摘    要:以子叶为外植体,研究了苗龄、培养基中Agar浓度、BA与NAA的组配和添加AgNO3、GA3对子叶产生不定芽的影响.结果表明,5~9d苗龄的外植体最易诱导出不定芽;培养基中BA浓度是影响不定芽形成的关键,BA浓度低于2mg/L,不定芽难以发生,BA6mg/L与NAA0.05mg/L的搭配有利于不定芽形成;在一定范围内不定芽发生频率随着Agar浓度的提高而提高;培养基中添加5mg/L AgNO3,2~3mg/LGA3可以提高不定芽的发生频率;在此基础上建立了青菜离体再生体系,利用该体系能够获得较高的芽苗再生频率,但存在着青菜基因型间的差异.

Abstract:The effects of factors including seedling age, different combinations of BA and NAA concentrations, Agar concentration,AgNO 3 and GA 3 on bud induction from cotyledon are analysed. The results indicate that the explants from 5 9 days old seedling are the most efficient to induce adventious buds. The concentration of BA is the key factor to adventious buds differentiation. BA lower than 2 mg/L, is harder to induce adventious buds. The combinations of 6 mg/L BA and 0.05 mg/L NAA can improve adventious buds induction. The culture medium with 12 g/L Agar, 5 mg/L AgNO 3 and 2 3 mg/L GA 3 can increase the frequence of differentiation. According to these results, a procedure for induction of adventious buds and regeneration system for Chinese cabbage is estabished. By using this system , high regeneration frequence can be obtained, but could be affected by genotypes.
