintless是单基因显性突变体,其性状表现为成熟的长纤维极度缩短,一般在4~6 mm,植株的叶片和茎呈扭曲状。本研究用陆地棉遗传标准系TM-1与棉花纤维突变体Ligon lintless的杂交一代(由于材料的特殊性即相当于回交一代BC1,后文称为BC1)的246个单株为作图群体,利用4000对SSR引物筛选出在双亲间表现多态性的引物28对,多态频率为0.7%。江利用这28对引物对BC1群体进行分析,共检测到23个多态性位点。用Joinmap3.0软件绘制连锁图,该图谱共覆盖168 cM,相当于棉花总基因组的3.74%,标记间的平均遗传距离为7.3 cM,将棉花纤维突变体Ligon lintless的L i基因连锁定位在22号染色体上。

Abstract:The Ligon lintless,with the expression of mature long fibers seriously shorten to 4-6 mm,which resulted in twisted leaf and stem of the plant,was a monogenic,dominant mutant in cotton.The objectives of this research were making use of the 246 plants segregating for Li-1 from the backcross of the upland cotton genetic standard line TM-1 and the Ligon lintless mutant in cotton,and making linkage map for the Ligon lintless mutant in cotton with the SSR method.28 pairs of SSR primer with the polymorphisms between parents TM-1 and Ligon lintless were screened from 4000 primers to analyze BC1 population.23 markers were detected by the primers mentioned above.And these markers were used to construct the molecular genetic linkage map by Joinmap 3.0.The map spanned about 168 cM and covered 3.74 % of cotton genome.The average distance of markers was 7.3 cM,and the location of Li was on chr.22.
